A continuous nutrient flow system has been developed to measure microbial activity in soil with various concentrations of
added substrate. The system consists of a thin soil layer through which substrate was added continuously over periods up to
4.5 days. Substrate utilization was determined by effluent analysis. Respiration was measured manually by injecting a sample
into a gas chromatograph or automatically by coupling the growth chamber to a computer-controlled gas sampling valve. This
permitted respiratory CO2 to be measured by the gas chromatograph at intervals selected by the investigator. Software controlling the valve and gas
chromatograph not only automated gas phase sampling, but also provided a scan of CO2 evolution and a preliminary data summary. This included the date and time of sample, peak height, and percent CO2 in the gas phase. Data for growth on glucose using a microbial population native to a California annual grassland soil demonstrated
that the direct cell count and respiratory techniques for biomass estimation give comparable results. This procedure provides
the potential for detailed analyses of substrate utilization in studies of the growth and maintenance of soil microorganisms. 相似文献

Summary Cucumber seedlings were grown in a Portsmouth soil-sand system to study how varying soil clay and organic matter content might
modify cucumber seedling response to ferulic acid, a reported allelopathic agent. Leaf area expansion of cucumber seedlings,
soil respiration, and soil solution concentrations of ferulic acid were monitored. Leaf area, mean absolute rates of leaf
expansion, and shoot dry weight of cucumber seedlings were significantly reduced by ferulic acid concentrations ranging from
10 to 70 μg/g dry soil. Ferulic acid was applied every other day, since it rapidly disappeared from soil solution as a result
of retention by soil particles, utilization by microbes and/or uptake by roots. The amount of ferulic acid retained (i.e., adsorbed, polymerized,etc.) by soil particles appeared to be secondary to microbial utilization and/or uptake by roots. Varying clay (5.3 to 9.8 g/cup)
and organic matter (2.0 to 0.04g/cup) contents of the soil appeared to have little impact on the disappearance of ferulic
acid from soil solution under “ideal” growth conditions for cucumber seedlings unless larger amounts of ferulic acid were
added to the soil; in this case 200 μg/g. The addition of ferulic acid to the soil materials substantially increased the activity
of the soil microbes. This latter conclusion is based on recovery of ferulic acid from soil solution and soil respiration
measurements.

Five plant species were cultivated on a soil from the Neckar alluvial fan near Heidelberg (FRG) polluted by the emissions
of a cement plant. Thallium, cadmium and lead concentrations in seedlings and mature plants were determined by atomic absorption
analysis. AdditionallyBrassica napus L.napus was grown on soils containing 5 different concentrations of heavy metals, achieved by mixing two similar soils, from the
same area but with different metal concentrations.
Thallium and cadmium were shown to be taken up by roots whilst lead which was also absorbed, was deposited mainly on the plant
surface. However during cultivation in the winter months, a remarkable deposit of lead via the roots was found. Thallium in
the soil from a anthrorogen source was more available to plants than thallium of geological origin.
During the lifetime of a plant concentrations of thallium and cadmium were always highest in the seedling. The decrease in
metal concentration with maturity depended on the plant species and the element, but was not a function of the metal concentration

Binding of gossypol by gossypin and congossypin and their succinylated and sulfhydryl group-blocked derivatives has been measured.
The binding by gossypin and congossypin is characterized by weak interaction. Succinylation of gossypin decreases the binding
affinity whereas that of congossypin increases it. Blocking of sulfhydryl groups of both the proteins does not significantly
affect gossypol binding, Succinylation dissociates gossypin and causes conformational changes whereas it does not dissociate
congossypin but causes conformational changes. Sulfhydryl group blocking does not dissociate gossypin or congossypin, nor

The galactose-binding lectin of human Placenta has been Purified to homogeneity by affinity chromatograPhy on asialo-fetuin
column. The Protein, extractable from the tissue only with lactose is aPParently membrane-bound. Molecular weight determination
of native Protein and subunit indicated a dimer of l3.4 kDa subunits. Inhibition of haemagglutination with various saccharides
indicate that thiodigalactoside is the best inhibitor followed by lactose. However,P-nitroPhenyl-and 1-O-methyl derivatives of galactose showed that α-anomers inhibited slightly better than β-anomer. Modification
of amino acid residues indicated involvement of arginine, lysine and histidine residues at the saccharidebinding site. Cysteine

Short-term species succession, and long-term community development, of planktonic rotifer populations of the temperate zone and factors influencing species' abundance (ie., physical and chemical limitations, food and exploitative competition, mechanical interference competition, predation, parasitism) are described and discussed. The long-term development of plankton communities in three European lakes is described and the major events are discussed in relation to ecological interactions. Frequently, the shortcomings of our knowledge about population ecology and ecophysiology of rotifers prevent explanations of short-term and, especially, of long-term developments of these plankton organisms. Summary A simple method for the evolutionary analysis of amino acid sequence data is presented and used to examine whether the number of variable sites (NVS) of a protein is constant during its evolution. The NVSs for hemoglobin and for mitochondrial cytochrome c are each found to be almost constant, and the ratio between the NVSs is close to the ratio between the unit evolutionary periods. This indicates that the substitution rate per variable site is almost uniform for these proteins, as the neutral theory claims. An advantage of the present analysis is that it can be done without knowledge of paleontological divergence times and can be extended to bacterial proteins such as bacterial c-type cytochromes. It is suggested that the NVS of cytochrome c has been almost constant even over the long period (ca. 3.0 billion years) of bacterial evolution but that at least two different substitution rates are necessary to describe the accumulated changes in the sequence. This two clock interpretation is consistent with fossil evidence for the appearance times of photosynthetic bacteria and eukaryotes. 相似文献

Experiments were conducted for 3 years at four locations and 1 year with six soil types at a common location in North Carolina to determine damage and control-cost functions for Heterodera glycines races 1 and 2 on soybean. In the experiments on native loamy sand and sandy soils, tolerance limits for initial population densities were 0 or very low, whereas in a muck, the tolerance limit was 315 eggs/500 cm³ soil. The aggressive race 2 was more damaging than race 1 in Lakeland sand and Norfolk loamy sand. The crop response was not different between races in the Appling sandy clay loam and Belhaven muck. Soybean yield responses to H. glycines were linear in six soil types in microplots at a common site. The amount of damage varied among these soil types, with lowest yields in the muck because of severe drought stress in this soil. An exponential function adequately described soybean yield response relative to nematode control with increasing rates of aldicarb in Norfolk loamy sand. Treatment with aldicarb in the Lakeland sand decreased the effective egg population of H. glycines but had only a minor effect in the muck. 相似文献

The effects of various insecticides on the mycelial growth, sporulation and conidial germination of Metarhizium anisopliae var. anisopliae isolate E9 were studied in the laboratory. Chlorpyrifos was the most toxic organophosphate to mycelial growth and sporulation at all concentrations. Temephos, malathion and leptophos were highly toxic to sporulation while malathion was the most inhibitory to germination. The carbamates, carbofuran, methomyl and oxamyl were moderately toxic to mycelial growth and sporulation while oxamyl had an adverse effect on germination. The pyrethroids (pyrethrin, permethrin and resmethrin) and the insect growth regulators (diflubenzuron and methoprene) were not inhibitory to the various developmental stages of isolate E9. The chlorinated hydrocarbons (chlordane, lindane and toxaphene) were more deleterious than all other insecticide groups tested.
