1. 1.|In the freshwater fish Chalcalburnus chalcoides, an increase in the body (standard) size caused decreases in the upper LT-50 from 36.6° to 36.0°C and lower LT-50 from 6.3° to 5.3°C

2. 2.|The fish acclimated to constant temperatures between 10°C and 30°C showed reasonable heat acclimation and also reasonable cold acclimation. Thus, an increase in the acclimation temperature from 10°C to 30°C caused increases in the upper LT-50 from 34° to 36.2°C and the lower LT-50 from 1.25 to 6.5°C.

3. 3|The mean survival time — temperature curves of 10°, 20° and 30°C acclimated fish at various constant temperatures showed decreased in the survival tim ewith increasing lethal temperatures. Furthermore, an increase in the acclimation temperature causes a shift in the survival duration-temperature curve to the right, i.e., the fish become more heat resistant. Thus, the mean survival duration of 10°, 20° and 30°C acclimated fish at 35°C were 7.5, 79.6 and 530 minutes, respectively.

4. 4.|The effect of the thermal experience to changing lethal temperatures depends on the first lethal temperature to which the fish were exposed as well as the sequence of temperature changes. In the experiments in which the first lethal temperatures were between 32° and 34°C and the temperature was varied in an ascending order, their thermal resistance was increased and the fish required 114 to 174% of the expected lethal doses to die while in the experiments in which the starting temperature were between 38° and 40°C and the temperature varied in descending order, the fish become more sensitive to the upper lethal temperature and they died after receiving only 62 to 81% of the expected lethal doses. Thus, with a gradual increase in the lethal temperature, the fish show additional acclimation in the zone of resistance which in turn causes an increase in the thermal resistance. This may have ecological significance in nature.

The thermal stability of excitation transfer from pigment proteins to the Photosystem II reaction center of Nerium oleander adjusts by 10 Celsius degrees when cloned plants grown at 20°C/15°C, day/night growth temperatures are shifted to 45°C/32°C growth temperature or vice versa. Concomitant with this adjustment is a decrease in the fluidity of thylakoid membrane polar lipids as determined by spin labeling. The results are consistent with the hypothesis that there is a limiting maximum fluidity compatible with maintenance of native membrane structure and function. This limiting fluidity was about the same as for a number of other species which exhibit a range of thermal stabilities. Inversely correlated shifts in lipid fluidity and thermal stability occurred during the time course of acclimation of N. oleander to new growth temperatures. Thus, the temperature at which the limiting fluidity was reached changed during acclimation while the limiting fluidity remained constant. Although the relative proportion of the major classes of membrane polar lipids remained constant during adjustments in fluidity, large changes occured in the abundance of specific fatty acids. These changes were different for the phospho- and galacto-lipids suggesting that the fatty acid composition of these two lipid classes is regulated by different mechanisms. Comparisons between membrane lipid fluidity and fatty acid composition indicate that fluidity is not a simple linear function of fatty acid composition.  相似文献

Suspensions of yeast cell (zygo Saccharomyces bailii) in a phosphate buffer solution were subjected to conventional (hot water) and ohmic (electric current) heating under identical temperature histories. Experiments were also conducted with cells of Escherichia coli to compare the lethal effect of combination of sublethal electrical preteatment and conventional heating with conventional heating. The kinetic parameters (D,Z,K and E(a)) were determined for both organisms during different treatments. There was no significant difference in the death rate of yeast cells during conventional and ohmic heating at the voltage range used in this study. Results of electrical pretreatment and conventional heating on E. coli indicated differences under certain conditions when compared with pure conventional heating. Thus it is concluded that microbial death during ohmic heating was due primarily to thermal effects with no significant effect of electric current per se. Sublethal electrical pretreatment appears to offer potential for increased bacterial inactivation in certain cases.  相似文献

The obligate shade plant, Tradescantia albiflora Kunth grown at 50 mol photons · m–2 s–1 and Pisum sativum L. acclimated to two photon fluence rates, 50 and 300 mol · m–2 · s–1, were exposed to photoinhibitory light conditions of 1700 mol · m–2 · s–1 for 4 h at 22° C. Photosynthesis was assayed by measurement of CO2-saturated O2 evolution, and photosystem II (PSII) was assayed using modulated chlorophyll fluorescence and flash-yield determinations of functional reaction centres. Tradescantia was most sensitive to photoinhibition, while pea grown at 300 mol · m–2 · s–1 was most resistant, with pea grown at 50 mol · m–2 · s–1 showing an intermediate sensitivity. A very good correlation was found between the decrease of functional PSII reaction centres and both the inhibition of photosynthesis and PSII photochemistry. Photoinhibition caused a decline in the maximum quantum yield for PSII electron transport as determined by the product of photochemical quenching (qp) and the yield of open PSII reaction centres as given by the steady-state fluorescence ratio, FvFm, according to Genty et al. (1989, Biochim. Biophys. Acta 990, 81–92). The decrease in the quantum yield for PSII electron transport was fully accounted for by a decrease in FvFm, since qp at a given photon fluence rate was similar for photoinhibited and noninhibited plants. Under lightsaturating conditions, the quantum yield of PSII electron transport was similar in photoinhibited and noninhibited plants. The data give support for the view that photoinhibition of the reaction centres of PSII represents a stable, long-term, down-regulation of photochemistry, which occurs in plants under sustained high-light conditions, and replaces part of the regulation usually exerted by the transthylakoid pH gradient. Furthermore, by investigating the susceptibility of differently lightacclimated sun and shade species to photoinhibition in relation to qp, i.e. the fraction of open-to-closed PSII reaction centres, we also show that irrespective of light acclimation, plants become susceptible to photoinhibition when the majority of their PSII reaction centres are still open (i.e. primary quinone acceptor oxidized). The effects of root-zone salinity (0, 30, and 60 mmol L–1 of NaCl) and root-zone temperature (10, 15, 20, and 25°C) and their interactions on the number of tillers, total dry matter production, and the concentration of nutrients in the roots and tops of barley (Hordeum vulgare L.) were studied. Experiments were conducted in growth chambers (day/night photoperiod of 16/8 h and constant air temperature of 20°C) and under water-culture conditions. Salinity and root temperature affected all the parameters tested. Interactions between salinity and temperature were significant (p<0.05) for the number of tillers, growth of tops and roots, and the concentration of Na, K, P in the tops and the concentration of P in the roots. Maximum number of tillers and the highest dry matter were produced when the root temperature was at the intermediate levels of 15 to 20°C. Effect of salinity on most parameters tested strongly depended on the prevailing root temperature. For example, at root temperature of 10°C addition of 30 mmol L–1 NaCl to the nutrient solution stimulated the growth of barley roots; at root temperature of 25°C, however, the same NaCl concentration inhibited the root growth. At 60 mmol L–1, root and shoot growth were maximum when root temperature was kept at the intermediate level of 15°C; most inhibition of salinity occurred at both low (10°C) and high (25°C) root temperatures. As the root temperature was raised from 10 to 25°C, the concentration of Na generally decreased in the tops and increased in the roots. At a given Na concentration in the tops or in the roots, respective growth of tops or roots was much less inhibited if the roots were grown at 15–20°C. It is concluded that the tolerance of barley plant to NaCl salinity of the rooting media appears to be altered by the root temperature and is highest if the root temperature is kept at 15 to 20°C.  相似文献

Nucleosome cores were digested with α-chymotrypsin until histone H3 was degraded to a partial histone, CP1. As we reported previously, cleavage occurred at leucine 20 to H3 and resulted in an increase in circular dichroism between 265 to 285 nm. Some modest core unfolding was also observed as determined by a small decrease in the sedimentation coefficient. Studies reported here deal with the analysis of core secondary structure and subsequent perturbation caused by treatment with α-chymotrypsin. Raman spectroscopy indicated that chymotryptic treatment promoted a change in the conformational environment of a population of core histone tyrosines. In addition, a shift from B-form to an intermediate B- or A-form was observed for core DNA. High-resolution thermal denaturation was used to determine alterations in the stabilization of core DNA related to perturbation of the core histones. Brief chymotryptic treatment indicated changes in both pre-melt and irreversible transitions.  相似文献

Crystals of bacteriophage T4 lysozyme used for structural studies are routinely grown from concentrated phosphate solutions. It has been found that crystals in the same space group can also be grown from solutions containing 0.05 M imidazole chloride, 0.4 M sodium choride, and 30% polyethylene glycol 3500. These crystals, in addition, can also be equilibrated with a similar mother liquor in which the sodium chloride concentration is reduced to 0.025 M. The availability of these three crystal variants has permitted the structure of T4 lysozyme to be compared at low, medium, and high ionic strength. At the same time the X-ray structure of phage T4 lysozyme crystallized from phosphate solutions has been further refined against a new and improved X-ray diffraction data set. The structures of T4 lysozyme in the crystals grown with polyethylene glycol as a precipitant, regardless of the sodium chloride concentration, were very similar to the structure in crystals grown from concentrated phosphate solutions. The main differences are related to the formation of mixed disulfides between cysteine residues 54 and 97 and 2-mercaptoethanol, rather than to the differences in the salt concentration in the crystal mother liquor. Formation of the mixed disulfide at residue 54 resulted in the displacement of Arg-52 and the disruption of the salt bridge between this residue and Glu-62. Other than this change, no obvious alterations in existing salt bridges in T4 lysozyme were observed. Neither did the reduction in the ionic strength of the mother liquor result in the formation of new salt bridge interactions. These results are consistent with the ideas that a crystal structure determined at high salt concentrations is a good representation of the structure at lower ionic strengths, and that models of electrostatic interactions in proteins that are based on crystal structures determined at high salt concentrations are likely to be relevant at physiological ionic strengths.  相似文献

The LT50 (lethal temperature) of first instar and adult stages of the peach-potato aphid Myzus persicae was lowered following long term acclimation at low temperatures.
First instars consistently showed greater cold hardiness than adult stages at each acclimation temperature, with the differential increasing as the temperature was lowered. When maintained at 5°C (the lowest acclimation regime) nymphs and adults had dLT508.3°C and 4.7°C respectively lower than those for non-acclimated individuals.
When 10°C acclimated adults were returned to 20°C, the acclimation effect was retained in full for 6 days but complete deacclimation occurred by day 10. In contrast the LT50 of their progeny increased gradually from the first day of adult deacclimation towards the level of the unacclimated control over a period of 10 days.
A change in cold hardiness was observed in first instars according to their position in the birth sequence. The LT50 of first-born nymphs (day 1 of reproduction) from 20°C parents was - 15.9°C rising to - 8.3°C by day 4 and remaining at this level until the end of the reproductive period.
The differential mortality between nymphs and adults observed in the laboratory was supported by the results of a field experiment. Adult aphids kept in clip-cages on a crop of oilseed rape showed greater mortality compared with those introduced as nymphs when the minimum temperature fell below -4°C for the first time in winter. At - 10°C mortality of aphids introduced as adults approached 100% whereas more than 50% of those introduced as nymphs were still alive at this temperature.  相似文献
