SYNOPSIS. The shock reaction of Euglena gracilis strain Z to a sudden increase in light intensity (the “direct photophobic response”) was examined by high speed cinemicrography. The response is expressed as a turning reaction toward the dorsal side of the cell, after a transduction time of 0.1–0.5 sec after the onset of stimulation. Transduction times, turning rates, and flagellar beat frequencies were measured by analyzing the filmed sequences. The experimental data are consistent with a mechanism of directional homeostasis in negative phototaxis that is based upon shading of the photoreceptor by the cell's posterior end.  相似文献

Kairomones from juvenile perch (Perca fluviatilis) enhance phototactic descent of Daphnia to the hypolimnion of lakes following upon light intensity increases at dawn. Kairomones are not present in the hypolimnion and ascent in the evening might be insufficient for the animals to reach the epilimnion again. The hypothesis that sensitization is maintained for some time in the absence of fish exudates was tested. Experiments showed that sensitization was indeed maintained for 5–6 days. Thus a kind of “memory”; for fish kairomones exists, resulting in an enhanced ascent from a fishless hypolimnion at sunset.  相似文献

Lipid-protein interactions are known to play a crucial role in structure and physiological activity of integral membrane proteins. However, current technology for membrane protein purification necessitates extraction from the membrane into detergent micelles. Also, due to experimental protocols, most of the data available for membrane proteins is obtained using detergent-solubilized samples. Stable solubilization of membrane proteins is therefore an important issue in biotechnology as well as in biochemistry and structural biology. An understanding of solubilization effects on structural and functional properties of specific proteins is of utmost relevance for the evaluation and interpretation of experimental results. In this study, a comparison of structural and kinetic data obtained for the archaebacterial photoreceptor/transducer complex from Natronomonas pharaonis (NpSRII/NpHtrII) in detergent-solubilized and lipid-reconstituted states is presented. Laser flash photolysis, fluorescence spectroscopy, and electron paramagnetic resonance spectroscopy data reveal considerable influence of solubilization on the photocycle kinetics of the receptor protein and on the structure of the transducer protein. Especially the protein-membrane proximal region and the protein-protein interfacial domains are sensitive towards non-native conditions. These data demonstrate that relevance of biochemical and structural information obtained from solubilized membrane proteins or membrane protein complexes has to be evaluated carefully.  相似文献

【目的】旨在获得铜绿丽金龟 Anomala corpulenta Motschulsky敏感的波谱范围及性别差异。【方法】本研究利用室内行为学的方法,测试了在波长为340~610 nm的14个单色光刺激下铜绿丽金龟的趋、避光行为反应,并计算趋、避光反应率曲线。【结果】观测显示,各单色光均能引起铜绿丽金龟产生一定的趋、避光反应,其雌雄虫的趋光敏感光区位于紫外光(405 nm)、蓝光(460 nm)和绿光区(505和570 nm),性别对趋光行为有一定的影响,但是仅存在于趋光反应率曲线波峰的大小,而对波峰的位置没有影响。铜绿丽金龟的避光行为反应,在雌雄之间无明显的性别分化,其避光敏感光谱为紫外光(380 nm)、蓝光(440 nm)和绿光区(492和505 nm)。【结论】铜绿丽金龟成虫对不同波长光的趋性存在差异,性别对其光谱行为反应有一定的影响。该研究结果为铜绿丽金龟光视觉的深入研究奠定必要的理论基础,也为利用其趋光性对该金龟甲进行综合治理提供科学依据。  相似文献

The silkworm, Bombyx mori, was reared aseptically on a synthetic diet with and without β-carotene and the effects of carotenoid and vitamin A deficiency on photosensitivities in larval phototaxis, visual function and adult eclosion were studied.β-Carotene or vitamin A acted as a growth-promoting factor in continuous darkness and under photoperiodic conditions. The deficiency of β-carotene decreased the larval phototactic response as growth proceeded. The offspring larvae from eggs laid by β-carotene-deficient moths also lost the phototactic response, but successive rearing with dietary β-carotene or vitamin A re-established the response. The deficiency of β-carotene caused the loss of the electric response by light stimuli in the ocelli of fifth instar larvae and the compound eyes of adult moths. These results indicate that vitamin A is essential for visual function in the silkworm, as reported in other insects. The lack of carotenoid did not affect the development of the pupae or the specific time of eclosion which is regulated by a photoperiodic condition of pupal stage. This observation suggests that the carotenoid and its derivative are not involved in photoreception for the entrainment of the adult eclosion of the silkworm.  相似文献

The reflection properties of the stigma in male gametes of Ectocarpus siliculosus (Dillw.) Lyngbye were investigated using confocal laser scanning microscopy in the epireflection contrast mode. The complex reflection pattern obtained after optical xy (horizontal) and xz (vertical) sectioning was consistent with stigma ultrastructure as revealed by serial thin sections. The intensity and pattern of the reflection signal varied with the orientation of the cell/stigma to the incident laser light. Maximal reflection occurred only in approximately normal orientation of the stigma to the light source. Focusing of reflected light from an elongated concave depression of the stigma on the region of the flagellar swelling was observed in xy and xz sections of living and fixed gametes. The results indicate the importance of mechanisms (focusing) other than quarter-wave interference reflection in signal amplification by the eyespot of flagellate algae.  相似文献

First-instar larvae of the rice leaffolder Cnaphalocrocis medinalis (Guene) oriented upwards on a vertically inclined filter paper and downwards on a rice leaf when the light source was from above. The direction of orientation was reversed when the light source was from below. The opposite directions of orientation on these substrates persisted when the substrates were held horizontal with light source from the sides. No difference was observed in the magnitude of reaction on susceptible and resistant rice varieties or when the rice leaf was held with its distal end pointing down. On non-host grasses, however, the directional orientation was lost. Dipping rice leaves in hexane decreased the magnitude of reaction. On dried rice leaf, the reaction was similar to that on filter paper but was of a lower magnitude. Application of rice plant volatile extracts on a filter paper decreased the intensity of upward orientation while 500 ppm of the volatile extracts applied on dried leaf caused a loss of oriented movement. Application of a mixture of seven volatile chemicals, identified from rice, decreased the magnitude of reaction significantly. It is concluded that rice plant volatiles along with some other hitherto unidentified rice plant factor cause a reversal of phototaxis, leading to downward orientation on the rice leaf.Based at the International Rice Research Institute  相似文献

All 3 photoreceptor types in the compound eye of Drosophila can evoke positive phototaxis. Here we describe input from R1–6 receptors which are very sensitive. Previous reports in this series of studies described input from R7 and R8, the other less sensitive receptors. Here we studied fast-walking phototaxis using extremely dim stimuli. We also studied input from the simple ocellar eyes. Thus, we can now summarize a complete synthesis of inputs and interactions among all compound eye and ocellar receptor types. Receptor-deficient mutants were used to establish receptor-specific input. Two other findings are presented: (1) eye colour pigments affect the spectral sensitivity for phototaxis; and (2) the ocelli interact to facilitate input from the compound eye receptor types. Possible mechanisms of receptor interaction are discussed in the light of these findings of positive input from all photoreceptor types in Drosophila.  相似文献
