Drug mediated induction of cytochrome P450 was studied in cultures of hepatocytes that had never been cultured in the presence of serum. Propylisopropylacetamide induced a five-fold increase in cytochrome P450, approximating in ovo induced levels, when triiodothyronine and/or dexamethasone were included in the culture medium. Insulin was apparently not required for this induction. Cytochrome P450, free of cytochrome oxidase, could be fully recovered from cell homogenates in a 8700g supernatant, by use of a buffer containing 0.2% Emulgen.  相似文献

Nucleotide sequence has been determined for the restriction fragments and cloned DNA from the pL-N-tL1 region of bacteriophage lambda. A unique reading frame for the N gene is defined by the absence of natural nonsense codons and by the presence of seven nonsense codons generated by mutations in N. This reading frame is initiated at two alternative ATG codons, the second of which is probably the in vivo translation start. Reading is stopped at a single TAG codon. The protein coded is therefore 133 or, more probably, 107 amino acids long, rich in lysine, arginine and proline.  相似文献

Leaf pocket resins of 11 species of the tropical arborescent genus Hymenaea are virtually identical qualitatively, but of widely varying quantitative proportions. Within this large range of variability, several strong positive quantitative correlations between resin constituents were found, especially between caryophyllene and β-humulene and between γ-muurolene and δ-cadinene. These data lead to clarification of sesquiterpene biosynthetic routes in Hymenaea. In addition, quantitative relationships found among caryophyllene, α- and β-selinene, γ-muurolene and δ-cadinene are explained only with difficulty by long accepted biosynthetic pathways, and the intermediacy of germacrenes is suggested.  相似文献

Phytochrome induces δ-aminolevulinate dehydratase (ALAD) activity in radish seedling cotyledons under continuous far red light. Analysis of the enzymatic activity in etioplasts vs total activity shows a constant ALAD level in these organelles (10 %) in etiolated seedlings. In far red irradiated seedlings, the percentage of enzyme detected into etioplasts increases up to 45 % of the total. Comparative kinetic studies of ALAD activity detected in the cytoplasm and the etioplasts indicate an increase in both compartments with a maximum value reached respectively at 96 and 120 hr from sowing. Treatment with cycloheximide shows a very fast abolition of cytoplasmic ALAD activity which is always correlated to an etioplast decrease with a time shift of ca 24 hr. Erythromycin acts only on the cytoplasmic level of ALAD, and only for far red irradiated seedlings, with an increase of activity twice the level detected in untreated ones. This unexpected effect is discussed.  相似文献

在水热条件下合成纳米δ-Bi2O3负载多孔沸石球填料(Bi-PZSF),研究其对水体中Cr(Ⅵ)的吸附性能(包括等温吸附、吸附动力学和吸附稳定性)以及溶解氧、pH等因素对吸附过程的影响及其吸附机理。结果表明:朗格缪尔模型和准二级动力学模型对实验结果具有良好的拟合度,在中性条件下,水体中的溶解氧含量对吸附速率影响较小,Bi-PZSF的最大吸附量为955.69 mg·kg^-1,比天然沸石的吸附量提升了近120倍;Bi-PZSF在NO3^-和SO4^2-的溶液中呈现出较强的选择吸收Cr(Ⅵ)能力,Cr(Ⅵ)去除率均在97%以上;同时,Bi-PZSF在吸附Cr(Ⅵ)后表现出高稳定性。Cr(Ⅵ)的去除主要是通过与附着在Bi-PZSF上的纳米δ-Bi2O3的表面羟基交换完成的。  相似文献

Difference among colonies in the population structure of otariids can be driven by philopatry and/or by specializations in the foraging ecology of females. In northern Patagonia, the South American sea lion (SASL) shows some degree of spatial genetic structure among colonies from north and south zones. This study aims to explore the isotopic niche of SASL females in the last period of the pregnancy from different colonies of northern Patagonia and to consider whether the fine scale genetic spatial structuring is potentially related to variation in trophic resources. Stable isotope analysis was performed on 101 skin samples of newborn pups in 10 colonies, as a proxy for the feeding ecology of their mothers. Differences among colonies in the metrics studied revealed the plasticity of the species and support individual trophic specialization of SASL females at a small geographic scale. Also, significant differences were found in all isotopic metrics between the north and south zones. Several hypotheses were proposed to explain the differences in SASL females' isotope values (e.g., use of different foraging areas or prey, isotopic baseline variation). Nonetheless, further research is needed to better understand the relation between fine scale genetic structuring and the foraging ecology of SASL females.  相似文献

Stable-isotope analysis (SIA) provides a valuable tool to address complex questions pertaining to elasmobranch ecology. Liver, a metabolically active, high turnover tissue (~166 days for 95% turnover), has the potential to reveal novel insights into recent feeding/movement behaviours of this diverse group. To date, limited work has used this tissue, but ecological application of SIA in liver requires consideration of tissue preparation techniques given the potential for high concentrations of urea and lipid that could bias δ13C and δ15N values (i.e., result in artificially lower δ13C and δ15N values). Here we investigated the effectiveness of (a) deionized water washing (WW) for urea removal from liver tissue and (b) chloroform-methanol for extraction of lipids from this lipid rich tissue. We then (a) established C:N thresholds for deriving ecologically relevant liver isotopic values given complications of removing all lipid and (b) undertook a preliminary comparison of δ13C values between tissue pairs (muscle and liver) to test if observed isotopic differences correlated with known movement behaviour. Tests were conducted on four large shark species: the dusky (DUS, Carcharhinus obscurus), sand tiger (RAG, Carcharias taurus), scalloped hammerhead (SCA, Sphyrna lewini) and white shark (GRE, Carcharodon carcharias). There was no significant difference in δ15N values between lipid-extracted (LE) liver and lipid-extracted/water washed (WW) treatments, however, WW resulted in significant increases in %N, δ13C and %C. Following lipid extraction (repeated three times), some samples were still biased by lipids. Our species-specific “C:N thresholds” provide a method to derive ecologically viable isotope data given the complexities of this lipid rich tissue (C:N thresholds of 4.0, 3.6, 4.7 and 3.9 for DUS, RAG, SCA and GRE liverLEWW tissue, respectively). The preliminary comparison of C:N threshold corrected liver and muscle δ13C values corresponded with movement/habitat behaviours for each shark; minor differences in δ13C values were observed for known regional movements of DUS and RAG (δ13CDiffs = 0.24 ± 0.99‰ and 0.57 ± 0.38‰, respectively), while SCA and GRE showed greater differences (1.24 ± 0.63‰ and 1.08 ± 0.71‰, respectively) correlated to large-scale movements between temperate/tropical and pelagic/coastal environments. These data provide an approach for the successful application of liver δ13C and δ15N values to examine elasmobranch ecology.  相似文献

该文旨在研究氧化低密度脂蛋白(ox-LDL)对大鼠肝星状细胞(HSC-T6)自噬的影响及机制,探讨非酒精性脂肪肝炎的发病机理。体外培养的HSC-T6细胞以不同质量浓度(0、10、20、40、60μg/mL)的ox-LDL分别处理不同时间(0、3、6、12、24 h)后,用Western blot检测LC3 II、Beclin1、p62的含量。不同质量浓度(0、10、20、40、60μg/mL)的ox-LDL处理HSC-T6细胞12 h后,Western blot检测Wnt5a、p-PKCδ、p-STAT3的含量。将HSC-T6细胞分为对照组(Control)、ox-LDL组、ox-LDL+si-NC组和ox-LDL+si-Wnt5a组,经相应处理后用Western blot、qRT-PCR分别检测LC3 II、Beclin1、p62、p-PKCδ、p-STAT3的蛋白和mRNA含量变化;免疫荧光检测LC3 II的变化;油红O染色观察HSC-T6脂滴含量变化;比色法检测各组细胞培养上清中羟脯氨酸(Hyp)含量;ELISA检测细胞培养上清中透明质酸(HA)和层黏连蛋白(LN)含量。PKCδ抑制剂Rottlerin预处理细胞:将HSC-T6细胞分为对照组(Control)、ox-LDL组、ox-LDL+DMSO组和ox-LDL+Rottlerin组,检测方法与敲低Wnt5a一致。经ox-LDL处理后,HSC-T6细胞中LC3 II、Beclin1含量增加(P<0.05),p62含量减少(P<0.01),且在ox-LDL质量浓度为20μg/mL、作用12 h时达到峰值。ox-LDL质量浓度为20μg/mL、作用12 h时,HSC-T6细胞中Wnt5a、p-PKCδ、p-STAT3蛋白表达显著升高(P<0.01)。敲低Wnt5a后,HSC-T6细胞中Wnt5a、LC3 II、Beclin1 mRNA和蛋白表达水平显著降低(P<0.001),p62蛋白表达增多(P<0.01),p-PKCδ、p-STAT3蛋白表达减少(P<0.05),细胞内LC3 II点状聚集减少,脂滴含量减少,细胞培养上清中Hyp、HA、LN含量也减少(P<0.05)。抑制PKCδ后,结果与敲低Wnt5a一致。ox-LDL可通过增强Wnt5a/PKCδ通路诱导HSC-T6细胞自噬。  相似文献

Abstract

Diatom genome sequences clearly reveal the presence of different systems for HCO3? uptake. Carbon-concentrating mechanisms (CCM) based on HCO3? transport and a plastid-localized carbonic anhydrase (CA, EC 4.2.1.1) appear to be more probable than the others because CAs have been identified in the genome of many diatoms. CAs are key enzymes involved in the acquisition of inorganic carbon for photosynthesis in phytoplankton, as they catalyze efficiently the interconversion between carbon dioxide and bicarbonate. Five genetically distinct classes of CAs exist, α-, β-, γ-, δ- and ζ and all of them are metalloenzymes. Recently we investigated for the first time the catalytic activity and inhibition of the δ-class CA from the marine diatom Thalassiosira weissflogii, named TweCA. This enzyme is an efficient catalyst for the CO2 hydration and its inhibition profile with sulfonamide/sulfamate and anions have also been investigated. Here, we report the detailed biochemical characterization and chemico-physical properties of the δ-CA of T. weissflogii. The δ-CA encoding gene was cloned and expressed in Artic Express cells and the recombinant protein purified to homogeneity. Interesting to note that TweCA has no intrinsic esterase activity with 4-nitrophenyl acetate (pNpA) as substrate although the phylogenetic analysis showed that δ-CAs are closer to the α-CAs than to the other classes of such enzymes.  相似文献
