Until 2000, efforts into organising tissue banks in Brazil had not progressed far beyond small “in house” tissue storage repositories,
usually annexed to Orthopaedic Surgery Services. Despite the professional entrepreneurship of those working as part time tissue
bankers in such operations, best practices in tissue banking were not always followed due to the lack of regulatory standards,
specialised training, adequate facilities and dedicated personnel. The Skin Bank of the Plastic Surgery Department of the
Hospital das Clinicas of Sao Paulo, the single skin bank in Brazil, was not an exception. Since 1956, restricted and unpredictable
amounts of skin allografts were stored under refrigeration for short periods under very limited quality controls. As in most
“tissue banks” at that time in Brazil, medical and nursing staff worked on a volunteer and informal basis undergoing no specific
training. IAEA supported the implementation of the tissue banking program in Brazil through the regional project RLA/7/009
“Quality system for the production of irradiated sterilised grafts” (1998–2000) and through two interregional projects INT/6/049 “Interregional Centre of Excellence in Tissue Banking”, during the period 2002–2004 and INT/6/052 “Improving the Quality of Production and Uses of Radiation Sterilised Tissue Grafts”, during the period 2002–2004. In 2001–2002, the first two years of operation of the HC-Tissue Bank, 53 skin transplants
were carried out instead of the previous 4–5 a year. During this period, 75 individuals donated skin tissue, generating approximately
90,000 cm2 of skin graft. The IAEA program were of great benefit to Brazilian tissue banking which has evolved from scattered make shift

The wild type lactose repressor of Escherichia coli is a tetrameric protein formed by two identical dimers. They are associated via a C-terminal 4-helix bundle (called tetramerization domain) whose stability is ensured by the interaction of leucine zipper motifs. Upon in vitro γ-irradiation the repressor losses its ability to bind the operator DNA sequence due to damage of its DNA-binding domains. Using an engineered dimeric repressor for comparison, we show here that irradiation induces also the change of repressor oligomerisation state from tetramer to dimer. The splitting of the tetramer into dimers can result from the oxidation of the leucine residues of the tetramerization domain. 相似文献

Plant plasma membrane aquaporins facilitate water flux into and out of plant cells, thus coupling their cellular function to basic aspects of plant physiology. Posttranslational modifications of conserved phosphorylation sites, changes in cytoplasmic pH and the binding of Ca2+ can regulate water transport activity by gating the plasma membrane aquaporins. A structural mechanism unifying these diverse biochemical signals has emerged for the spinach aquaporin SoPIP2;1, although several questions concerning the opening mechanism remain. Here, we describe the X-ray structures of the S115E and S274E single SoPIP2;1 mutants and the corresponding double mutant. Phosphorylation of these serines is believed to increase water transport activity of SoPIP2;1 by opening the channel. However, all mutants crystallised in a closed conformation, as confirmed by water transport assays, implying that neither substitution fully mimics the phosphorylated state. Nevertheless, a half-turn extension of transmembrane helix 1 occurs upon the substitution of Ser115, which draws the Cα atom of Glu31 10 Å away from its wild-type conformation, thereby disrupting the divalent cation binding site involved in the gating mechanism. Mutation of Ser274 disorders the C-terminus but no other significant conformational changes are observed. Inspection of the hydrogen-bond interactions within loop D suggested that the phosphorylation of Ser188 may also produce an open channel, and this was supported by an increased water transport activity for the S188E mutant and molecular dynamics simulations. Question: How does above‐ground net primary production (ANPP) differ (estimated from remotely sensed data) among vegetation units in sub‐humid temperate grasslands? Location: Centre‐north Uruguay. Methods: A vegetation map of the study area was generated from LANDSAT imagery and the landscape configuration described. The functional heterogeneity of mapping units was analysed in terms of the fraction of photosynthetically active radiation absorbed by green vegetation (fPAR), calculated from the normalized difference vegetation index (NDVI) images provided by the moderate resolution imaging spectroradiometer (MODIS) sensor. Finally, the ANPP of each grassland class was estimated using NDVI and climatic data. Results: Supervised classification presented a good overall accuracy and moderate to good average accuracy for grassland classes. Meso‐xerophytic grasslands occupied 45% of the area, Meso‐hydrophytic grasslands 43% and Lithophytic steppes 6%. The landscape was shaped by a matrix of large, unfragmented patches of Meso‐xerophytic and Meso‐hydrophytic grasslands. The region presented the lowest anthropic fragmentation degree reported for the Rio de la Plata grasslands. All grassland units showed bimodal annual fPAR seasonality, with spring and autumn peaks. Meso‐hydrophytic grasslands showed a radiation interception 10% higher than the other units. On an annual basis, Meso‐hydrophytic grasslands produced 3800 kg dry matter (DM) ha?1 yr?1 and Meso‐xerophytic grasslands and Lithophytic steppes around 3400 kg·DM·ha?1·yr?1. Meso‐xerophytic grasslands had the largest spatial variation during most of the year. The ANPP temporal variation was higher than the fPAR variability. Conclusions: Our results provide valuable information for grazing management (identifying spatial and temporal variations of ANPP) and grassland conservation (identifying the spatial distribution of vegetation units). 相似文献

PurposeTo investigate and improve the domestic standard of radiation therapy in the Republic of Korea.MethodsOn-site audits were performed for 13 institutions in the Republic of Korea. Six items were investigated by on-site visits of each radiation therapy institution, including collimator, gantry, and couch rotation isocenter check; coincidence between light and radiation fields; photon beam flatness and symmetry; electron beam flatness and symmetry; physical wedge transmission factors; and photon beam and electron beam outputs.ResultsThe average deviations of mechanical collimator, gantry, and couch rotation isocenter were less than 1 mm. Those of radiation isocenter were also less than 1 mm. The average difference between light and radiation fields was 0.9 ± 0.6 mm for the field size of 20 cm × 20 cm. The average values of flatness and symmetry of the photon beams were 2.9% ± 0.6% and 1.1% ± 0.7%, respectively. Those of electron beams were 2.5% ± 0.7% and 0.6% ± 1.0%, respectively. Every institutions showed wedge transmission factor deviations less than 2% except one institution. The output deviations of both photon and electron beams were less than ±3% for every institution.ConclusionsThrough the on-site audit program, we could effectively detect an inappropriately operating linacs and provide some recommendations. The standard of radiation therapy in Korea is expected to improve through such on-site audits. 相似文献

The appearance of a malignant disease during pregnancy is relatively rare. The use of external-beam radiation therapy is limited to non-pelvic tumors which are usually located above the diaphragm. However, supradiaphragmatic radiotherapy unavoidably exposes the fetus to secondary radiation due to head leakage, scatter from the machine and scatter produced inside the patient. This fetal exposure may be associated with an elevated risk for the development of deterministic harmful effects and/or carcinogenesis. The decision about the administration of radiotherapy in a pregnant patient is influenced by the fetal dose which must always be estimated before the patient’s treatment course. The methods employed for fetal dosimetry in external-beam radiotherapy are described in this review study. Direct dose measurements using thermoluminescent dosemeters or large ionization chambers placed on physical phantoms may be used. Monte Carlo simulations on computational phantoms may also provide accurate fetal dose calculations. The physical and/or computational phantoms need to simulate the full-scatter geometry of the pregnant patient. Typical fetal dose values attributable to radiation therapy for brain tumors, head and neck cancer, breast carcinoma and Hodgkin lymphoma at the first, second and third trimesters of gestation are presented. The effectiveness of different shielding devices for fetal dose reduction in radiotherapy is discussed. The effect of the dimensions and setup of the shielding material on the radiation dose received by the fetus is described. Moreover, practical methods for reducing the fetal dose by selecting the appropriate irradiation parameters are presented. 相似文献

Secretins form large oligomeric assemblies in the membrane that control both macromolecular secretion and uptake. Several Pasteurellaceae are naturally competent for transformation, but the mechanism for DNA assimilation is largely unknown. In Haemophilus influenzae, the secretin ComE has been demonstrated to be essential for DNA uptake. In closely related Aggregatibacter actinomycetemcomitans, an opportunistic pathogen in periodontitis, the ComE homolog HofQ is believed to be the outer membrane DNA translocase. Here, we report the structure of the extra-membranous domains of HofQ at 2.3 Å resolution by X-ray crystallography. We also show that the extra-membranous domains of HofQ are capable of DNA binding. The structure reveals two secretin-like folds, the first of which is formed by means of a domain swap. The second domain displays extensive structural similarity to K homology (KH) domains, including the presence of a GxxG motif, which is essential for the nucleotide-binding function of KH domains, suggesting a possible mechanism for DNA binding by HofQ. The data indicate a direct involvement in DNA acquisition and provide insight into the molecular basis for natural competence. 相似文献
