Eukaryotic cells have the ability to uptake and transport endogenous and exogenous DNA in their nuclei, however little is known about the specific pathways involved. Here we show that the nuclear transport receptor importin 7 (imp7) supports nuclear import of supercoiled plasmid DNA and human mitochondrial DNA in a Ran and energy‐dependent way. The imp7‐dependent pathway was specifically competed by excess DNA but not by excess of maltose‐binding protein fused with the classical nuclear localizing signal (NLS) or the M9 peptides. Transport of DNA molecules complexed with poly‐l ‐lysine was impaired in intact cells depleted of imp7, and DNA complexes remained localized in the cytoplasm. Poor DNA nuclear import in cells depleted of imp7 directly correlated with lower gene expression levels in these cells compared to controls. Inefficient nuclear import of transfected DNA induced greater upregulation of the interferon pathway, suggesting that rapid DNA nuclear import may prevent uncontrolled activation of the innate immune response. Our results provide evidence that imp7 is a non‐redundant component of an intrinsic pathway in mammalian cells for efficient accumulation of exogenous and endogenous DNA in the nucleus, which may be critical for the exchange of genetic information between mitochondria and nuclear genomes and to control activation of the innate immune response . 相似文献

Phylogenies with even a rough time scale can be used to investigate the history of non‐volant taxa with disjunct distributions in widely separated land areas that were once connected. Basic methods for doing this are discussed. A partial phylogeny of Mabuya based on mtDNA (305 bp cytochrome b, 379 bp 12S rRNA and 388 bp 16S rRNA) is used to show that this genus invaded tropical America from Africa twice in the last 9 Myr, once reaching the American mainland and once the oceanic island of Fernando de Noronha, two journeys each of at least 3000 km. In general, phylogenetic evidence for multiple invasions is less equivocal than that suggesting a single invasion, which is more prone to sampling artefacts. Two alternative hypotheses explaining the presence of Mabuya in both Africa and tropical America are refuted on the basis of molecular clock considerations, namely that the occurrence of Mabuya in these continents pre‐dated their separation over 100 My ago and that it was introduced from one continent to the other by human activities. Like several other lizard groups that have made successful long‐distance transmarine colonizations, Mabuya has done this on many occasions. Phylogenetic results are also compatible with a SE Asian or Australasian origin of Mabuya followed by westward expansion. 相似文献

MtDNA sequences (396 bp cytochrome b and 369 bp 12S rRNA) from recent material and old museum specimens indicate Pleurodeles poireti and P. waltl form independent clades with 7.76% genetic divergence. Within P. poireti, populations from Djebel Edough, NE Algeria are very distinct with 6.12% genetic divergence from the remainder and may deserve separate species status. Away from Djebel Edough, P. poireti consists of three distinct clades (coastal NW Tunisia; central N Algeria; Constantine plus inland NW Tunisia) with a maximum genetic divergence of only 1%. P. waltl contains two clades with 2.96% genetic divergence, one in SE and E Spain plus north Morocco, the other in Portugal and SW and central Spain. Pleurodeles probably invaded NW Africa from SW Europe during the Messinian Salinity Crisis, when land contact was first established at 5.6 Ma, and then interrupted at 5.3 Ma. Molecular clocks, calibrated in the assumption that the latter event separated P. waltl and P. poireti, suggest that Pleurodeles diverged from its sister taxon, Tylototriton, at about 8.6–10 Ma. Djebel Edough P. poireti separated at about 4.2 Ma, perhaps through isolation on a temporary, now ‘fossil’, island initiated by the Messinian crisis. Differentiation in remaining P. poireti may have been caused by Pleistocene climatic fluctuations, while bifurcation in P. waltl appears to have taken place in the Pliocene approximately between 3.2 and 2 Ma. This species reached Morocco very recently, perhaps as a result of human introduction. Use in Pleurodeles of the slower divergence rates estimated in some other salamandrids results in a less parsimonious historical hypothesis that does not fit known geophysical events. 相似文献

Darevskia praticola differs from the other species of the genus in having a large but disjunct distribution, covering the Balkan and the Caucasus regions. Furthermore, most Darevskia species occupy saxicolous habitats, whereas D. praticola inhabits meadows and forest environments. Here we determine the phylogeographic and phylogenetic relationships of Darevskia praticola sensu lato and evaluate the current, morphology-based taxonomy. We sequenced two mtDNA genes (Cyt-b and ND4) and two nuclear loci (MC1R and RELN) for samples collected across the species range. Because our sequences amplified with the Cyt-b primers appear to represent a nuclear pseudogene we excluded this marker from the final analysis. Our results support monophyly of D. praticola and show its division into three clades. The first divergence, dated to the Late Pliocene, is between the Balkans and the Caucasus. The Caucasus lineage is further subdivided in a western Greater Caucasus and a Transcaucasia clade, likely due to subsequent differentiation during the Pleistocene. Our findings do not support the current taxonomic arrangement within D. praticola. The main geographic divergence likely happened due to a vicariance event associated with Plio-Pleistocene climatic and vegetation oscillations. 相似文献

The conservation of unique populations of animals is critical in order to preserve valuable genetic diversity and, where populations are free‐living, maintain their irreplaceable influence upon habitat ecology. An accurate assessment of genetic diversity and structure within and between populations is crucial in order to design and implement conservation strategies in natural and domesticated species. Moreover, where it is possible to identify relic populations that are related to a structured breed an ideal opportunity presents itself to model processes that reveal historical factors that have shaped genetic diversity. The origins of native UK mountain and moorland ponies are uncertain, but they may have directly descended from prehistoric populations and potentially harbour specific adaptations to the uplands of Britain and Ireland. To date, there have been no studies of population structure and genetic diversity present within a free‐living group of ponies in the Carneddau mountain range of North Wales. Herein, we describe the use of microsatellites and SNPs together with analysis of the mitochondrial control region to quantify the extent and magnitude of genetic diversity present in the feral Carneddau pony and relate this to several recognised British and Irish pony breeds. Our results establish that the feral Carneddau ponies represent a unique and distinctive population that merits recognition as a defined population and conservation priority. We discuss the implications for conservation of this population as a unique pool of genetic diversity adapted to the British uplands and potentially of particular value in maintaining the biodiversity of these habitats. 相似文献

Summary. By combining different methods we evaluate whether the ant Tetramorium rhenanum is specifically separated from T. moravicum or whether it is a conspecific microgyne form. High-precision morphometry shows a clear difference in queen size. Sequence comparison of 1031 bp of COI reveals that T. rhenanum falls into a clade with T. moravicum, which is significantly separated from T. forte and T. chefketi. T. rhenanum shares at least two haplotypes with T. moravicum and is considered as a junior synonym. Sexual behaviour and colony foundation experiments corroborate conspecificity. Over eight kilobases (kb) of sequence from eight genes including two mitochondrial loci, Cyt b and 12S, and six nuclear loci, B2m , Zp3 , Tcp1, Sry, Smcx and Smcy , were used to investigate phylogenetic relationships among 11 taxa representing eight species within the rodent genus Mus . Particular attention was given to discerning relationships among species within the subgenus Mus including members of a Palearctic clade ( M. musculus , M. spicilegus , M. macedonicus and M. spretus ) and members of an Asian clade ( M. caroli , M. cookii and M. cervicolor ), as previous studies using different datasets have produced different topologies for taxa within these two groups. While parsimony analyses of the combined eight-gene dataset yielded a single, fully resolved tree, support values were lower for nodes resolving relationships within the Palearctic and Asian clades than they were elsewhere in the tree. In addition, a maximum likelihood analysis of the same eight-gene dataset yielded different topologies for both the Palearctic and the Asian clades. Both observations are indicative of clade instability. The nature of this instability was explored through a comparison with our previous study in which we included the two mitochondrial loci and only four of the six nuclear genes, and through an analysis of partitioned data, specifically mitochondrial vs. nuclear genes. The phylogeographical patterns and demographic history of mitochondrial DNA (cytochrome b, N = 327; D‐loop, N = 252) and nuclear DNA (IRBP gene, N = 235) haplotypes were studied for the Meriones meridianus complex in northern China, a desert‐dwelling gerbil species complex. The phylogenetic analyses, which were performed on the separate and combined (mitochondrial + nuclear) datasets, revealed two divergent clades (Clade A and Clade B) corresponding to distinct geographical regions. Clade A contained the haplotypes found mostly in individuals from the Tianshan Mountains area. Clade B contained haplotypes from populations located in other deserts in northern China. The divergence times indicated that the history of the M. meridianus complex was influenced by the uplift of the Tianshan Mountains and climate‐induced habitat fluctuations. In the Pleistocene, the expansion of forests and grasslands during interglacial period led to the isolation of the M. meridianus complex, which preferred to inhabit deserts. Hence, long geological isolation and the M. meridianus complex adaptation to local ecological conditions led to its genetic divergence. Clade A had long‐lasting demographic stability, most likely because the populations of this clade remained in a stable desert environment for a long time. However, the extension of other deserts and disappearance of palaeolakes during the last glacial period resulted in demographic expansion of Clade B. The aim of this work was to gather information about the origin and genetic characterization of the Central European Hucul horse based on 71 horses using 17 microsatellites and the D‐loop region of mtDNA. Their genetic relationship to the Polish Konik (N = 7), German (N = 4) and Hungarian wild Przewalski horses (N = 4) and 200 horse sequences from GenBank was also analysed. Both microsatellite and mtDNA analysis showed a high genetic variation in the Hucul. A total of 130 alleles were detected, the mean number of observed alleles per microsatellite was 7.647, and the number of effective alleles was 4.401. The average observed and expected heterozygosity were 0.706 and 0.747, respectively. The high heterozygosity values and Wright's fixation index (FIS) (?0.128) indicated a low level of inbreeding, low or no selection pressure, and large number of alleles. mtDNA analysis revealed 18 haplotypes for the Hucul population with a total of 23 variable sites. Haplotype and nucleotide diversities were 0.935 ± 0.011 and 0.022 ± 0.012, respectively. Neutrality tests (Tajima's D and Fu's Fs) were non‐significant, and mismatch distribution was ragged, indicating that the Hucul population is in genetic equilibrium. The most frequent mtDNA D‐loop region belonged to haplogroup A (48%), which was also present in Przewalski Wild horse samples, while Polish Konik samples belonged to three haplotypes and C, F, and G haplogroups. Large and significant pairwise ΦST values along with a small number of common haplotypes indicated a low level of gene flow and lack of genetic structure among the three studied breeds (Hucul, Konik, and Przewalski Wild horse). Nucleotide polymorphism at 12 nuclear loci and two mitochondrial gene fragments was studied in three closely related pine species from the Pinus mugo complex in populations across the species distributional range in Europe. Despite large differences in the census sizes of the populations, high and similar levels of nucleotide diversity (θsil = ~0.013–0.017) were found at nuclear loci in the three pine species. More rapid decay of overall linkage disequilibrium (LD) and recombination to diversity ratio (ρ/θ) was observed across the species distributional range in P. mugo (ρ = 0.0369 ± 0.0028; ρ/θ = ~2.2) than in P. uncinata (ρ = 0.0054 ± 0.0011; ρ/θ = ~0.4) and P. uliginosa (ρ = 0.0051 ± 0.0010, ρ/θ = ~0.4). However, regional groups of P. mugo showed similar levels of LD and ρ/θ ratio to the other species. An excess of rare nucleotide variants was found in P. mugo at four loci, but, overall, the allelic frequency spectrum in the three species did not deviate significantly from neutrality (multilocus Tajima's D = ?0.681, D = ?0.118 and D = ?0.266, P > 0.05, respectively). Bayesian clustering methods showed no clear correspondence of clusters to species or geographical regions. Some differences between populations and species were found in a hierarchical analysis of molecular variance (AMOVA) and in the distribution of the mitochondrial DNA haplotypes, suggesting rather limited gene flow between the taxa and ongoing divergence.
