GABAergic cortical interneurons, derived from the embryonic medial and caudal ganglionic eminences (MGE and CGE), are functionally and morphologically diverse. Inroads have been made in understanding the roles of distinct cortical interneuron subgroups, however, there are still many mechanisms to be worked out that may contribute to the development and maturation of different types of GABAergic cells. Moreover, altered GABAergic signaling may contribute to phenotypes of autism, schizophrenia and epilepsy. Specific Cre-driver lines have begun to parcel out the functions of unique interneuron subgroups. Despite the advances in mouse models, it is often difficult to efficiently study GABAergic cortical interneuron progenitors with molecular approaches in vivo. One important technique used to study the cell autonomous programming of these cells is transplantation of MGE cells into host cortices. These transplanted cells migrate extensively, differentiate, and functionally integrate. In addition, MGE cells can be efficiently transduced with lentivirus immediately prior to transplantation, allowing for a multitude of molecular approaches. Here we detail a protocol to efficiently transduce MGE cells before transplantation for in vivo analysis, using available Cre-driver lines and Cre-dependent expression vectors. This approach is advantageous because it combines precise genetic manipulation with the ability of these cells to disperse after transplantation, permitting greater cell-type specific resolution in vivo.  相似文献

Summary The aim of this work was to investigate the effect of a short-term exposure to somatostatin (SS), its receptors (SSTR) selective agonists as well as muscarinic receptors agonists upon acetylcholine-induced release of 3H-MPP+ from bovine adrenal medullary cells. Acetylcholine (ACH, 100, 500 μM) was found to increase the release of 3H-MPP+ by these cells (to 175 and 171% of basal release, respectively). ACH-elicited 3H-MPP+ release was significantly reduced by hexamethonium (100 μM) and atropine (100 μM), selective nicotinic and muscarinic antagonists, respectively. Previous exposure to any of two muscarinic agonists, oxotremorine or pilocarpine, led to a significant reduction of 3H-MPP+ release in response to 100 μM ACH, to about a maximum of 51% and 78% of control, respectively. Somatostatin (SS, 0.01–0.1 μM), previously applied to the preparation, depressed ACH-elicited 3H-MPP+ release by 25–27%, but only when a 500 μM ACH concentration was used. The inhibition exerted by SS upon ACH-evoked 3H-MPP+ release appeared to be mediated by its SSTR: (1) SSTR2, 3 and 4 subtype agonists mimicked the effects seen with SS, and (2) the SSTR non-selective antagonist, cyclo-SS, counteracted the SS inhibitory effect. When SS was tested in the presence of any of the muscarinic agonists, oxotremorine or pilocarpine, its inhibitory effect on 500 μM ACH-induced 3H-MPP+ release was no longer detectable. These results, showing a somewhat similar effect of short-term exposure to SS and muscarinic agonists over ACH-induced release of 3H-MPP+, as well as the loss of effect of SS by the presence of the muscarinic agonists, suggest that these compounds may share signalling pathways.  相似文献

TT-232 is a structural derivative of the peptide hormone somatostatin with selective anti-proliferative and anti-inflammatory properties. It has a strong anti-tumour activity both in vitro and in vivo on a wide range of tumour models and induces apoptosis. Its anti-tumour activity is mediated through the SSTR1 receptor and by the tumour specific isoform of pyruvate kinase. TT-232 has been shown to be a potent neurogenic inflammation inhibitory, anti-inflammatory and analgesic agent with a broader spectrum than presently available anti-inflammatory/analgesic drugs. In animal models it is effective against neurogenic inflammation and blocks neuropathic hyperalgesia where COX-1 or COX-2 inhibitors (e.g. diclofenac or meloxicam) proved ineffective. TT-232 has passed phase I clinical trials without toxicity and significant side effects. Human phase II efficacy studies are ongoing in melanoma indication. Two more oncological indications and phase II clinical trials in inflammatory diseases, including rheumatoid arthritis and burn injuries are in preparation. This compound has the perspective to become the first drug in molecularly targeted therapy of inflammation where a combined effect of anti-inflammatory, analgesic and neurogenic inflammation inhibiting activity can be achieved.  相似文献

生长抑素(somatostatin,SST)通过与细胞膜上的G蛋白偶联的生长抑素受体(somatostatin receptors,SSTRs)结合而发挥其抑制细胞增殖的作用,因而生长抑素类似物(somatostatin analogue, SSA)常被用于肿瘤辅助治疗。然而,治疗效果存在相当大的个体差异,推测生长抑素类似物治疗效果不佳,与内源性生长抑素受体表达缺失或者表达量和亚型组合有关。为此,检测各亚型SSTR在几例罕见的神经内分泌肿瘤中的表达,并检测过表达SSTR2和SSTR5以及受体激活对细胞增殖的抑制效果,分析受体激活的可能机制,有助于临床筛选适合SSA肿瘤辅助治疗的病例,预估SSA的治疗效果。免疫组化检测肿瘤组织SSTR1-5的表达。在培养的293T细胞中过表达SSTR2和SSTR5,免疫共沉淀检测受体相互作用,免疫荧光和共聚焦显微镜检测受体细胞内定位。用MTT法检测受体过表达及激活对培养的人肺癌细胞NCI-H460细胞增殖的影响,用流式细胞技术检测细胞周期分布。SSTR1-5在10例神经内分泌肿瘤组织中均有不同程度的表达,表达亚型及表达量与肿瘤类型和年龄无关,SSTR5在所有肿瘤组织中均表达。SSTR2与SSTR5可形成受体相互作用。SSTR2与SSTR5活化后相互作用增加并定位于细胞质。共表达SSTR2和SSTR5显著抑制细胞增殖,并与受体激活剂呈现剂量相关性。SSTR2/SSTR5的共表达及激活显著减少S期的细胞而滞留于G1期。  相似文献

The binding of somatostatin was studied in the cytosolic fraction of bovine gallbladder mucosa. The binding reaction depended on time, temperature and pH, and was reversible, saturable and specific. Stoichiometric data suggested the presence of two classes of binding sites: a class with high affinity (K d=23.6 nM) and low capacity (3.7 pmol somatostatin/mg protein) and a class with low affinity (K d=284.6 nM) and high capacity (85.0 pmol somatostatin/mg protein) at 37°C and pH 7.4. The binding sites were highly specific for somatostatin since peptides such as [Leu]enkephalin, neurotensin, vasoactive intestinal peptide and substance P showed practically no effect upon somatostatin binding. The presence of somatostatin-binding sites in the cytosolic fraction of gallbladder mucosa, together with the known occurrence of somatostatin nerve endings in the gallbladder strongly suggests that this peptide may be involved in the physiology and physiopathology of gallbladder mucosa.  相似文献

本工作利用血管灌流离体大鼠胃研究促胰液素和生长抑素对泌酸的影响及其与内源性前列腺素E(PGE)和前列环素(PGI_2)释放的关系。结果表明:(1)促胰液素和生长抑素都能有效地抑制五肽胃泌素(G_5)促进胃酸分泌的作用,消炎病可翻转这种抑制作用。(2)促胰液素能显著促进PGE和PGI_2代谢产物6-酮-前列腺素F_(1α)(6-Keto-PGF_(1α))释放;生长抑素只能促进FGE释放。消炎痛分别阻断促胰液素对PGE和6-keto-PGF_(1α)释放及生长抑素对PGE释放的促进作用。上述结果提示:(1)促胰液素的抑酸效应由促进PGI_2和PGE释放介导;(2)生长抑素的抑酸效应通过促进PGE释放介导。  相似文献

1. The goal of this work was to determine the effects of typical and atypical neuroleptics on the level of preprosomatostatin messenger RNA (mRNA) in regions of the rat brain innervated by dopaminergic neurons. 2. Quantitative in situ hybridization histochemistry was used to measure the levels of mRNA encoding preprosomatostatin in neurons of the striatum, the nucleus accumbens, and the medial and lateral agranular areas of the frontal cortex in adult rats treated with either haloperidol or clozapine. 3. In untreated animals, the density of neurons containing preprosomatostatin mRNA was higher in the nucleus accumbens than in the striatum and frontal cortex. The intensity of labeling per neuron, however, was higher in the striatum than in the two other areas examined, suggesting that the expression of preprosomatostatin mRNA is differentially regulated in these brain regions. Chronic administration of haloperidol (1 mg/kg for 28 days) induced a significant decrease in the labeling for preprosomatostatin mRNA in neurons of the nucleus accumbens, frontal cortex, and medial but not lateral striatum. Treatment with clozapine (20 mg/kg for 28 days) increased the levels of preprosomatostatin mRNA in the nucleus accumbens but not in the striatum or the frontal cortex. 4. These results support a role for dopamine in the regulation of central somatostatinergic neurons. The differences in the effects of haloperidol, a neuroleptic which induces extrapyramidal side effects, and clozapine, which does not, suggest that somatostatinergic neurons may play an important role in the regulation of motor behavior.  相似文献

We report the conformational analysis by 1H‐NMR in DMSO and computer simulations involving distance geometry and molecular dynamics simulations of peptoid analogs of the cyclic hexapeptide c‐[Phe11‐Pro6‐Phe7‐d ‐Trp8‐Lys9‐Thr10] L‐363,301 (the numbering refers to the positions in native somatostatin). The compounds c‐[Phe11‐Nphe6‐Nal7‐d ‐Trp8‐Lys9‐Thr10] ( Nphe 6 ‐ Nal 7 analog 1 ), c‐[Nal11‐Nphe6‐Phe7‐d ‐Trp8‐Lys9‐Thr10] ( Nal 11 ‐ Nphe 6 analog 2 ) and c‐[Phe11‐Nnal6‐Phe7‐d ‐Trp8‐Lys9‐Thr10] ( Nnal 6 analog 3 ), where Nphe denotes N‐benzylglycine and Nnal denotes N‐(1‐naphthylmethyl)glycine, are subjected to SAR studies in order to investigate the influence of the bulky naphthyl aromatic ring on the conformation. The Nal 11 ‐ Nphe 6 and Nphe 6 ‐Nal 7 analogs exhibit potent binding to the hsst2, hsst3 and hsst5 receptors, whereas the Nnal 6 analog has decreased binding affinity to all receptors but is more selective towards the hsst2 than the other two analogs and L‐363,301. The conformational search employing distance geometry, energy minimization and molecular dynamic simulations gives insight into the conformational flexibility of these analogs. The molecules adopt both cis and trans orientations of the peptide bond between residues 11 and 6. The cis isomers of these analogs adopt type II′ β‐turns with d ‐Trp in the i+1 position and type VIa β‐turns with the cis peptide bond between residues 6 and 11. The results of free and distance restrained molecular dynamics simulations at 300 K indicate that the Nphe 6 ‐Nal 7 and Nal 11 ‐Nphe 6 compounds adopt a preferred backbone conformation which can be described as ‘folded’ about residues 7 and 10. The Nnal 6 analog, which binds less effectively to the hsst receptors, has a more flexible backbone structure than the Nal 11 ‐Nphe 6 and Nphe 6 ‐Nal 7 analogs and prefers a ‘flat’ structure with regard to the orientations about Phe7 and Thr10 during molecular dynamics simulations.
