Cytosine methylation appears to be established as an important DNA base modification involved in regulation of gene expression but is poorly understood from an evolutionary viewpoint. Xiphophorus progeny from inter-species crosses and backcrosses that are utilized in contemporary tumor induction studies were analyzed for cytosine methylation pattern inheritance using Southern blot analyses. Methylation patterns at CCGG sequences of 411 independent chromosomes in three distinct inter-species crosses were analyzed. In every case the non-recurrent parental methylation pattern remained unaltered for each of the genes studied, once introduced into the recurrent parental genetic background. Through F1 inter-species hybridization and succeeding meiosises leading to first generation (BC1) and second generation (BC2) backcross hybrid progeny, we demonstrate that parental species methylation patterns are stable.  相似文献

Summary The aim of the experiment was to study the possibility of facilitating the gene transfer and reducing the number of required backcrosses through pollen irradiation and subsequent selection of F1M1 plants containing a very high proportion of sterile pollen as male parent for backcrossing. Anthers of a donor line, C-3-1, were irradiated with 1,500 rad -rays and the pollen used for pollination of a recipient genotype W-8 which posseses a number of recessive marker genes. Five F1M2 plants containing more than 80% sterile pollen grains and one semi-sterile plant were selected and used for backcross to W-8. The segregation pattern of four characters expressed in the first backcross generation [W–8×(W–8×C–3–1)] was assessed and compared with the non-irradiated control. A changed segregation pattern was observed (in some cases even non-transfer of a paternal allele) as well as a shift towards more plants possessing the investigated maternal alleles. A scheme for backcross procedure in combination with pollen irradiation is discussed.  相似文献

Summary Each of two sorghum (Sorghum bicolor (L.) Moench) cultivars were crossed with representatives of three wild sorghum races. Backcross-derived sorghum populations containing 3.125 to 50% wild germplasm were evaluated for grain yield, 100-kernel weight, days to flower, and plant height. Population means increased linearly with backcrossing for kernel weight, increased curvilinearly for grain yield, decreased curvilinearly for plant height, and changed erratically for days to flower. For all traits, the relationship between genetic variance and level of backcrossing deviated significantly from that expected based on an additive model. Genetic variance usually reached a maximum in the BC1 or BC2. The BC1 genetic variance for grain yield, averaged over matings, was twice as large as the average BC0 genetic variance.  Populations derived by multiple backcrosses are potentially useful for quantitative trait locus (QTL) mapping studies. Comparisons of relative power to detect QTL using populations derived by multiple back-crosses are needed to make decisions when mapping projects are initiated. The objective of this study was to theoretically compare the power to detect QTL in populations derived by multiple backcrosses relative to mapping in a recombinant inbred population of equal size. Backcrossing results in a reduction in genetic variance with each generation and also results in an increasing frequency of the recurrent parent marker genotype. The relevant outcome for QTL mapping is a reduction in genetic variance to partition between marker genotype classes and increasing unbalance of the number of individuals contributing to the mean of the marker genotypes. Both of these factors lead to a decrease in the power to detect a QTL as the number of backcross generations increases. Experimental error was held constant with the populations compared. From a theoretical standpoint, backcross-derived populations offer few advantages for QTL detection. If, however, a backcrossing approach is the most efficient method to achieve a desired breeding objective and if QTL detection is an objective of equal or less importance, backcross-derived populations are a reasonable approach to QTL detection. 本研究采用一种新型甘蓝型油菜细胞质雄性不育系(EruCMS)(Brassicanapus2n=4x=38)与红菜薹(B.campestris L.ssp.chinensis L.var.utilis Tsen et Lee.2n=2x=20)进行远缘杂交,并以红菜薹为轮回亲本连续回交,对甘蓝型油菜细胞质雄性不育系与红菜薹的杂种一代及其回交后代进行植物学性状及品质性状调查测定。结果表明,供体亲本和受体亲本在回交转育过程中发生明显的核置换,其杂交及回交后代的农艺性状及品质性状出现波动,并且随着回交次数的增多,世代越高的材料越相似于轮回亲本红菜薹,而世代越低材料遗传变异范围更大。  相似文献

Advanced backcross QTL (AB-QTL) analysis was deployed to identify allelic variation in wild barley (Hordeum vulgare ssp. spontaneum) of value in the improvement of grain yield and other agronomically important traits in barley (Hordeum vulgare ssp. vulgare) grown under conditions of water deficit in Mediterranean countries. A population of 123 double haploid (DH) lines obtained from BC1F2 plants derived from a cross between Barke (European two-row cultivar) and HOR11508 (wild barley accession) were tested in replicated field trials, under varying conditions of water availability in Italy, Morocco and Tunisia, for seven quantitative traits. Significant QTL effects at one (P 0.001) or more trial sites (P 0.01) were identified for all traits. At 42 (52%) of the 80 putative QTLs identified, the allele increasing a “traits' value” was contributed by H. spontaneum. For example, though the majority (67%) of QTL alleles increasing grain yield were contributed by H. vulgare, H. spontaneum contributed the alleles increasing grain yield at six regions on chromosomes 2H, 3H, 5H and 7H. Among them, two QTLs (associated to Bmac0093 on chromosome 2H and to Bmac0684 on chromosome 5H) were identified in all three locations and had the highest additive effects. The present study shows the validity of deploying AB-QTL analysis for identifying favourable QTL alleles from wild germplasm and indicates its potential as an enhancement strategy for the genetic improvement of cultivars better adapted to drought-prone environments.  相似文献

Summary The extent of correlation was estimated between isozyme genotypes and the four widely segregating characters — leaf segment W/L ratio, stigma exsertion, fruit weight, and seed weight — in the first backcross of F1 Lycopersicon esculentum x Solanum pennellii to the former parent. The inbred parents differ in their alleles at the 12 tested isozymic loci, which are known to mark a minimum of eight of the twelve tomato chromosomes. Based on the isozyme data, a mean heterozygosity value, ¯H, was calculated which estimates the proportion of pennillii alleles in each individual. Correlations between mean heterozygosity and observed levels of each quantitative trait were highly significant and positive or negative as expected from the relative parental values. Plants with the lowest mean heterozygosity — i.e., closest to the esculentum zymotype also had mean values closest to those of this parent amongst the whole backcross population for each of the quantitative traits.Bivariate and multiple regression analysis was used to evaluate the ability of isozymes vs diagnostic morphological characters to estimate the portion of recurrent parent genes carried in each backcross individual. The results suggest that isozyme data gives better estimates than single diagnostic morphological characters and approach the level obtained by combinations of three morphological traits. Since electrophoretic determinations are made on small seedlings, selection at that stage can effect great savings of space and effort by greatly deminishing the size of the population needed at maturity. As such, isozyme selection would precede morphological selection but not replace it, thus the predictive value of these biochemical markers as well as diagnostic morphological characters could be obtained.  相似文献

Twelve somatic hybrid lines were raised through polyethylene glycol-mediated intergeneric protoplast fusion between Volvariella volvacea and Pleurotus florida using a double selection method. Four hybrid lines belonging to two distinct colony morphology types could be maintained in culture. Basidiocarps could be generated from two hybrid lines, one of which showed resemblance to the P. florida parent while the other showed intermediate parental morphology. Hybridity of the fusant lines was established on the basis of colony morphology, mycelial growth rate, hyphal traits, fruit body morphology, isozyme and RAPD analysis. Four isozyme activities were analyzed to provide biochemical criteria for detection of polymorphism among the hybrids and their parents. Using 20 random decamers a total of 203 RAPD bands ranging from 0.09 to 1.6 kb were observed. The UPGMA method of clustering exhibited two major phylogenetic clusters, the first of which comprised of the parents, two fruit body generating hybrid lines and their subsequent fruit body derived lines, while the two non-fruit body generating hybrid lines formed the out group of the first major cluster. Screening of a Pleurotus type hybrid line having high biological efficiency and generation of a temperature tolerant Pleurotus type line through backcross mating between a non-fruit body generating somatic hybrid and V. volvacea parent are the key achievements of this fusion programme.  相似文献

Mus terricolor I, II and III are the three chromosomal species which differ in stable autosomal short-arm heterochromatin variations established in homozygous condition. Analysis of meiosis in the laboratory-generated F1 male hybrids from crosses (both ways) betweenM. terricolor I and II and betweenM. terricolor I and III shows high frequencies of pairing abnormalities at pachytene. The backcross (N3 generation) male hybrids betweenM. terricolor I and II have meiotic abnormalities as in the F1male hybrids, though to a lesser extent. They show difference in pairing abnormalities in the different karyotypic forms; the backcross hybrids heterozygous for the heterochromatic short arms have more anomalies compared to the homokaryotypic hybrids. This suggests a negative influence of the heterochromatin heterozygosity in meiotic pairing. The results indicate a role for heterochromatin variations in the development of a reproductive barrier in the speciatingM. terricolor complex.  相似文献
