Round wormwood (Artemisia sphaerocephala Krasch) seeds were germinated on Murashige & Skoog (1962) medium without plant growth regulators. The hypocotyls of seedlings were sliced and cultured on M1 medium with 2,4-dichlorophenoxyacetic acid (9.05 M) to induce callus. The induced calluses were subcultured on the same medium. Ten day old calluses were used to isolate protoplasts in an enzyme solution with 0.65 M mannitol. Protoplast yield strongly depended upon the state of callus cultures. Certain amount of hemicellulase could improve protoplast isolation. Purified protoplasts were cultured in modified Kao & Michayluk (1975) medium with 0.60 M mannitol as osmoticum, suggesting that protoplasts of A. sphaerocephala need a high initial osmolarity. Protoplasts generally divided evenly and the percentage of first division could reach 10%. Kinetin exhibited a positive effect on initial cell division. Furthermore, we studied the effect of protoplast density and vitamin C on sustained growth of protoplasts. The role of ethylene in anther culture of durum wheat ( Triticum durum Desf. cv. Ardente) was analyzed by testing the effects of 2-chloroethylphosphonic acid (ethrel) silver thiosulfate (Ag+), a -aminooxyacetic acid (AOA) and 1-aminocyclopropane-l-carboxylic acid (ACC) on microspore division observed after 21 days of culture and on development of calli estimated at day 45. The use of ethrel and Ag+ indicated a positive effect of ethylene on microspore division, whereas the use of AOA, and to a lesser extent ACC, snowed a negative effect. In contrast, the addition of ethrel or Ag+ indicated that ethylene inhibits the development of microspore-derived calli. AOA gave contradictory results. Ethylene production by anthers was about 7 pl anther−1h−1 and decreased during culture. ACC content in the anthers was maximal at day 9, whereas malonyl ACC (MACC) increased sharply from day 0 to day 3 and then decreased. The addition of AOA or ACC to the culture medium decreased or increased, respectively, ethylene production of anthers and the ACC and/or MACC content, but at concentrations higher than those that modified the formation of calli. This formation seems to occur in two successive phases: induction and initiation of microspore division, which was promoted by ethylene, followed by callus development, which was inhibited by ethylene.  相似文献

Acyl lipids and pigments were analyzed in young plants of garden pea, spring wheat and spinach exposed to < 5 or 65 nl l?1 ozone 12 h per day for 6 days. In one set of experiments, the plants were exposed to 14CO2 for 2 h 3 days prior to ozone exposure. The plants responded differently to the moderately enhanced level of ozone used Spinach was not at all sensitive while in both pea and wheat, leaves of different ages differed in ozone sensitivity. In pea, ozone sensitivity increased with leaf age. In the second and third oldest leaves, the amounts of galactolipids per leaf area and the proportions of 18:3 of the total lipid extract and of phosphatidylglycerol decreased. In the second oldest leaf, ozone also caused a decreased proportion of 18:3 of monogalactosyldiacylglycerol. In the fourth oldest leaf, lipid composition and galactolipid unsaturation was unaffected, but ozone caused decreased leaf expansion resulting in increased acyl lipid content per leaf area. In both the first and second leaves of wheat, ozone fumigation caused a marked decrease in the content of monogalactosyldiacylglycerol and in the first leaf, the contents of phosphatidylcholine and phosphatidylethanolamine increased. The proportion of 18:3 in phosphatidylcholine was larger in ozone-fumigated than in control plants, while the reverse applied for phosphatidylglycerol. In the oldest sampled leaves of pea and wheat, ozone caused an increase in the radioactivity associated with β-carotene, indicating increased turnover. Thus, while spinach was unaffected, in both pea and wheat ozone caused a decrease in the proportion of chloroplast membrane lipids to non-chloroplast membrane lipids in older leaves while younger leaves were less sensitive.  相似文献

A crown rot disease in wheat caused by the fungusFusarium graminearum Schw. Group 1 is a widespread problem in chronically Zn-deficient Australian soils. A link between crown rot and Zn deficiency was established by Sparrow and Graham (1988). This paper reports a test of a further hypothesis, that wheat genotypes more efficient at extracting zinc from low-zinc soils are more resistant to infection by this pathogen. Three wheat cultivars (Excalibur, Songlen and Durati) of differential Zn efficiency were tested at three zinc levels (0.05, 0.5 and 2.0 mg Zn kg−1 of soil) and three levels ofF. graminearum S. Group 1 inoculum (0.1 g and 0.3 g kg−1 live chaff-inoculum and control having 0.1 g kg−1 dead chaff inoculum). Six weeks after sowing dry matter production of shoots and roots was decreased byFusarium inoculation at 0.05 mg and 0.5 mg kg−1 applied Zn.Fusarium inoculum at 0.1 g was as effective as 0.3 g kg−1 for infection and decreasing dry matter. The infection at the basal part of culm decreased significantly by increasing the rate of Zn application. Excalibur, a Zn-efficient cultivar (tolerant to Zn deficiency) produced significantly more shoot and root dry matter, and showed less disease infection compared with Zn-inefficient cultivars (Durati and Songlen) at low (0.05 mg Zn kg−1 soil) and medium (0.5 mg Zn kg−1 soil) Zn fertilization rates. Higher rate of Zn fertilization (2.0 mg Zn kg−1 soil) reduced the disease level in Durati to the level of Excalibur but the disease level of Songlen was still high, indicating its high Zn requirement and or sensitivity to crown rot. The data on Zn uptake show that Excalibur, being Zn-efficient, was able to scavenge enough Zn from Zn-deficient soil, we suggest that besides sustaining growth Excalibur was able to build and maintain resistance to the pathogen; inefficient cultivars needed extra Zn fertilization to achieve performance comparable to that of Excalibur. The present study indicates that growing Zn-efficient cultivars of wheat along with judicious use of Zn fertilizer in Zn-deficient areas where crown rot is a problem may sustain wheat production by reducing the severity of the disease as well as by increasing the plant vigour through improved Zn nutrition. ei]Section editor: R Rodriques-Kalana  相似文献

We have investigated the water use efficiency of whole plants and selected leaves and allocation patterns of three wheat cultivars (Mexipak, Nesser and Katya) to explore how variation in these traits can contribute to the ability to grow in dry environments. The cultivars exhibited considerable differences in biomass allocation and water use efficiency. Cultivars with higher growth rates of roots and higher proportions of biomass in roots (Nesser and Katya) also had higher leaf growth rates, higher proportions of their biomass as leaves and higher leaf area ratios. These same cultivars had lower rates of transpiration per unit leaf area or unit root weight and higher biomass production per unit water use. They also had higher ratios of photosynthesis to transpiration, and lower ratios of intercellular to external CO2 partial pressure. The latter resulted from large differences in stomatal conductance associated with relatively small differences in rates of photosynthesis. There was little variation between cultivars in response to drought, and differences in allocation pattern and plant water use efficiency between cultivars as found under well-watered conditions persisted under dry conditions. At the end of the non-watered treatment, relative growth rates and transpiration rates decreased to similar values for all cultivars. High ratios of photosynthesis to transpiration, and accordingly high biomass production per unit of transpiration, is regarded as a favourable trait for dry environments, since more efficient use of water postpones the decrease in plant water status.  相似文献

A role for nitrogen reserves in forage regrowth and stress tolerance   总被引:20,自引:0,他引:20  
Carbohydrate accumulation and utilization during shoot regrowth after defoliation and winter has been studied extensively in most species used as forage. However, recent work suggests that N reserves found in vegetative tissues also are important for defoliation tolerance and winter hardiness. Results suggest that these N reserves constitute an alternative N source used when N2 fixation and/or mineral N uptake are reduced. 15N labelling experiments indicate that a large proportion of herbage N is derived from N reserves mobilized from stem bases or roots to developing leaves and shoots. Amino acids and specific proteins (i.e. vegetative storage proteins, VSPs) are deposited in roots and stem bases and, in the case of VSPs, are degraded rapidly after defoliation. Identification and characterization of VSPs will increase our understanding of the role N reserves play in stress tolerance and may lead to innovative approaches for improving forage persistence and productivity.  相似文献

Time courses of formation of inositol 1,4,5-trisphosphate (IP3) were followed in the leaves of non-acclimated and cold (2°C)-acclimated winter oilseed rape ( Brassica napus L. var. oleifera ) plants, subjected to different freezing temperatures or to polyethylene glycol 8000 (PEG) and abscisic acid (ABA) treatments. Changes in water potential (Ψw) and in ABA level in the frost- and PEG-treated tissues were also determined. Results obtained indicate that temperatures sligthly higher than LT50 induced a transient and substantial increase in IP3 level, both in non-acclimated and cold-acclimated tissues. At comparable freezing temperature (–5°C) the response of cold-acclimated leaves was lower than that of non-acclimated ones. The PEG-depedent decrease in Ψw to –0.9 MPa or ABA (0.1 m M ) treatment gave rise to a transient increase in IP3 content in non-acclimated tissues only. Collectively, the data indicate that cold acclimation of plants may lead to lower cell responsiveness to the factors studied in terms of induction of IP3 formation. Changes in the IP3 content, observed in the present experiments, support our previous suggestion that non-killing freezing temperatures may induce the phosphoinositide pathway, both in non-acclimated and cold-acclimated tissues. Lowering of tissue water potential to some threshold value or a high exogenous ABA supply may mimic the freezing-dependent reaction in the non-acclimated leaves.  相似文献

Many crop models relate the allocation of dry matter between shoots and roots exclusively to the crop development stage. Such models may not take into account the effects of changes in environment on allocation, unless the allocation parameters are altered. In this paper a crop model with a dynamic allocation parameter for dry matter between shoots and roots is described. The basis of the model is that a plant allocates dry matter such that its growth is maximized. Consequently, the demand and supply of carbon, nitrogen, and water is maintained in balance. This model supports the hypothesis that a functional equilibrium exists between shoots and roots.This paper explains the mathematical computation procedure of the crop model. Moreover, an analysis was made of the ability of a crop model to simulate plant dry matter production and allocation of dry matter between plant organs. The model was tested using data from a greenhouse experiment in which spring wheat (Triticum aestivum L.) was grown under different soil moisture and nitrogen (N) levels.Generally, the model simulations agreed well with data recorded for total plant dry matter. For validation data the coefficient of determination (r2) between simulated and measured shoot dry weight was 0.96. For the validation treatments r2 was slightly lower, 0.94. In addition to dry matter production the model succeeded satisfactorily in simulating the dry weight of different plant organs. The response of simulated root to shoot ratio to the level of soil moisture was mainly in accordance with the measured data. In contrast, the simulated ratio seemed to be insensitive to the changes in the levels soil N concentration used in the experiment.The data used in the present study were not extensive, and more data are needed to validate the model. However, the results showed that the model responses to the changes in soil N and water level were realistic and mostly agreed with the data. Thus, we suggest that the model and the method employed to allocate dry matter between roots and shoots are useful when modelling the growth of crops under N and water limited conditions.  相似文献

The fluxes of NO and NO2 between wheat canopy monoliths and the atmosphere were investigated with the dynamic chamber technique. For this purpose monoliths were dug out at different plant growth stages from a field site, transported to the institute, and placed in an environmental growth chamber. The wheat canopy monoliths were exposed over a period of four days to the average ratios of atmospheric NO2 and NO measured at the field site, i.e. NO2 concentration of about 18 mL L-1 plus NO concentration lower than 0.5 nL L-1. Under these conditions NO emission into the atmosphere and NO2 deposition into canopy monoliths was observed. Both fluxes showed diurnal variation with maximum rates during the light and minimum rates during darkness. NO2 fluxes correlated with soil temperature as well as with light intensity. NO fluxes correlated with soil temperature but not with light intensity. From the investigation performed the diurnal variation of the NO and NO2 compensation points, the maximum rates of NO and NO2 emission, and the total resistances of NO and NO2 fluxes were calculated. Under the assumption that the measured data are representative for the whole vegetation period, annual fluxes of NO and NO2 were estimated. Annual NO emission into the atmosphere amounted to 87 mg N m-2 y-1 (0.87 kg ha-1 y-1), annual NO2 deposition into canopy monoliths amounted to 1273 mg N m-2 y-1 (12.73 kg ha-1 y-1). Apparently, the uptake of atmospheric nitrogen by the wheat field from NO2 deposition is about 15 times higher than the loss of nitrogen from NO emission. It can therefore be assumed that even in rural areas wheat fields are a considerable sink for atmospheric nitrogen. The annual sink strength estimated in the present study is ca. 12 kg N ha-1 y-1. The possible origin of the NO emitted and the fate of atmospheric NO2 taken up by the wheat canopy monoliths are discussed.Preliminary results of this paper were presented at the Joint Workshop COST 611/Working Party 3 and EUROTRAC in Delft, The Netherlands (Ludwig et al., 1991).  相似文献
