新疆甜瓜子叶原生质体的培养和植株再生   总被引:14,自引:0,他引:14  
从新疆甜瓜(Cucumts melo L.)的无菌苗子叶游离原生质体,用改良的 Miller 培养基(Ma)培养,得到了再生细胞的高频率分裂。比较了液体浅层培养、双层培养与琼脂糖珠看护培养等方法,发现由烟草瘤细胞 B_6S_3看护的琼脂培珠培养,最宜于新疆甜瓜子叶的原生质体。再生的愈伤组织经液体与固体两步培养程序分化出完整的小植株。  相似文献

Two sites for -oxidation of fatty acids in pea (Pisum sativum L.) cotyledons exist. One site is the microbody, the other the mitochondrion. Mitochondrial -oxidation of fatty acids is carnitine-dependent. The fatty acid permeates the membrane as palmitoylcarnitine which is formed from cytosolic-side palmitoyl-CoA by a carnitine palmitoyltransferase located on the exterior face of the inner mitochondrial membrane as a peripheral protein. A single-gated pore integral membrane translocator is proposed to exchange the palmitoylcarnitine for carnitine or acetylcarnitine across the membrane. An internal (matrix side) carnitine palmitoyltransferase then reforms palmitoyl-CoA which enters -oxidation and subsequently the tricarboxylic-acid cycle.  相似文献

Embryonic axes and cotyledons of three soybean ( Glycine max L. cv. Gudzon) seed lots designated as "normal", "naturally aged" and "acceleratedly aged" were analyzed for their organic free radical contents. No signals of free radicals were found in ESR spectra from cotyledonary material of the three samples investigated. High levels of organic free radicals were observed in the embryonic axes. There were significant differences in the free radical contents of the embryonic axes: the relative values of free radicals were 100, 190 and 170% for samples aged normally, naturally and in an accelerated manner, respectively. These results point to the physiological role of the embryonic axes during natural or accelerated aging of seeds, notwithstanding that the axis is a small part as compared to the cotyledons. It is suggested that lipid peroxidation in embryonic axes may play an important role in the seed deterioration during aging.  相似文献

研究了外源多胺对离体黄瓜(CucumissativusL.)子叶内在多胺含量和花芽构建的影响。在MS培养基中添加5×10-2mmol·L-1亚精胺(Spd)、精胺(Spm)能明显提高子叶内Spd含量和SpdPut值,子叶成花率也明显高于对照组,而添加5×10-2mmol·L-1Put、5mmol·L-1环己胺硫酸盐(CHAS)培养的结果则相反。表明子叶内Spd含量和SpdPut与成花率呈正相关关系。在添加5mmol·L-1D精氨酸(DArg)处理后,子叶内SpdPut高于Spd处理,但子叶成花率低于对照组,表明Put、Spd含量也影响离体子叶花芽的构建。在实验中添加CHAS处理后子叶内Spm含量高出对照组10倍,子叶成花率却为0,其它各处理与对照组的内源Spm变化基本一致,成花率却差别明显,表明子叶内Spm含量与黄瓜子叶花芽构建之间无明显相关性。  相似文献

The inhibitors of cytokinin N-glucosylation are known to influence the growth of some plant objects including cotyledons. The use of the plate meristem of zucchini cotyledon as an experimental system allowed us to study for the first time the way in which the changes in the cell division are integrated in this growth reaction. Roscovitine, a potent inhibitor of cytokinin N-glucosylation and cycline-dependent kinases, did not show to have an effect on the meristem activity when applied in 100 μM to cultivated zucchini cotyledons, and acted as an inhibitor in concentrations higher than 400 μM. A 200 μM roscovitine stimulated both palisade cell division and growth. In different seed batches, 400 μM roscovitine acted as a stimulator or an inhibitor. A much stronger stimulating effect on growth and cell division was observed after application of benzyladenine (BA, 10 μM). In contrast to BA, roscovitine provoked a formation of principally flat lamina. In combined treatments, it lowered the stimulating effect of BA; 400 μM roscovitine combined with BA severely suppressed the growth and division activity. This cellular behavior and changes in cotyledon growth could be due to the roscovitine-provoked changes in endogenous cytokinin levels via the inhibition of cytokinin N-glucosylation. Roscovitine-caused stimulation of cell growth and division is stronger in the marginal meristem than that registered in central regions of the cotyledon blade. In this region it also changed the pattern of cell division and lowered the adhesion between the clusters, which enhanced the appearance of local ruptures of the cotyledon edges. The first palisade layer of the plate meristem of cultured zucchini cotyledons, the natural mono-layer of proliferating palisade cells, may be used for screening the inhibitors of cycline-dependent kinases and cytokinin N-glucosylation with regard to their effects on cell division and growth.  相似文献

本文对无菌培养条件下黄瓜子叶外植体龄对子叶生根能力的影响进行了研究,结果表明,对于分别萌发5天、10天、15天、20天的子叶在MS基本培养基上,无论有生长系NAA,均以5天龄的子叶生根能力最强。附加1.0mg/LNAA,显著促进子叶生根。对子叶起始过氧化物酶活性测定表明,子叶起始过氧化物酶活性与生根能力之间存在有一定的关系。  相似文献

A method for regenerating pigeonpea [Cajanus cajan (L.) Millsp.] plants has been developed using distal cotyledonary segments of mature seeds as explants. A large number of shoot buds were induced directly from explants of genotypes T-15-15 and GAUT-82-90 when cultured on six different basal media fortified with 22.2 μm N6-benzylaminopurine, 2.3 μm kinetin, and 271 μm adenine sulfate. The shoot buds developed into shoots when they were subcultured on the same medium but with one-tenth concentrations of cytokinins and adenine sulfate. The shoots elongated by subculturing first two to three times on Murashige and Skoog (MS) basal medium supplemented with 2.22 μm N6-benzylaminopurine and 0.54 μm α-naphthaleneacetic acid or on half-strength MS medium containing 2.89 μm gibberellic acid, and then once on the same medium without growth regulators. Elongated shoots were rooted with 80–85% efficiency on MS medium with 4.92 μm indole-3-butyric acid and the plantlets were transferred for hardening. Plants survival in pots was 70–75%.
