Increases in emergency room visits due to abuse of designer drugs, popularly known by the street names “K2” and “Spice,” are a cause for social, judicial, and clinical concerns. The psychoactive components in these herbal drugs mainly consist of different synthetic cannabinoids, and users of these street drugs are primarily within the age group of 12 to 20 years old. The abusive use of synthetic cannabinoids results in anxiety, nausea, vomiting, tachycardia, elevated blood pressure, tremors, seizures, hallucinations, and paranoid behavior, but the effects of maternal use of synthetic cannabinoids during pregnancy are ambiguous due to limited studies in humans and a relative short history of the drugs. In this review, we discuss the known and potential adverse effects of synthetic cannabinoids on human pregnancy using knowledge gathered from studies in mice and limited studies in humans. In mice, multiple sites and stages of pregnancy are potential targets of synthetic cannabinoids, including preimplantation embryo development, oviductal embryo transport, implantation, placentation, and parturition. It is anticipated that maternal use of synthetic cannabinoids would result in severely compromised female fertility and pregnancy outcome.  相似文献

Cannabinoids (CBs) are psychoactive compounds, with reported anticancer, anti-inflammatory, and anti-neoplastic properties. The study was aimed at assessing the hepatoprotective effects of CB against ethanol (EtOH)-induced liver toxicity in rats. The animals were divided into seven groups: control (Group I) and Group II were treated with 50% ethanol (EtOH 5 mg/kg). Groups III, IV, and VI were treated with (EtOH + CB 10 mg/kg), (EtOH + CB 20 mg/kg), and (EtOH + CB 30 mg/kg), respectively. Groups V and VII consisted of animals treated with 20 and 30 mg/kg, of CB, respectively. Biochemical analysis revealed that Group IV (EtOH + CB 20 mg/kg) had reduced levels of ALT—alanine transferase, AST—aspartate aminotransferase, ALP—alanine peroxidase, MDA—malondialdehyde and increased levels of GSH-reduced glutathione. Histopathological analysis of liver and kidney tissues showed that EtOH + CB (20 and 30 mg/kg) treated animal groups exhibited normal tissue architecture similar to that of the control group. ELISA revealed that the inflammatory markers were reduced in the animal groups that were treated with EtOH + CB 20 mg/kg, in comparison to the animals treated only with EtOH. The mRNA expression levels of COX-2, CD-14, and MIP-2 showed a remarkable decrease in EtOH + CB treated animal groups to control groups. Western blot analysis revealed that CB downregulated p38/JNK/ERK thereby exhibiting its hepatoprotective property by inhibiting mitogen-activated protein kinase pathways. Thus, our findings suggest that CB is a potential candidate for the treatment of alcohol-induced hepatotoxicity.  相似文献

Using electrophysiological techniques (a patch-clamp technique in the whole-cell configuration and intracellular perfusion of neurons), we studied the effect of cannabinoids on the characteristics of glycine-activated currents in freshly isolated pyramidal neurons of the rat hippocampus. We found that endocannabinoids (anandamide and 2-arachidonoyl glycerol), as well as a synthetic cannabinoid, WIN 55,212-2, when applied in physiological concentrations, decreased the amplitude of glycine-activated currents. The agents under study accelerated the kinetics of activation and desensitization of glycine-induced Cl currents. The characteristics of the currents recovered after washout from cannabinoids. Changes in the kinetics of desensitization of glycine-activated currents depended noticeably on the holding potential; at positive potentials the sensitivity to cannabinoids was higher. These effects of cannabinoids were also observed in the presence of antagonists of CB1/CB3 receptors and an inhibitor of G proteins, GDPβS. These data indicate that under our experimental conditions cannabinoids exerted direct effects on glycine receptors. Experimental data suggest that the endogenous cannabinoid system is involved in gastric function in different animal species. In most of them, CB(1) receptors have been localized on vagal terminals innervating the external wall of the stomach. We aimed at studying the putative presence and distribution of these receptors in the human gastric mucosa. To this end, we first performed Western blotting, RT-PCR, in situ hybridization, and immunohistochemical analysis of CB(1) protein distribution in biopsy samples of healthy individuals. To determine the precise cell populations expressing CB(1) receptors, we performed double immunofluorescence plus confocal microscopy analysis of the same samples. Our results show that CB(1) receptors are present in the gastric epithelium of the mucosa. Specifically, they are expressed by a subpopulation of mucosal cells, the acid-secreting parietal cells, as shown by double immunohistochemical staining and by their differential abundance in subregions of the gastric mucosa. These results reinforce the notion of a prominent role for the endocannabinoid system in the gastric function in humans and postulate the use of cannabinoid CB(1) receptors in parietal cells as new therapeutic targets for the regulation of gastric acid production.  相似文献

Four populations of Cannabis sativa L. grown from seeds collected in Panama, Jamaica, Nepal, and east central Illinois were grown under controlled conditions in growth chambers. One set was grown under warm conditions (32° day and 23° night) and the other set was grown under lower temperatures (23° day and 16° night). CO2 exchange and transpiration were examined under various temperatures and light intensities. Observations on growth, and analyses for chlorophyll and Δ1THC (tetrahydrocannabinol) content were made. Under warm growth conditions, the central Illinois population had the highest photosynthetic rate at all temperatures investigated. The Nepal population had intermediate rates, while the Jamaica and the Panama populations had the lowest rate. The Jamaica and Panama populations had insignificant changes in photosynthetic response to changes in temperatures between 15° and 30°. Under cool growing conditions the central Illinois population had the highest rate of photosynthesis with a definite peak at 25°. Nepal plants had intermediate rates of photosynthesis, while the Panama and Jamaica populations had the lowest rate. Differences in chlorophyll and drug content were also significant between these populations. From these data it is suggested that the four populations can be grouped into different ecotypes genetically adapted to their respective environments.  相似文献
