The efficacy of eight isolates of Pichia angusta against three common postharvest pathogens of apple fruit was evaluated for the first time. All tested strains showed significant biocontrol activity against both Botrytis cinerea and Monilia fructicola , whereas efficacy against Penicillium expansum was poor. A leucine-auxotrophic mutant had no significant biocontrol activity against brown rot of apple, while the addition of 0.6–1.2 g L−1 leucine in the fruit wound fully restored the biocontrol activity of this mutant against M. fructicola . Given the extremely well-developed classical and molecular genetics, the availability of genomic libraries, and its complete genomic sequence, this species can serve to elucidate the mechanisms related to biocontrol capacity.  相似文献

The immobilization of an endoglucanase, benzoylformate decarboxylase (BFD) from Pseudomonas putida, as well as of lipase B from Candida antarctica (CALB) onto the carrier supports Sepabeads EC-EP, Sepabeads EC-EA, and Sepabeads EC-BU was accomplished. It is shown that via these immobilized biocatalysts the synthesis of both fine and bulk chemicals is possible. This is illustrated by the syntheses of polyglycerol esters and (S)-hydroxy phenyl propanone. The benefit of immobilization is illustrated by repetitive use in a bubble column reactor as well as in a stirred tank reactor. High stability of two biocatalysts was achieved and reusability up to eight times was demonstrated. The comparison of CALB immobilized on Sepabeads EC-EP to Novozym 435 shows similar activity. A total of 123 Alternaria isolates were obtained from roting fruits of Pyrus bretschneideri "Ya Li" collected in Hebei Province, China. The isolates, according to morphological characteristics of conidia and sporulation patterns, were segregated into four groups: A. alternata group, A. tenuissima group, A. yaliinficiens group and Alternaria sp. group. Molecular characteristics of part of the isolates were determined using random amplified polymorphism DNA (RAPD) analysis. Based on cluster analysis of RAPD data, large-and small-spored Alternaria spp. were evidently distinguished, and the small-spored species can form five clusters that fundamentally paralleled the morphological groupings.  相似文献

Zuili plum (Prunus salicina L.) trees usually set fruit poorly, although they produce high quality fruit. To elucidate the causes of the poor fruit set, pollen tube growth into pistils and fruit set percentage were investigated after cross-, self- and open-pollination. Ovule development in Zuili pistils was also investigated. Pollen tube penetration into the ovules via the obturator and micropyle was best when Zuili pistils were pollinated by cv. Black Amber (P. domestica) pollen grains, although cross-pollinations with Hongxinli and Miili (P. salicina) pollen were more effective than self- and open-pollination. The fruit set percentage was also highest in pistils pollinated with Black Amber pollen grains. Morphological observation of Zuili pistils revealed that the trees produce "double pistils", developing two ovaries from a basal pistil, at a rate as high as 28%. In such abnormal pistils, most ovules were lacking an embryo sac or were entirely degenerated. The percentage of normally developed ovules was 24.3% and 8.9% in normal and double pistils, respectively. From these results, we conclude that the main causes of poor fruit set of Zuili plums are a lack of effective cross-pollination and the production of high percentages of double pistils in which normally developed ovules are scarcely formed.  相似文献

利用透射电子显微镜技术研究了北柴胡营养器官中分泌道的发育以及挥发油的积累.并重点探讨了挥发油的分泌机制.结果显示.分泌道细胞的质体、细胞基质以及线粒体参与挥发油或其前体物质的合成,而内质网参与挥发油向分泌道腔隙的转运.在分泌道发育的后期,大量小泡与分泌细胞相邻壁的质膜融合,并将其内的物质释放入此部分细胞壁中,与此同时,此部分壁在靠近腔隙的位置结构变得松散.然后围绕腔隙的壁在相邻两分泌细胞相接的位置面向腔隙外突形成许多小泡,并将小泡释放入腔隙中.由此可见.北柴胡分泌道中挥发油主要以胞吐方式被排入分泌道的腔隙中.  相似文献

The first set of 15 expressed sequence tag-simple sequence repeat markers was developed in sea urchin Hemicentrotus pulcherrimus. Number of alleles per locus ranged from two to 17. The observed and expected heterozygosities ranged from 0.022 to 0.911 and from 0.022 to 0.916, respectively. These informative marker loci will be useful for the assessment of genetic variation and population structure of this species.  相似文献

The white sea bream, Diplodus sargus (Teleostei, Sparidae), is a species with a high commercial importance in Mediterranean aquaculture. There is currently little information available about the genetic characteristics of cultured populations. In this survey, we have developed eight polymorphic microsatellites for the white sea bream using an enriched genome library protocol. All of them were polymorphic in the 67 individuals tested, 32 of which were wild specimens, and 35 were individuals from a captive F(1) broodstock. These markers can potentially be useful tools for use in population genetic studies.  相似文献

For experiments at the torque meter, flies are kept on standard fly medium at 25°C and 60% humidity with a 12hr light/12hr dark regime. A standardized breeding regime assures proper larval density and age-matched cohorts. Cold-anesthetized flies are glued with head and thorax to a triangle-shaped hook the day before the experiment. Attached to the torque meter via a clamp, the fly''s intended flight maneuvers are measured as the angular momentum around its vertical body axis. The fly is placed in the center of a cylindrical panorama to accomplish stationary flight. An analog to digital converter card feeds the yaw torque signal into a computer which stores the trace for later analysis. The computer also controls a variety of stimuli which can be brought under the fly''s control by closing the feedback loop between these stimuli and the yaw torque trace. Punishment is achieved by applying heat from an adjustable infrared laser.  相似文献

Biodiesel has become more attractive recently because of its environmental benefits, and the fact that it is made from renewable resources. Generally speaking, biodiesel is prepared through transesterification of vegetable oils or animal fats with short chain alcohols. However, the lack of oil feedstocks limits the large-scale development of biodiesel to some extent. Recently, much attention has been paid to the development of microbial, oils and it has been found that many microorganisms, such as algae, yeast, bacteria, and fungi, have the ability to accumulate oils under some special cultivation conditions. Compared to other plant oils, microbial oils have many advantages, such as short life cycle, less labor required, less affection by venue, season and climate, and easier to scale up. With the rapid expansion of biodiesel, microbial oils might become one of potential oil feedstocks for biodiesel production in the future, though there are many works associated with microorganisms producing oils need to be carried out further. This review is covering the related research about different oleaginous microorganisms producing oils, and the prospects of such microbial oils used for biodiesel production are also discussed.  相似文献

研究在发酵培养基中添加色醇对斯达氏油脂酵母发酵的影响.结果表明,在接种后0 h或12 h时添加色醇,能够明显抑制菌体生长和油脂积累;而在培养24 h或36 h添加,能够明显促进菌体生长,并增强菌体对底物利用率.与对照组比较,在36 h添加100 μmol/L色醇,生物量、油脂量和脂肪系数分别增加7.4%、13.9%和14.2%,发酵时间缩短13.3%,明显提高了油脂生产效率.气相色谱分析表明,添加色醇对菌油脂肪酸组成及其相对含量无显著影响.实验结果有助于建立调控油脂发酵的新策略,具有重要的理论和工程应用意义.  相似文献
