A 16S rRNA sequence analysis of Syntrophobacter wolinii was done by using PCR amplification of the 16S rRNA-genes from DNA isolated from the S. wolinii-Desulfovibrio sp. coculture. Phylogenetic analysis using the obtained sequence revealed that S. wolinii was not related to bacteria growing syntrophically on other fatty acids than propionate, but was related to sulfate-reducing bacteria. The increase in sister-chromatid exchanges induced by 5 chemicals, with different DNA damaging and carcinogenic activities, was studied in short-term foetal-mouse cultures. A significant increase in SCE was induced by N-methyl-N'-nitro-N-nitrosoguanidine, N-diazoacetylglycine-amide, azaserine and methotrexate. k-Strophantin, on the contrary, was totally inactive. On a molar basis, MNNG was the most active chemical followed by MTX, AZS and DGA, in that order. At equitoxic concentrations (D37), the order of SCE-inducing abilities was MNNG, DGA, AZS and MTX. Compared with previous data, at equitoxic concentrations, the most DNA-damaging agents were also the most effective in inducing SCE. The SCE increase seems to correlate not with unspecific cytotoxicity but more with DNA damage or other damage at the genome level. MTX, a non-mutagen, which induced SCE only at toxic levels, could be considered a false positive because this positivity may reflect an enhancement of incorporation of 5-BrdUrd into DNA. The positive results obtained with AZS suggest a sufficient sensitivity of the method for detecting relatively weak carcinogens. 相似文献

Summary The sequences of two rubredoxins isolated from the sulfate reducing bacteria:Desulfovibrio vulgaris andDesulfovibrio gigas have been elucidated. They have similar sequences but many more differences occur than would be expected from two bacteria of the same genus. Of the 52 sites, only 37 are occupied by identical residues. The primary structures are compared with those of the anaerobic bacteria rubredoxins ofClostridium pasteurianum, Micrococcus aerogenes, Pseudomonas oleovorans andPeptostreptococcus elsdenii: only 12 identities are found, mostly in the two clusters that contain two iron-bound cysteines each. A phylogenetic tree based on the primary structures is presented and possible relations with plant and bacterial ferredoxins are discussed. During a 9 day period after anthesis the concentration of reducing sugars showed a 6-fold increase in fruits of Citrullus lanatus, and a 2-fold increase in those of Capsicum annuum. These increases were associated with acid invertase, the specific activity of which was high in ovaries at anthesis and which increased 5-fold in watermelon and 1.5-fold in pepper during the same period. Sucrose synthase apparently plays only a minor role in sucrose hydrolysis. Changes in sugar concentrations and both acid invertase and sucrose synthase activities were similar in fruits developed both after pollination or hormone (NAA) treatment of ovaries. In non-pollinated ovaries of watermelon there was also an increase in invertase activity up to 6 days after anthesis which paralleled the increase in activity in seeded and parthenocarpic fruits. However, there was no increase in either reducing sugars or sucrose, indicating that sucrose is not imported into non-pollinated ovaries. Two new compounds with the aldehyde group, cryptoporusal A (1) and 9-formyl-4-hydroxy-6, 9-dimethyl-1-O-β-D-xylosopyranosyl-8a,9-dihydro-ace-naphthylen-8-one (2), were isolated from the ethyl acetate extract of the fruiting bodies of Cryptoporus volvatus (Pk.) Hubbard. Their structures were elucidated by extensive spectroscopic studies, and comparison with literature data. Furthermore, they were evaluated for their in vivo cytotoxicity against five human cancer cell lines and antioxidant activity. Reactive oxygen species play an important role in adriamycin (ADR) nephropathy. We showed by in vivo electron paramagnetic resonance (EPR) that renal reducing ability (RRA) declined on the 7th day after ADR administration. Proteinuria appeared after the decline in RRA. The aim of this study was to prove by in vivo EPR whether the decline in RRA is altered by scavengers such as dimethyl sulfoxide (DMSO) and dimethylthiourea (DMTU) and that it is this change which is responsible for the proteinuria in ADR nephropathy. By showing that DMSO and DMTU ameliorate the RRA, we demonstrate that the decline in RRA is related to ADR-induced proteinuria. 相似文献

Ferredoxin, cytochrome c3 and hydrogenase are specific partners of the sulfate reduction pathway of Desulfovibrio desulfuricans Norway and might be exemplary for electron exchange mechanism studies. Cytochrome c3 contains four low redox potential haems for 13 000 molecular weight. Two ferredoxins isolated from the same bacteria are dimers of 6 000 molecular weight per subunit (Ferredoxin I: one (4 Fe-4S) cluster per subunit, ferredoxin II: two (4 Fe-4 S) clusters per subunit). The amino acid sequence of ferredoxin I is reported and compared to the ferredoxin II sequence. The structural characteristics of ferredoxins and cytochrome c3 should allow a discussion on the nature of the interaction. 1H-NMR spectra of ferredoxin I and cytochrome c3 in the absence and presence of ferredoxin are presented. 相似文献
