The kinetic component (39 ps) for the energy transfer from a phycobilisome (PBS) to the photosystems was temperature-dependent while the components related to the kinetic processes within PBS, photosystem 2 (PS2) or PS1 were temperature-independent. The 39 ps component possessed the amplitude maximum at 647 nm but the minimum at 715 nm (room temperature) or 685 nm (0 °C), suggesting a direct energy transfer from C-phycocyanin to PS1 at room temperature but to PS2 at 0 °C. The effect of three different concentrations of amitrole (AM), a bleaching herbicide affecting carotenogenesis, on chloroplast ultrastructure, photosynthetic pigment contents, and photochemical activity was studied in two maize genotypes differing in photosynthetic characteristics. The content of photosynthetic pigments in leaves of plants treated with low (20 M) AM concentration was similar to control plants and no damaging effect of the herbicide on the ultrastructure of either mesophyll (MC) or bundle-sheath (BSC) cell chloroplasts was observed. Higher (60 and 120 M) concentrations of AM caused a significant decrease in the content of carotenoids (especially xanthophylls), which was followed by photooxidative destruction of chlorophylls and some alterations of chloroplast ultrastructure. MC chloroplasts appeared more sensitive to the damaging effect of AM compared to BSC chloroplasts. A significant decrease in the amount of both granal and intergranal thylakoids in MC chloroplasts was observed with the increasing concentration of AM. As regards BSC chloroplasts, rapid decrease in the volume density of starch inclusions was found in plants treated with higher concentrations of AM. When 120 M AM was used, both MC and BSC chloroplasts contained just a few thylakoid membranes that were strongly altered. The changes in the ultrastructure of MC chloroplasts were accompanied by the changes in their photochemical activity. The formation of chloroplast protrusions after treatment of plants with AM as well as in control plants was also observed. 相似文献

LIGHT ACCLIMATION IN PORPHYRIDIUM PURPUREUM (RHODOPHYTA): GROWTH,PHOTOSYNTHESIS, AND PHYCOBILISOMES1
Acclimation to three photon flux densities (10, 35, 180 μE.m?2.s?1) was determined in laboratory cultures of Porphyridium purpureum Bory, Drew and Ross. Cultures grown at low, medium, and high PPFDs had compensation points of <3, 6, and 20 μE-m?2.s?1, respectively, and saturating irradiances in the initial log phase of 90, 115, 175 μE.m?2.s?1 and up to 240 μE.m?2.s?1 in late log phase. High light cells had the smallest photosynthetic unit size (phycobiliproteins plus chlorophyll), the highest photosynthetic capacity, and the highest growth rates. Photosystem I reaction centers (P700) per cell remained proportional to chlorophyll at ca. 110 chl / P700. However, phycobiliprotein content decreased as did the phycobilisome number (ca. 50%) in high light cells, where as the phycobilisome size remained the same as in medium and low light cells. We concluded that acclimation of this red alga to varied PPFDs was manifested by the plasticity of the photosystem II antennae with little, if any, effect noted on photosystem I. 相似文献

The chlorophyll b-containing alga Mantoniella squamata was analyzed with respect to its capacity to balance the energy distribution from the light-harvesting antenna to photosystem I or photosystem II. It was shown, that this alga is unable to alter the absorption cross section of the two photosystems in terms of short-time regulations (state transitions). The energy absorbed by the LHC, which contains 60% of total photosynthetic pigments, is transferred to both photosystems without any preference. The stoichiometry of the two photosystems is found to be extremely unequal and variable during light adaptation. In high light, the molar ratio of P-680 per P-700 is found to be two, whereas under low light conditions this ratio accounts to nearly four. This very unbalanced stoichiometry of the reaction centers gives some new insights into the concept of the photosynthetic unit as well as in the importance of the regulation of the energy distribution. It is assumed that the high concentration of photosystem II can be understood as a mechanism to prevent the overexcitation of photosystem I. In addition, the changes im membrane protein pattern are not accompanied by variations in the ratio of appressed to nonappressed membranes as probed by ultrastructural analysis. It is suggested that the thylakoids are organized like a homogenous pigment bed. The lack of state transitions can be interpreted as a consequence of this unusual membrane morphology.Abbreviations Chl

The adaptability of the thylakoid membrane to extended photoperiod (from natural to 24 h) was studied using a photoperiod-sensitive species ( Lycopersicon esculentum Mill. cv. Trend) and a non-photoperiod-sensitive species ( Capsicum annuum L. cv. Delphin). Our results have shown that thylakoid membranes of both species adapt to an extended photoperiod by increasing their photosystem II to photosystem I ratio (PSII/PSI) in order to provide a more balanced energy distribution between both photosystems to improve quantum yield. In tomato plants, these results correspond with a lower chlorophyll (Chl) a/b ratio, a decrease in Chl associated with PSI light-harvesting chlorophyll a/b protein complexes and with an increase in Chl associated with PSII light-harvesting chlorophyll a/b protein complexes. In spite of these changes, the electron transport capacity through PSII and PSI per unit of Chl and the light saturation point of PSII remained unchanged. The inability of tomato plants to use supplemental light for an extended photoperiod is not the result of photoinhibitory conditions. In pepper plants a significant increase in electron transport capacity and in the light saturation point of PSII was found. There was a significant increase in CO2 assimilation when the light period was increased from 12 to 24 h. In contrast to tomato, pepper plants adapt to a 24-h photoperiod by increasing their carboxylation capacity which is accompanied by an increase in electron transport capacity and the light saturation point. 相似文献

Synechocystis sp. PCC 6803 when grown in the presence of sublethal (M) levels of cobalt chloride shows an enhancement of Photosystem II (PS II) catalyzed Hill reaction. This stimulation seems to be induced by cobalt ions as other metal ions inhibit para-benzoquinone catalyzed Hill reaction. At saturating white light intensity, this enhancement is two times over that of the control cells on unit chlorophyll basis. Analysis of the PS II electron transport rate at varying intensities of white, blue or yellow light suggests an increased maximal rates but no change in the quantum yield or effective antenna size of CoCl2-grown cells. There were no structural and functional changes in the phycobilisome as judged by the absence of changes in the phycocyanin/allophycocyanin ratio, fluorescence emission spectra, second derivative absorption spectra at 77 K and SDS-PAGE analysis of isolated phycobilisomes. The 77 K fluorescence emission spectra of the cells showed a decrease in the ratio of Photosystem I emission (F725) to Photosystem II emission (F685) in CoCl2-grown cells compared to the control cells. These observations indicate three possibilities: (1) there is an increase in the number of Photosystem II units; (2) a faster turnover of Photosystem II centers; or (3) an alteration in energy redistribution between PS II and PS I in CoCl2-grown cells which causes stimulation of Photosystem II electron transport rate.Abbreviations APC

Colored light modifies the relative concentration of chlorophyll-forms of the diatom Phaeodactylum tricornutum compared to white-light control. No change in the ratio carotenoids/chlorophylls was observed after 4 days exposure to green light (max: 530 nm), blue light (max: 470 nm) or red light ( > 650 nm) of same intensity.However, the absorption spectra were modified, the content in Ca 684, Ca 690, Ca 699 forms increased in red and green light cultures and photosynthetic unit size of PS II decreased by 30% in green and blue light cultures.Fluorescence emission and fluorescence excitation spectra according to the Butler and Kitajima method (1975) were carried out for each culture. Ca 669 form was predominant in the two photosystems. The newly appeared far red forms fluoresce at 715 nm like PS I forms.We conclude that these new forms originated in a rearrangement of PS II forms. They do not transmit excitation energy to reaction center of PS I and are disconnected from the other chlorophyll-forms of the photosynthetic antennae.Abbreviations ABS

Activity of the photosynthetic apparatus of synchronized cultures was studied with the xanthophycean alga Bumilleriopsis filiformis, following the kinetics of fluorescence induction and photooxidation of cytochrome f (= cytochrome c-553) of intact cells. During the beginning of the cell-division phase, minimum cellular photosynthetic activity is observed and a maximum after its completion, which is accompanied by corresponding changes in Hill reaction activity and re-reduction of cytochrome f by photosystem II light. At minimum activity, the level of steady state fluorescence was higher than at the maximum. This is due, at least in part, to the diminished electron flow between the two photosystems seemingly caused by decreased photosystem I activity. This explanation was suported by the kinetics of cytochrome-f photooxidation.Thus, electron transport activity of both photosystems appears to vary during the cell cycle.Abbreviations pBQ

The functional organizations of thylakoid membranes from wild type pea ( Pisum sativum L. cv. Kapital) and two viable mutants with low chlorophyll (Chl) contents were compared. Nuclear mutations in mutants 7 and 42 led to two- and three-fold decrease in total chlorophyll content, respectively. In spite of low Chl content mutants showed 80% photosynthetic activity, biological productivity, and seed production. It has been shown that mutant membranes differed from that of wild type by Chl distribution between the pigment-protein complexes and by stoichiometry of the main electrontransport complexes. The ratio photosystem I (PSI): photosystem II (PSII): cytochrome (Cyt) bjf complex: Chl was 1:1.1:1.2:650 in wild type chloroplasts, 1:1.8:1.7:600 in mutant 7 , and 1:1.5:1.9:350 in mutant 42 . PSI- and PSII-dependent electron-transport activities were enhanced in the mutants per mg Chl in proportion to number of reaction centers. The activity of the non-cyclic electron-transport chain increased in proportion to PSII and Cyt bjf complexes. The amount of ATP synthetase per unit of Chl as estimated by H− ATPase activity was much greater in mutant thylakoids, which is favorable for photosynthetic energy transduction. The low content of the light-harvesting complexes (LHC) in mutants is compensated by an increase of the number of PSII and Cyt bjf complexes, which eliminates the bottleneck at the site of plastoquinone oxidation. 相似文献
