We examined the possibility of using the acidification and bubblingmethod (ABM) to determine phytoplankton primary production incoastal marine environments. Our results from a number of laboratoryand field experiments, suggested that (i) incubations of samplescan be carried out in glass vials instead of using the normalincubation bottles, and (ii) storage of samples fixed with formalinor glutaraldehyde can be carried out for at least 8 weeks withouta change in radioactivity. In the course of our work, we designeda new manifold removing excess 14C via ABM. A linear correlationbetween the G24-max values obtained by the two techniques (r=0.93)was found On average the ABM gave values 1.8 times higher thanthose yielded by the particulate filtration procedure On a squaremetre basis, the ABM gave values 1.4 times higher than thoseyielded by the particulate filtration procedure. The annualproduction at six marine locations was 57% higher when usingthe ABM. The ABM is superior to the traditional filtration procedure,because filtration artefacts are avoided, precision is improved,the dissolved primary production is included in the measurement,and the immediate amount of field work is considerably reduced. 相似文献
Measurements of biomass and growth rate of two axenic algalcultures were carried out using three different methodologicalapproaches: the specific 14C-labelling of chlorophyll a, [3H]adenineincorporation into DNA and net organic carbon assimilation.Time-course experiments revealed that the specific activitiesof chlorophyll a were significantly higher than the specificactivity of total algal carbon in six of seven experiments.When the specific activity of chlorophyll a is used to calculatethe carbon biomass and growth rate, the carbon biomass of thealgae will thus be underestimated and the specific growth ratewill be too high. Determination of growth rates from incorporationof [3H]adenine gave lower values than those obtained from netorganic carbon assimilation and from 14C incorporation intochlorophyll a. Problems with adenine saturation are suggested.When [3H]adenine is used to measure growth rates in dense algalcultures, additions of >1 µM [3H]adenine are oftenrequired to maximally label the extracellular and intracellularadenine pools and hence DNA. 相似文献

The evolution of the protonephridial terminal organ across Rotifera with particular emphasis on Dicranophorus forcipatus, Encentrum mucronatum and Erignatha clastopis (Rotifera: Dicranophoridae). —Acta Zoologica (Stockholm) 91 : 199–211 We report on the ultrastructure of the protonephridial terminal organ in three species of dicranophorid rotifers (Dicranophorus forcipatus, Encentrum mucronatum and Erignatha clastopis). Differences between the three species relate to shape and size, the morphology of the filter region and the number of microvilli and cilia inside the terminal organ. A comparison across Rotifera indicates that the terminal organs in D. forcipatus display a number of plesiomorphic characters, but are modified in E. mucronatum and Er. clastopis. This is in accordance with the results of phylogenetic analyses suggesting a basal position of D. forcipatus compared with the more derived species E. mucronatum and Er. clastopis. Moreover, we survey available data on the terminal organ in Rotifera and discuss its evolutionary transformations. The protonephridial terminal organ in the common ancestor of Rotifera consisted of a cytoplasmic cylinder with cilia united into a vibratile flame and a single circle of circumciliary microvilli. Depending on the topology on which characters are optimized, the site of ultrafiltration was formed by longitudinal cytoplasmic columns spanned by a fine filter diaphragm or by pores in the wall of the terminal organ. In several taxa of Rotifera, the terminal organ – probably independently – lost its circumciliary microvilli. 相似文献
Five regions are recognized in the accessory glands of the Mediterranean flour moth, Anagasta kuehniella (Zeller), on the basis of cellular morphology and aggregates of secretory material in the lumen. Some variation is found in each of the posterior four regions, especially the third one. In the most anterior region (region 1) the epithelium is composed of a single type of cell, while in each of the other regions there are two classes of cells. The cells of region 1 and one class in each of the other four regions are fairly typical exocrine cells with extensive rough endoplasmic reticula. Secretion is primarily via Golgi-derived vesicles. Apocrine secretion in the form of sloughing off of the apical cytoplasm probably also occurs in all regions but is most prominent in the posterior two regions. One class of cells is very similar in morphology in each of the posterior four regions though their secretory products form characteristic aggregates in the lumen. The second class of cells (foliate cells) occurring in the posterior four segments is most notably characterized by elongate apical projections that extend out into the lumen. The apical projections contain large quantities of glycogen, some microtubules, and, in some cases, many minute mitochondria. The membrane content of the projections is also very high. In the anterior regions, the membranes are mostly fused in pairs and typically form multilayered whorls. Fusion and whorl formation decrease in the posterior regions. The cytoplasm of the foliate cells has a high organelle content including many lysosomes and mitochondria. The latter exhibit considerable polymorphism, with particular forms occurring in the different regions of the glands. The apical projections of the foliate cells are detached during copulation, presumably as the result of nervous stimulation, and become a part of the ejaculate. Species of the freshwater gastrotrich taxon Dasydytidae show a set of conspicuous structural and behavioural adaptations to a semi-planktonic life. Conspicuously, most dasydytids have several groups of strong, moveable spines that can actively be abducted to perform saltatory movements, change the overall direction of locomotion, or enable the animals to rest in a defensive position. So far, there are only vague ideas of how these spine movements are achieved in dasydytid species. In order to gain insight into the possible morpho-functional coupling of body musculature and motile spines, we have carried out a study targeting the muscular system in two species of Dasydytidae by means of phalloidin staining and confocal laser scanning microscopy.
For spine movements in both species studied, Stylochaeta scirtetica and Dasydytes (Setodytes) tongiorgii, we have identified an antagonistic system of segmented longitudinal and oblique somatic muscle pairs. In both species, contraction of the musculi obliqua abduct the paired groups of ventro-lateral spines; contraction of the segments of musculi laterales causes their adduction.
A comparison of the muscular system of the studied species to that of other gastrotrichs reveals several homologous muscle pairs, visceral as well as somatic, that might be features of the stem species of a clade comprising all Paucitubulatina exclusive of Xenotrichulidae. The pairs of oblique somatic muscles are most probably an autapomorphy of Dasydytidae. 相似文献
