Summary In common with other cyclostomata, the Japanese river lamprey (Lampetra japonica) has a retina consisting of distinct types of photoreceptor cells called long and short photoreceptor cells. After freeze-fracture, disc membranes of these photoreceptor cells were characterized in common by a homogeneous distribution of intramembrane particles on the protoplasmic fracture faces, in contrast to those of the myeloid bodies bearing scattering particles.Immunofluorescent examination was applied to the retina with monoclonal antibodies raised against bovine and chicken rhodopsins. Positive immunoreactivity was found to be limited to outer segments of the short cell, leaving the entire body of the long cell and all other components of the retina negative. The results suggest that the short cell is more closely related to a rod-type photoreceptor cell characterized by rhodopsin as its visual pigment.  相似文献

Summary Horseradish peroxidase (HRP) uptake (through a Corneal incision) in photoreceptor cells of the compound eye of Boettcherisca peregrina, resulted in the labeling of two types of very long visual fibers. One of them (the long fiber, If) penetrates through the lamina and medulla, and directly terminates within the lobula. The other (the bypass fiber, bpf) terminates in the medulla, like the axons of R7 and R8 photoreceptor cells, but the fibers run a considerably roundabout course. The photoreceptor cells with these very long fibers are region-dependent within the retina. Both the If and bpf are found almost exclusively in the male fly.  相似文献

Summary Ion-selective microelectrodes inserted into the compound eyes of Calliphora, Locusta and Apis were used to monitor the changes in extracellular concentration of Ca2+ (Cao) brought about by a 1-min exposure to white light (maximal luminous intensity ca. 103 cd/m2).In the blowfly retina such stimulation causes a decrease in Cao. At high light intensities the Cao signal is phasic, falling over about 6 s to a transient light-induced minimum (Cao= -6.2% ± 0.4%, n = 20, SE) and then rising to an approximately stable plateau (-3.3% ± 0.6%). In migratory locusts the light-induced minimum corresponds to a Cao of -13.8% ± 1.6% (n = 10), and at the plateau the Cao decrease is-13.2% ± 1.5%. In honey-bees Cao at first decreases only slightly, by -2.6% ± 1.0% (n = 10); by the end of the 1-min stimulus the extracellular concentration averages 33.6% ± 14.6% above the dark level.The results suggest a relationship between the position of the characteristic curve of the photoreceptor in the dark-adapted state, the occurrence of quantum bumps, and light-induced increases or decreases in Cao. We investigated the mechanism by which rat retina conserves docosahexaenoic acid during essential fatty acid deficiency. Weanling female albino rats were fed diets containing either 10% by weight hydrogenated coconut oil, safflower oil, or linseed oil for 15 weeks. Plasma and rod outer segment (ROS) membranes were prepared for fatty acid and phospholipid molecular species analysis. In addition, retinas were removed for morphometric analysis. We found the following: (1) Plasma phospholipids and cholesterol esters from coconut oil, safflower oil, and linseed oil diet groups were enriched in 20:3(n-9), 20:4(n-6), and 20:5(n-3), respectively. The levels of these 20-carbon fatty acids in the ROS, however, were only slightly affected by diet. (2) The fatty acids and molecular species of ROS phospholipids from the safflower oil and coconut oil groups showed a selective replacement of 22:6(n-3) with 22:5(n-6), as evidenced by a reduction of the 22:6(n-3)-22:6(n-3) molecular species and an increase in the 22:5(n-6)-22:6(n-3) species. (3) The renewal rate of ROS integral proteins, determined by autoradiography, was 10% per day for each diet group. (4) Morphometric analysis of retinas showed no differences in the outer nuclear layer area or in ROS length between the three groups. We conclude that the conservation of 22:6(n-3) in ROS is not accomplished through reductions in the rate of membrane turnover, the total amount of ROS membranes, or in the number of rod cells. The retina may conserve 22:6(n-3) through recycling within the retina or between the retina and the pigment epithelium, or through the selective uptake of 22-carbon polyunsaturated fatty acids from the circulation.  相似文献

Summary Photoreceptor cells, nerve cells and supporting cells were dissociated from the pineal organ of the river lamprey, Lampetra japonica, by the use of 10 U/ml papain solution at 28° C for 20 min, followed by repeated trituration. With the aid of Nomarski interferencecontrast optics, photoreceptor cells, nerve cells and supporting cells were readily identified. Electron-microscopic examination revealed that isolated photoreceptor cells display an outer segment endowed with a few lamellar disks and connected to the inner segment (ellipsoid) via a connecting cilium. The structural features of the dissociated photoreceptor and supporting cells strongly resemble the morphology of the respective cellular elements in situ. We succeeded in culturing dissociated cells for time periods up to 48 h when the procedure described in detail was applied.  相似文献

Summary In this study, immunohistochemistry on cryostat sections is used to demonstrate anti-histamine immunoreactivity in the Drosophila brain. The results support earlier findings that histamine is probably a transmitter of insect photoreceptors. It is further shown that, in Drosophila, all imaginal photoreceptors including receptor type R7 are anti-histamine immunoreactive, whereas the larval photoreceptors do not seem to contain histamine. In addition to the photoreceptors, fibres in the antennal nerve and approximately 12 neurons in each brain hemisphere show strong histamine-like immunoreactivity. Summary Histological staining of wild-type and sevenless transgenic Drosophila melanogaster bearing Rh3-lacZ fusion genes permits the selective visualization of polarization-sensitive R7 and R8 photoreceptor cells located along the dorsal anterior eye margin. Diffusion of -galactosidase throughout these cells reveals that they project long axons to the two most peripheral synaptic target rows of the dorsal posterior medulla, defining a specialized marginal zone of this optic lobe. Comparison of the staining patterns of marginal and nonmarginal Rh3-lacZ-expressing photoreceptor cells in the same histological preparations suggests that the marginal cells possess morphologically specialized axons and synaptic terminals. These findings are discussed with reference to the neuroanatomy of the corresponding dorsal marginal eye and optic lobe regions of the larger dipterans Musca and Calliphora, and in relation to the ability of Drosophila to orient to polarized light.  相似文献

Abstract: The aim of the present study was to examine the roles of membrane depolarization, calcium influx, and cyclic AMP synthesis in regulating the stability and inactivation of serotonin N -acetyltransferase activity (NAT) in cultured chick photoreceptor cells. NAT activity was induced by pretreating cells for 6 h with 1 µ M forskolin. Cycloheximide was subsequently added, and the rate of loss of enzyme activity (inactivation) was determined. After induction, in the presence of cycloheximide, NAT activity declined with a half-life of ∼30 min. The rate of inactivation was greatly reduced when depolarizing concentrations of K+, forskolin, 8-bromoadenosine 3',5'-cyclic monophosphate, or 3-isobutyl-1-methylxanthine were added together with cycloheximide. The apparent increase in NAT stability caused by K+ was abolished by addition of EGTA or nifedipine and potentiated by Bay K 8644, indicating the involvement of Ca2+ influx through dihydropyridine-sensitive channels. MDL-12330A, an inhibitor of K+-stimulated cyclic AMP formation, blocked the effect of depolarizing concentrations of K+. This result suggests that the effect of Ca2+ influx on the stability of NAT is at least partially mediated by increased levels of cyclic AMP. Thus, depolarization-evoked Ca2+ influx and cyclic AMP formation have two roles in the regulation of NAT activity in chick photoreceptor cells. First, they stimulate the de novo synthesis of NAT or a regulatory protein required for NAT activity. Second, they increase the half-life of the enzyme, presumably by regulating the turnover of existing enzyme molecules.  相似文献

Phase responses to red and blue light pulses were measured at different times during the circadian cycle (phase response curves, PRC) in the marine unicellular dinoflagellate Gonyaulaxpolyedra Stein. Pulses were given during a 24-h period of darkness; thereafter, cultures were released into constant dim red light for the assessment of phase and period. The results confirmed earlier findings that the Gonyaulax circadian system receives light signals via two distinct input pathways. During the subjective day and for the first 3 h of the subjective night, red and blue light pulses led to identical phase responses. For the rest of the circadian cycle, however, phase responses to pulses of either red or blue light differed drastically both in their amplitude and direction (advances or delays). Thus, the Gonyaulax light PRC is generated by two distinct light responses. One of these represents responses via a light input that is responsive both to red and blue light mainly producing small delays. The other represents responses of a primarily blue-sensitive input system leading to large advances restricted to the subjective night. Summary The eighth retinular cell (R 8) of Grapsus lacks cytoplasmic pigment granules and basically resembles those previously known in the ghost crab Ocypode and the mysid Praunus. Distally located, R 8 comprises four lobes inserted between the outer ends of the seven regular retinular cells (R 1–R 7). A thin cytoplasmic bridge connects these lobes. One lobe adjacent to R 1 contains the nucleus of R 8 and gives rise proximally to the cell's axon. The short distal eighth rhabdomere consists of microvilli (mvl) protruding axially from all four lobes. Similar R 8's were found also in two other crab families and in two other genera of mysids.In Grapsus the eighth rhabdomere is extraordinary in possessing mvl oriented in two orthogonal directions parallel to the mvl of R 1–R 7. The distal 20% of the rhabdom consists of mvl originating exclusively from R 8. These appear in somewhat irregular bands and are alternately oriented parallel to the animal's vertical or horizontal axis. More proximally the retinula contains eleven sectors but the rhabdom still comprises bands of alternating mvl with those from R 8 joined respectively by the rhabdomeres of R 1, 4, and 5 (horizontal) and R 2, 3, 6 and 7 (vertical).
