蕲蛇酶对大白鼠和家兔实验性脑血栓的溶栓作用   总被引:3,自引:1,他引:2

采用血小板聚集诱导剂二磷酸腺苷(ADP)加肾上腺素(Ad)从家兔颈内动脉或大白鼠颈总动脉注入,以形成脑血栓,2h后各组动物分别从静脉给予不同剂量蕲蛇酶(150、300和600mg/kg),阳性对照药潘生丁(10mg/kg)及空白对照生理盐水(1ml/kg)。以脑组织对伊文思蓝通透性改变及脑组织切片计算血栓数目为指标,判断蕲蛇酶对血栓的影响。结果不同剂量蕲蛇酶组对大鼠或家兔的实验性脑血栓均比生理盐水组血栓消褪加快,形成的血栓数目减少,脑组织伊文思蓝含量亦明显减少;潘生丁组亦显示促血栓消褪的效果  相似文献

The haemorrhagin AaH III isolated from the snake venom ofAgkistrodon acutus is one of the few alkaline ones in snake venoms. Its crystals belong to space group P212121 witha = 9. 573 4 nm,b = 4. 996 7 nm andc = 4.728 8 nm. Its crystal structure was determined by the molecular replacement method according to the model of metalloproteinase Adamalysin II from eastern rattlesnake venom. The AaH III structure has been refined by PROLSQ. The finalR factor was 0.254 and the RMS deviations of bond lengths and angles were 0.001 8 nm and 1.5°. The structure comparison suggested that AaH III has a similar structure to other snake venom zinc-metalloproteinases. They all belong to matrix metalloproteinases super-family. The experiments were perfomed on transvcrsus abdominis muscle of Elaphe dione by subendothelial recording. The results indicate that in snake motor nerve endings there exist four types of K* channels, i.e. voltage-dependent fast and slow K channels, Ca2 -activated K channel and ATP-sensitive K channel, (i) The typical wave form of snake terminal current was the double-peaked negativity in standard solution. The first peak was at-tributed to Na influx (INa) in nodes of Ranvier. The second one was blocked by 3, 4-aminopyridine (3, 4-DAP) or te-traethylammonium (TEA), which corresponded to fast K outward current (IKF) through the fast K* channels in terminal part, (ii) After IKF as well as the slow K current (IKS) were blocked by 3, 4-DAP, the TEA-sensitive Ca2 -dependent K current (IK(Ca)) passing through Ca2 -activated K channel was revealed, whose amplitude depended on [K ]and [Ca2 ] It was blocked by Ba2 , Cd2 or Co2 . SYNOPSIS. Monocercomonas shares many fine-structural features with all other trichomonads. These include the basic arrangement of the kinetosomes as well as of the recurrent and 3 anterior flagella. The pelta-axostyle complex and the parabasal apparatus, i.e. the Golgi complex and the periodic filaments, also conform to the trichomonad pattern. Of interest with regard to the crucial evolutionary position of Monocercomonas, considered to represent the most primitive trichomonad type, is the fact that it has some structures in common with other Monocercomonadidae and Trichomonadinae and others in common with Devescovinidae and Tritrichomonadinae. Among the former organelles are the marginal lamella and the costal base, and among the latter, the comb-like organelle situated between the infrakinetosomal body and parabasal filament 2 as well as the infrakinetosomal body. No traces of either costa or undulating membrane have been noted, but a complex structure homologous to the marginal lamella of Hypotrichomonas and Trichomonadinae is found underlying the short anteriormost portion of the recurrent flagellum that is attached to the body surface. Observations of sections of selected division stages indicate the potential of parental kinetosomes #1 and #3 to become daughter kinetosome #2.  相似文献

Proton nuclear magnetic resonance (NMR) spectra have been recorded of various neurotoxins from snake venoms.pH dependence of the chemical shifts and resonance intensity has been followed for the functionally essential Trp-29. The indole N-1 proton of Trp-29 in -bungarotoxin, toxin B, and cobrotoxin exhibits appreciably large upfield shifts as thepH is lowered and the suppressed exchange with the solvent hydrogen atpH 3–4, but not inNaja haje annulifera 10 where Asp-31 is replaced with Gly-31. This observation strongly suggests the presence of a hydrogen bond between Trp-29 and Asp-31 that is probably important in stabilizing the arrangement of the functionally essential residues to form a distinct binding region for the receptor.  相似文献

A radioiodinated derivative of OS2 (125I–OS2), a neurotoxic monochain phospholipase A2 isolated from taipan venom, was previously found to bind to a specific brain membrane receptor with very high affinity.125I–OS2 is now used to identify the properties of neurotoxic phospholipase receptors in other tissues. Heart, skeletal muscle, kidney, lung, liver, pancreas, and smooth muscle membranes also contain high-affinity binding sites for toxic phospholipases A2. In most tissues, two different types of receptor sites have been characterized for125I–OS2 with Kd1 and Kd2 values in the 1–5 pM and the 10–50 pM range respectively. Whereas all receptors are similar in the different tissues in terms of their affinity for125I–OS2, maximal binding site capacities were very different, varying from 1.4 pmol/mg of protein in brain to 0.01 pmol/mg of protein in pancreaas. In brain, heart, and skeletal muscle, receptor densities vary with in vivo development. Affinity labeling experiments have identified the subunit composition of OS2 receptors and indicated that these receptors do not have identical structures in the different tissues. Binding competition studies with OS2 and other toxic phospholipases showed tissue-dependent pharmacological profiles. Early environments can profoundly influence an organism in ways that persist over its life. In reptiles, early thermal environments (nest temperatures) can impact offspring phenotype and survival in important ways, yet we still lack an understanding of whether general trends exist and the magnitude of impact. Understanding these patterns is important in predicting how climate change will affect reptile populations and the role of phenotypic plasticity in buffering populations. We compiled data from 175 reptile studies to examine, and quantify, the effect of incubation temperature on phenotype and survival. Using meta‐analytic approaches (standardized mean difference between incubation treatments, Hedges' g), we show that across all trait types examined there is, on average, a moderate to large magnitude of effect of incubation temperatures (absolute effect: |g| = 0.75). Unsurprisingly, this influence was extremely large for incubation duration, as predicted, with warmer temperatures decreasing incubation time overall (g = −8.42). Other trait types, including behaviour, physiology, morphology, performance, and survival experienced reduced, but still mostly moderate to large effects, with particularly strong effects on survival. Moreover, the impact of incubation temperature persisted at least one‐year post‐hatching, suggesting that these effects have the potential to impact fitness in the long term. The magnitude of effect increased as the change in temperature increased (e.g. 6°C versus 2°C) in almost all cases, and tended to decrease when temperatures of the treatments fluctuated around a mean temperature compared to when they were constant. The effect also depended on the mid‐temperature of the comparison, but not in consistent ways, with some traits experiencing the greatest effects at extreme temperatures, while others did not. The highly heterogeneous nature of the effects we observe, along with a large amount of unexplained variability, indicates that the shape of reaction norms between phenotype and temperature, along with ecological and/or experimental factors, are important when considering general patterns. Our analyses provide new insights into the effects of incubation environments on reptile phenotype and survival and allow general, albeit coarse, predictions for taxa experiencing warming nest temperatures under climatic change.  相似文献
