Abstract Bacteria from an anaerobic enrichment reductively removed chlorine from the ortho- position of 2,3,6-trichlorobenzoic acid (2,3,6-TBA) producing 2,5-dichlorobenzoate (2,5-DBA). The strictly aerobic bacterium Pseudomonas aeruginosa JB2 subsequently used 2,5-DBA as a growth substrate in the presence of oxygen. The anaerobic dechlorinating microbial population was grown with P. aeruginosa JB2 in continuous culture. Inside the liquid culture, a nylon netting, on a stainless-steel support, contained vermiculite particles to provide a strictly anaerobic environment within the aerated culture. Complete mineralization of 2,3,6-TBA depended on the extent of oxygen input into the reactor. Under strictly anaerobic conditions 2,5-DBA and Cl were produced stoichiometrically through the reductive dechlorination of 2,3,6-TBA. This process of reductive dechlorination was not inhibited by (moderate) aeration resulting in an O2-concentration of 0.3–0.5 μM in the culture liquid.  相似文献

目的:研究4周中等强度有氧运动结合饮食控制对肥胖女青年、少年血清总胰岛素样生长因子1(IGF-1)、IGF-1结合蛋白3(IGFBP-3)水平和IGF-1活性的影响及其在体脂减少和糖脂代谢改善中的作用。方法:招募9名18~19岁肥胖女青年和30名14~16岁肥胖女少年,进行全封闭的4周中等强度有氧运动结合饮食控制干预。运动项目有游泳、跑步、健身操等,每周运动6 d,每天运动4 h,每运动30 min,休息5 min。运动强度从第1周的低强度(运动后即刻心率约100~120次/分)递增至第2~4周的中强度(运动后即刻心率约120~140次/分)。根据基础代谢率给予每日1 400或1 600 kcal的总能量。另招募正常体重女青年和女少年各9名作为对照组。检测肥胖女青年、少年在4周干预前、后和对照组女青年、少年的体重、身体质量指数(BMI)、腰围、空腹血糖、胰岛素和血脂水平以及血清总IGF-1和IGFBP-3的水平和IGF-1活性。结果:①与对照组相比,肥胖女青年、少年的血清总IGF-1和IGFBP-3水平均降低且肥胖女少年的IGF-1活性降低;②4周中等强度有氧运动结合饮食控制在显著降低肥胖女青年、少年的体脂、腰围和改善糖脂代谢的同时,降低血清IGFBP-3水平、增加IGF-1活性,但血清总IGF-1水平没有显著改变。且相关性分析显示IGF-1活性增加可能与肥胖女青年的腰围减少有关,但血清IGFBP-3水平的降低和IGF-1活性的增加与糖脂代谢的改善没有显著相关性。结论:4周中等强度有氧运动结合饮食控制降低肥胖女青年、少年的血清IGFBP-3水平、增加IGF-1活性;且IGF-1活性的增加可能与运动结合饮食控制降低肥胖女青年的腰围有关。  相似文献

目的:探讨小半夏加茯苓汤对小鼠心脏移植模型中移植心存活时间的延长效果及作用机制。方法:将接受过腹腔心脏移植手术的小鼠分为实验组和对照组,自移植手术当日起至术后7天,每日经口给药,通过腹部触诊判断移植心脏跳动情况,移植术后30日,取部分小鼠移植心脏进行病理检查,取脾细胞进行淋巴细胞培养,并检测培养液中白细胞介素(IL)-2、IL-4、IL-10和干扰素(IFN)-γ的浓度。取移植心脏存活时间超过30天的小鼠脾脏,通过流式细胞术检测调节性T细胞(Regulatory Tcells,Tregs)的含量。结果:与对照组相比,实验组的移植心存活时间明显延长(P0.01),心脏病理切片结果提示实验组的心脏血管病变受到了抑制,脾细胞培养液中IL-2和IFN-γ浓度明显降低,而IL-4和IL-10的浓度升高,脾细胞CD4阳性细胞中CD25和Foxp3均阳性的细胞含量明显高于对照组中同类细胞含量。结论:小半夏加茯苓汤可以显著延长小鼠移植心脏的存活时间,可能与其诱导调节T细胞的产生、改变受体小鼠体内Th-1和Th-2细胞因子的平衡及抑制IL-2、IFN-γ减轻心脏损伤有关。  相似文献

Lymphoblastoid cell lines (LCLs) established from chromosomal breakage syndromes or related genetic disorders have been used to study the effects of mutagens on human lymphoid cells. The disorders studied include xeroderma pigmentosum, ataxia telangiectasia, Fanconi's anemia, Bloom's syndrome and Cockayne's syndrome. Three approaches were used to assess the cells' ability to cope with a particular mutagen: (1) assaying recovery of DNA snythetic capabilities as measured by [3H]thymidine (dT) incorporation; (2) measurements of classical excision DNA repair by isopyknic sedimentation of DNA density labeled with 5-bromo-2-deoxyuridine (BrdU); (3) determining cell survival by colony formation in microtiter plates. LCLs established from xeroderma pigmentosum showed increased sensitivities to ultraviolet (354 nm) light and N-acetoxy-2-acetylaminofluorene (AAAF) as determined by DNA synthesis or colony formation and had diminished levels of excision-repair. Cockayne's syndrome LCLs, on the other hand, had increased sensitivities to ultraviolet (UV) light, AAAF and N-methyl-N′-nitro-N-nitrosoguanidine (MNNG) while showing near normal levels of DNA-repair after treatment with each agent. An LCL established from ataxia telangiectasia had decreased DNA repair synthesis and defective colony-forming ability following treatment with MNNG. LCLs, in addition to ease of establishment, appear likely to provide useful material for the study of DNA repair replication and its relationship to carcinogenesis.  相似文献

Evidence is accumulating that early emotional experience interferes with the development of the limbic system, which is involved in perception and regulation of emotional behaviors as well as in learning and memory formation. Limbic brain regions, as well as hypothalamic regions and other, nonlimbic areas contain specific neuron subpopulations, which express and release corticotropin releasing factor (CRF). Since these neurons serve to connect limbic function to endocrine, stress-related responses, we proposed that stressful experience during early postnatal brain development should interfere with the development of CRF-containing neurons, particularly in brain regions essential for emotional regulation. Applying neonatal separation stress (daily 1 h separation from the parents and litter mates) as stressor, the number of immunocytochemically identified CRF-expressing neurons/fibers was quantified in the amygdala, hippocampus, paraventricular nucleus of the hypothalamus, piriform cortex, and the somatosensory cortex of 3-week-old stressed and nonstressed Octodon degus, a semi-precocial rodent. Compared to controls neonatally stressed animals showed significantly lower levels of CRF-positive fibers (-60%) in the central amygdala, significantly less CRF-positive neurons in the dentate gyrus (-28%) and the CA1 region (-29%) and significantly lower CRF cell densities in the somatosensory cortex (-26%). On the other hand, we found significantly higher numbers of CRF-immunoreactive neurons in the basolateral amygdaloid complex (+192%) of stressed animals compared to nonstressed controls. No differences in CRF-immunoreactive cell densities were detected in the other regions. Additional behavioral analysis revealed significantly elevated exploratory behavior (+34%) in stressed animals compared to controls, which might indicate reduced anxiety in the stressed animals.  相似文献

目的:探究顺铂联合针灸疗法对晚期肺癌患者血清集落刺激因子(CSF)、肿瘤坏死因子(TNF-alpha)与白细胞介素2(IL2)水平的 影响。方法:选取我院肿瘤科收治的晚期肺癌患者60 例,并将其随机分为对照组和实验组。对照组30 例患者仅予顺铂常规治疗; 实验组30 例患者顺铂在常规治疗的基础上加用针刺肺俞、丰隆、中府、太渊、尺泽、心俞、天宗、膻中、膏肓、尺泽、背压痛点、灸丹 田穴位。治疗前后,检测和比较两组患者的血清CSF、TNF-alpha与IL2 水平。结果:治疗前,两组患者的血清CSF、TNF-alpha、IL2 水平比 较均无统计学差异(P>0.05);治疗后,两组的血清CSF、TNF-alpha、IL2水平均较治疗前降低,且实验组显著低于对照组,差异均有统计 学意义(P<0.05)。实验组的治疗总有效率明显高于对照组,脱发、失眠、白细胞减少的发生率均明显低于对照组,差异均具有统计 学意义(P<0.05)。结论:顺铂联合针灸治疗能够明显降低晚期肺癌患者的血清CSF、TNF-alpha与IL2 水平,进而可能提高患者的免疫 功能和临床疗效,值得临床进一步研究和推广应用。  相似文献

以果实性状差异较大的一年生辣椒材料C.annuum B9431(P1)和灌木辣椒材料C.frutescens H108(P2)为亲本,构建4世代群体(P1、P2、F1、F2),应用数量性状主基因+多基因混合遗传模型方法对辣椒6个果实性状进行遗传分析,为辣椒果实性状QTL定位及分子标记辅助育种研究奠定理论基础。结果表明:6个果实性状均符合2对主基因+多基因遗传模型。其中,单果重量、果实纵径、果实横径、果形指数和果肉厚度5个性状均符合E-0模型,即2对加性-显性-上位性主基因+加性-显性-上位性多基因混合遗传模型;果柄长度最佳遗传模型为E-5,即2对完全显性主基因+加性-显性多基因模型。单果重量、果实纵径、果实横径、果形指数、果肉厚度和果柄长度主基因遗传率分别为87.64%、37.67%、82.46%、94.82%、83.33%和40.00%,多基因遗传率分别为7.50%、54.56%、10.53%、0.27%、12.96%和37.78%。  相似文献
