A major challenge in neurophysiology has been to characterize the response properties and function of the numerous inhibitory cell types in the cerebral cortex.We here share our strategy for obtaining stable, well-isolated single-unit recordings from identified inhibitory interneurons in the anesthetized mouse cortex using a method developed by Lima and colleagues1. Recordings are performed in mice expressing Channelrhodopsin-2 (ChR2) in specific neuronal subpopulations. Members of the population are identified by their response to a brief flash of blue light. This technique – termed “PINP”, or Photostimulation-assisted Identification of Neuronal Populations – can be implemented with standard extracellular recording equipment. It can serve as an inexpensive and accessible alternative to calcium imaging or visually-guided patching, for the purpose of targeting extracellular recordings to genetically-identified cells. Here we provide a set of guidelines for optimizing the method in everyday practice. We refined our strategy specifically for targeting parvalbumin-positive (PV+) cells, but have found that it works for other interneuron types as well, such as somatostatin-expressing (SOM+) and calretinin-expressing (CR+) interneurons.  相似文献

Parvalbumins beta (β-PRVBs) are considered the major fish allergens. A new strategy for the rapid and direct detection of these allergens in any foodstuff is presented in this work. The proposed methodology is based on the purification of β-PRVBs by treatment with heat, the use of accelerated in-solution trypsin digestion under an ultrasonic field provided by High-Intensity Focused Ultrasound (HIFU) and the monitoring of only nineteen β-PRVB peptide biomarkers by Selected MS/MS Ion Monitoring (SMIM) in a linear ion trap (LIT) mass spectrometer. The present strategy allows the direct detection of the presence of fish β-PRVBs in any food product in less than 2 hours.  相似文献

Hippocampal interneurons are local circuit neurons which are responsible for inhibitory activity in the hippocampus. Parvalbumin (PV) is one of useful markers for GABAergic interneurons, not for principle cells, in the hippocampus. In the present study, we investigated age-related changes in PV immunoreactive neurons and protein levels in the gerbil hippocampus during normal aging. PV immunoreactive neurons were detected in all hippocampal subregions of all groups. PV immunoreactive neurons, which innervated principal neurons, were non-pyramidal neurons in the hippocampal CA1-3 regions, and were polymorphic neurons in the dentate gyrus. In the hippocampal CA1 region, the number of PV immunoreactive neurons was significantly reduced in the postnatal month 3 (PM 3) group, which was sustained by PM 18, and, at PM 24, the number of PV immunoreactive neurons was significantly decreased. In the CA2/3 region and dentate gyrus, the number of PV immunoreactive neurons was significantly decreased at PM 6: Thereafter, the number of PV immunoreactive neurons was sustained until PM 24. In addition, changes in PV protein levels in the gerbil hippocampus were similar to immunohistochemical changes during normal aging: PV protein levels were significantly decreased with age by PM 6: Thereafter, PV protein levels were sustained by PM 24. These results suggest that PV immunoreactive interneurons were decreased in the hippocampus with age in gerbils.  相似文献

对由Morris水迷宫分得的青年、老年记忆正常和记忆减退鼠的脑组织分别进行突触、AChE纤维、突触素、小白蛋白神经元以及突触体钙离子浓度、膜流动性的定量分析。结果表明老年记忆减退鼠新皮质、海马结构突触素含量、突触、胆碱能纤维、小白蛋白阳性神经元密度及突触体膜流动笥较老年记忆正常和青年鼠明显降低。老年记忆正常鼠与青年鼠各项均无显著差异。本研究提示各研究指标的异常与老年学习记忆减退密切相关。  相似文献

The parvalbumin pattern in white muscle from seven Cyprinidae has been determined by starch gel electrophoresis analysis. The immunochemical discrimination of the various components using several monospecific antisera against a number of distinct Cyprinidae parvalbumins has been qualitatively evaluated by immunoelectrophoresis. The results show that immunologically different groups of parvalbumins can be distinguished within these patterns. One of these groups includes at least one component which is present in each of the seven species investigated. A similar component has been found in a species of Siluridae which, like the Cyprinidae, belongs to the well defined systematic superorder of the Ostariophysi.  相似文献

将65例妊娠14天SD大白鼠胚胎视网膜移植至新生第1天大白鼠中脑背面偏左侧,同时摘除新生鼠右眼。应用免疫细胞化学方法,HRP逆行追踪及其结合免疫细胞化学双标记技术,观察手术后第10天至50天移植视网膜、与其相应年龄的正常和宿主视网膜免疫反应(Parvalbuminimmunoreactive,PV-Ⅰ)神经元的发育规律和定位分布,以及移植视网膜与宿主上丘的联系。结果表明:移植视网膜具有正常视网膜各层结构和生长发育规律;移植和正常视网膜PV-Ⅰ神经元是无长突细胞、移位无长突细胞和节细胞。PV-Ⅰ神经元在生后第3天移植视网膜的节细胞层出现,在生后第14天至19天移植视网膜中达很高程度,而在生后第3天移植视网膜节细胞层中有些PV-Ⅰ神经元同时具有HRP标记,提示这些细胞是节细胞,并与上丘建立了联系。  相似文献

Recent evidence has linked striatal amino-3-hydroxy-5-methyl-4-isoxazole propionic acid (AMPA) receptor function to the adverse effects of long-term dopaminergic treatment in Parkinson’s disease. The phosphorylation of AMPA subunit, GluR1, reflects AMPA receptor activity. To determine whether serine phosphorylation of GluR1 subunit by activation of Ca2+/calmodulin-dependent protein kinase II (CaMKII) contributes to the process, we examined the effects of unilateral nigrostriatal depletion with 6-hydroxydopamine and subsequent l-dopa treatment on motor responses and phosphorylation states. Three weeks of l-dopa administration to rats shortened the duration of the rotational response. We found a significant reduction in the abundance of both phosphorylated GluR1 at serine-831 site (pGluR1S831) and GluR1 in the cell plasma membrane of lesioned striatum. Chronic treatment of lesioned rats with l-dopa markedly upregulated the phosphorylation of GluR1 in lesioned striatum with a concomitant normalization of the plasma membrane GluR1 abundance, which lasted at least 1 day after withdrawal of chronic l-dopa treatment. Our immunostaining data showed that these changes were confined to parvalbumin-positive neurons where GluR1 subunits are exclusively expressed. Both the altered motor response duration and the degree of pGluR1S831 were attenuated by the intrastriatal administration of CaMKII inhibitor KN-93. These findings suggest that activation of CaMKII contributes to both development and maintenance of motor response duration alterations, through a mechanism that involves an increase in pGluR1S831 within parvalbumin-positive neurons.Maowen Ba and Min Kong are contributed equally to this work.  相似文献

The intrinsic circuitry of the motor cortex comprises a complex network of connections whose synaptic relationships are poorly understood. This study was designed to determine the characteristics of subsets of GABAergic neurons containing the calcium-binding proteins parvalbumin (PV) and calbindin (CB), and their relationships with intrinsic axons in motor cortex. Immunohistochemically identified PV-containing neuronal profiles were more evenly distributed across cortical laminae (38% in II-III, 32% in V, 30% in VI) and more numerous (2.1/1) than CB-containing neuronal profiles (71% in II-III, 17% in V, 12% in VI). Relationships between neurons and axons intrinsic to motor cortex were visualized with fluorescent markers using the laser scanning confocal microscope. Similar percentages of PV (43%) and CBimmunoreactive (IR) (40%) neurons formed sparsely distributed appositions (1-5/neuron) with anterogradely labeled axons. The mean distances of such appositions from the somata were significantly different for the two groups (PV, mean =22 mum, range = 1.6-93 mum; CB, mean = 32 mum, range = 6.2-132 mum). PV-IR neurons had a lower ratio of axosomatic/ axodendritic appositions (1/99) compared with CB-IR neurons (14/86). Ultrastructural studies confirmed these findings. Fifty-seven percent of CB-IR neurons and 38% of PV-IR neurons formed synapses with intrinsic axons. Both populations received sparse input (1-6 synapses/neuron). Nearly all appositions between labeled terminals and postsynaptic profiles formed one synapse. Postsynaptic dendrites of PV-IR neurons (mean = 1.4 mum diameter) were larger than those of CB-IR neurons (mean = 1.1 mum), indicating more proximal synapses. Distinct input patterns of intrinsic axons to the two populations of neurons suggest unique roles in cortical processing.  相似文献

Carboxylate (COO) groups can coordinate to metal ions in of the following four modes: ‘unidentate’, ‘bidentate’, ‘bridging’ and ‘pseudo-bridging’ modes. COO stretching frequencies provide information about the coordination modes of COO groups to metal ions. We review the Fourier-transform infrared spectroscopy (FTIR) of side-chain COO groups of Ca2+-binding proteins: pike parvalbumin pI 4.10, bovine calmodulin and Akazara scallop troponin C. FTIR spectroscopy of Akazara scallop troponin C has demonstrated that the coordination structure of Mg2+ is distinctly different from that of Ca2+ in the Ca2+-binding site. The assignments of the COO antisymmetric stretch have been ensured on the basis of the spectra of calcium-binding peptide analogues. The downshift of the COO antisymmetric stretching mode from 1565 cm-1 to 1555-1540 cm−1 upon Ca2+ binding is a commonly observed feature of FTIR spectra for EF-hand proteins.  相似文献
