There are three different sodium transport systems (Ena1-4p, Nha1p, Nhx1p) in Saccharomyces cerevisiae. The effect of their absence on the tolerance to alkali-metal cations and on the membrane potential was studied. All three sodium transporters were found to participate in the maintenance of Na+, Li+, K+ and Cs+ homeostasis. Measurements of the distribution of a fluorescent potentiometric probe (diS-C3(3) assay) in cell suspensions showed that the lack of all three transporters depolarizes the plasma membrane. The overexpression of the Na+,K+/H+ antiporter Nha1 resulted in the hyperpolarization of the plasma membrane and consequently increased the sensitivity to Cs+, Tl+ and hygromycin B. This is the first evidence that the activity of a Na+,K+/H+ antiporter could play a role in the homeostatic regulation of the plasma membrane potential in yeast cells. 相似文献

AIMS: To test the suitability of the Agrobacterium tumefaciens-mediated transformation (AMT) method with Paecilomyces fumosoroseus, a fungal pathogen that causes diseases in a wide range of insects including whiteflies. METHODS AND RESULTS: Conidia of P. fumosoroseus were successfully transformed to hygromycin B resistance using the hph gene of Escherichia coli as the selectable marker. Transformation frequencies were 58.3 +/- 18.5, 98.3 +/- 24.8 and 169.7 +/- 35.5 (+/-SEM) transformants per 10(5), 10(6) and 10(7) target conidia respectively. After confirmation by PCR, transformants were subjected to Southern analysis, and the results revealed that 45% (four of nine) of the transformants contained single-copy integration of the T-DNA. CONCLUSIONS: In our AMT system, we efficiently transformed conidia of P. fumosoroseus. The employment of this method circumvents time-consuming protoplast preparation and allows the isolation of transformants containing single-copy integration of the T-DNA. SIGNIFICANCE AND IMPACT OF THE STUDY: Considering the efficiency of Ag. tumefaciens-mediated transformation, this method represents a useful tool for insertional mutagenesis to characterize genes that are important for the pathogenicity of P. fumosoroseus. 相似文献

The cell wall–lacking mutant CW-15 of the unicellular green alga Chlamydomonas reinhardtii was transformed by electroporation using plasmid pCTVHyg, which was constructed with the hygromycin phosphotransferase genehpt as the selective marker and the Tn5 transposon of Escherichia coli under the control of the virus SV40 early gene promoter. Under optimal conditions (106 mid-exponential cells/ml; electric field strength 1 kV/cm; and pulse length 2 ms), the transformation yielded 103 HygR transformants per 106 recipient cells. The exogenous DNA integrated into the nuclear genome of Ch. reinhardtii was persistently inherited through more than 350 cell generations. A pCTVHyg plasmid was constructed in a unicellular green alga Chlamydomonas reinhardtii Dang. by using the hygromycin phosphotransferase gene (hpt) as a selectable marker and the Escherichia coli transposon Tn5 under the early SV40 viral gene promoter. CW-15 mutant cells devoid of cell walls were transformed by electroporation in an electric field of 1 kV/cm and a pulse duration of 2 ms. A suspension density of 106 cell/ml and the mid-logarithmic growth phase were the optimum conditions for transformation, producing up to 103 hygromycin-resistant (HygR) clones per 106 HygR recipient cells. Exogenous DNA integrated in the nuclear genome of C. reinhardtii was steadily inherited in subsequent generations within at least a 8-month period; however, the HygR trait manifestation was not stable. The comparative analysis of frequencies in codon usage in hpt and in the nuclear genes of C. reinhardtii significantly excluded the possibility that the bias in codon usage was the primary factor affecting foreign gene expression. The advantages of using theCW-15 mutant and the described selection system are discussed in the context of heterologous transformation of C. reinhardtii. 相似文献

A simple and efficient protocol for the Agrobacterium-mediated transformation of an agronomically useful abiotic sensitive popular indica rice cv. ADT 43 has been developed. Initiation of calli were best achieved from the leaf bases of 4 days old rice seedlings on LS medium supplemented with 2.5 mg/L 2,4-D and 1.0 mg/L thiamine-HCl. Rice calli immersed in Agrobacterium suspension (strain EHA 105, OD600 = 0.8) were co-cultured on LS30-AsPC medium for 2 days at 25 ± 2 °C in the dark. Based on GUS expression analysis, 10 min co-cultivation time with 100 μM acetosyringone was found optimum for the delivery of gus gene. Calli were proved to be very sensitive to Agrobacterium infection and we found that the level of necrotic response can be minimized after co-cultivation with 30% LS, 10 g/L PVP, 10% coconut water and 250 mg/L timentin which improved the final transformation efficiency to 9.33%. Molecular and genetic analysis of transgenic plants reveals the integration, expression and inheritance of transgene in the progeny (T1) of these plants. Calcium reduces toxicity of aminoglycoside antibiotics in sugar beet explants in vitro 总被引:3,自引:0,他引:3
Aminoglycoside antibiotics are frequently used for the selection of transgenic plant cells. However, for a number of species aminoglycoside selection is inefficient. The objective of the present study was to elucidate factors affecting the phytoloxic effects of aminoglycoside antibiotics. Using non-transgenic sugar beet cotyledonary explants the interaction between three aminoglycoside antibiotics, kanamycin, neomycin and hygromycin. and Ca2+ was studied by monitoring the effects on growth and shoot formation. The phytotoxic effects of the aminoglycoside antibiotics were strongly dependent on the calcium concentration in the growth media. At comparable levels of the antibiotics (kanamycin 170 μ M , neomycin 220 μ M , hygromycin 9.5 μ M , an elevation of the calcium concentration from 1 to 10 m M resulted in growth increases of approximately 3-, 2.5- and 8-fold, respectively, and shoot formation was enhanced 1.5-, 2-and 6-fold, respectively. At lower concentrations of the antibiotics, the toxic effect was nearly abolished by increasing the calcium concentration. Additional magnesium, sodium and ammonium did not affect the phytotoxic effects of the aminoglycoside antibiotics. Moreover, the phytotoxic effects of the herbicides glyphosate and phosphinothricin were not decreased by additional calcium. These data suggest the existence of a specific interaction between calcium and aminoglycoside anfibiotics in plants. The implications of these results for the use of aminoglycosides as selective agents in plant transformation are discussed. 相似文献

Bt/CpTI转基因稻及其非转基因亲本对照在间隔种植条件下的转基因漂移 总被引:11,自引:1,他引:10
随着转基因技术的迅速发展,越来越多的转基因作物被培育出来。转基因作物的外源转基因通过花粉传播向非转基因作物的漂移,会影响非转基因作物品种的种子纯度,从而可能导致一系列生物安全问题。为了研究转基因栽培水稻(Oryzasativa)中的外源转基因通过花粉介导向非转基因水稻品种逃逸的可能性及其频率,我们选用3个含双价抗虫基因(Bt/CpTI)的转基因水稻品系及其相对应的非转基因水稻亲本品种(近等基因系)进行了转基因漂移的实验。为了获取在近距离状况下转基因水稻与非转基因水稻品种之间的转基因漂移频率,采用了转基因与非转基因水稻品种间隔种植的栽培方式,分别在福建省福州市和海南省三亚市的转基因环境安全实验地进行实验,并利用潮霉素抗性筛选标记基因来鉴定转基因和非转基因稻的杂种。共检测了从非转基因水稻品种随机收获的70,056颗种子,以此计算转基因漂移频率。结果表明,在相邻种植的情况下,由这3个转基因水稻向对应的非转基因水稻品种的转基因漂移的频率比较低(0.275–0.832%)。如此近距离条件下获得的低转基因漂移频率表明,对于严格自花授粉的水稻而言,通过一定的隔离措施,能有效地降低由花粉介导的转基因漂移导致的非转基因种子混杂。 相似文献
