Summary Current procedures for isolating intestinal epithelial cell surface and intracellular membranes are based on the assumption that each organelle is marked by some unique constitutent. This assumption seemed inconsistent with the dynamic picture of subcellular organization emerging from studies of membrane turnover and recycling. Therefore, we have designed an alternative fractionation which is independent ofa priori marker assignments. We subjected mucosal homogenates to a sequence of separations based on sedimentation coefficient, equilibrium density, and partitioning in aqueous polymer twophase systems. The resulting distributions of protein and enzymatic markers define a total of 17 physically and biochemically distinct membrane populations. Among these are: basal-lateral membranes, with Na,K-ATPase enriched 21-fold; brush-border membranes, with alkaline phosphatase enriched as much as 38-fold; two populations apparently derived from the endoplasmic reticulum; a series of five populations believed to have been derived from the Golgi complex; and a series of five acid phosphatase-rich populations which we cannot identify unequivocally. Each of the five enzymatic markers we have followed is associated with a multiplicity of membrane populations. Basallateral, endoplasmic reticulum, and Golgi membranes contain alkaline phosphatase at the same specific activity as the initial homogenate. Similarly, Na,K-ATPase appears to be associated branes at specific activities two-to seven-fold that of the initial homogenate.  相似文献

Considerable inherent variations in the relation between macropterous and brachypterous wing forms, and nymphal density were found in field populations of the brown planthopper, Nilaparvata lugens Stål (Homoptera: Delphacidae), collected from various locations in Japan. When compared under uniform laboratory rearing conditions, most of the female populations exhibited higher ratios of macropters with increasing nymphal density, but some showed extremely high proportion of brachypters and the others were highly macropterous, over broad ranges of density. These results indicate the possibility that the planthoppers in Japan, which are known not to persist in winter, are derived from different migration sources.About ten generations of successive selection for brachyptery from a population showing usual density-dependent wing morphism generated populations similar to highly brachypterous ones mentioned above. Genetic analysis of the inheritance of wing morphism revealed that brachyptery in the females was controlled by a single pair of dominant alleles. However, in the males wing forms did not segregate so clearly in the crossing experiments. This suggests that wing morphism in N. lugens in under sex-limited inheritance.
Einwanderung von Nilaparvata lugens mit unterscheidlicher Reaktion auf Populationsdischte bei der Flügelausbildung
Zusammenfassung In Feldpopulationen von Nilaparvata lugens Stål., welche in verschiedenen Regionen Japans gesammelt wurden, bestand in der Beziehung zwischen makropteren bzw. brachypteren Flügelformen und der Larvendichte eine beträchtliche Variation. Unter einheitlichen Zuchtbedingungen im Laboratorium stieg der Makropterenanteil bei den meisten Weibchenpopulationen mit steigender Temperatur; bei einigen Populationen hingegen war entweder der Brachypterenanteil oder der Makropterenanteil extrem hoch und zwar über weite Dichtebereiche. Dies deutet auf die Möglichkeit hin, dass die Zikade in Japan, wo sie bekanntlich nicht überwintert, jeweils aus verschiedenen Quellen einwandert.Wenn eine Population mit der üblichen dichteabhängigen Flügelausbildung 10 Generationen lang auf Brachypterie selektioniert wurde, entstanden Populationen, die den erwähnten hochbrachypteren Populationen aus dem Feld glichen. Die genetische Analyse der Vererbung der Brachypterie ergab, dass bei Weibchen ein einzelnes dominantes Allel verantwortlich ist. Bei Männchen dagegen trennten sich bei Kreuzungsexperimenten die Flügelformen nicht so klar. Dies deuted auf Unterschiede zwischen den Geschlechtern bei der Vererbung der Flügelformen.

The softening of wet lipid bilayer membranes during their gel-to-fluid first-order phase transition is studied by computer simulation of a family of two-dimensional microscopic interaction models. The models include a variable number, q, of lipid chain conformational states, where 2q10. Results are presented as functions of q and temperature for a number of bulk properties, such as internal energy, specific heat, and lateral compressibility. A quantitative account is given of the statistics of the lipid clusters which are found to form in the neighborhood of the transition. The occurrence of these clusters is related to the softening and the strong thermal density fluctuations which dominate the specific heat and the lateral compressibility for the high-q models. The cluster distributions and the fluctuations behave in a manner reminiscent of critical phenomena and percolation. The findings of long-lived metastable states and extremely slow relaxational behavior in the transition region are shown to be caused by the presence of intermediate lipid chain conformational states which kinetically stabilize the cluster distribution and the effective phase coexistence. This has as its macroscopic consequence that the first-order transition apperas as a continuous transition, as invariably observed in all experiments on uncharged lecithin bilayer membranes. The results also suggest an explanation of the non-horizontal isotherms of lipid monolayers. Addition of 15L-hydroperoxy-5,8,11,13-eicosatetraenoic acid (15-HPETE) to human leukocytes led to the formation of a novel series of compounds containing four conjugated double bonds. The yield of tetraenes was increased approx. 100-fold when ionophore A23187 (5 μM) was added simultaneously with 15-HPETE. The structure of the major tetraene was established by physical methods as well as by chemical degradation and found to be 5,6,15L-trihydroxy-7,9,11,13-eicosatetraenoic acid.  相似文献

The purpose of this study was to determine and compare the follicular phase steroid hormone secretion into the utero-ovarian vein by the ovary with a dominant follicle and the contralateral ovary in the same baboon. Serial utero-ovarian vein blood from both sides was collected in 25 baboons by the use of a laparoscope on alternate days, starting on day 1 or 3 of the cycle and continuing through 2 to 3 days post-ovulation. Approximately 3–4 days before the day of expected ovulation, samples were collected at 8-hr intervals. Steroids estradiol (E2) and progesterone (P) were measured in all utero-ovarian vein plasma by radioimmunoassay. In the peripheral plasma, E2, P, LH, and FSH measurements were carried out. Concentrations of steroids were significantly higher on the side of the ovulating ovary by day 5 before ovulation. Individual plots however, indicated that some baboons may establish the dominant side as early as day 11 before ovulation. The preovulatory gonadotropins had a differential effect on the two ovaries. For example, E2 values on the ovulatory side ovary declined after increases in LH/FSH, whereas on the contralateral side these values had increased. Both sides showed increases in the level of P with the increases in LH. The mean interval from E2 peak to LH peak was 24 hrs and LH peak to ovulation was 24 hrs.  相似文献

We present a theoretical model of a lipid bilayer in its gel state which explicity couples the vertical displacements of the lipid chains to their conformational state. In this model the chains are free to move longitudinally under a potential due to the neighbouring chains. The potential is due to a restoring force with a force constant k and thus acts to keep them in the local plane as defined by their nearest neighbours. It is demonstrated that the force constant k is directly related to the internal bilayer pressure, , and that if a value =33 dynes/cm is assumed then k=17.3 dynes/cm. Steric effects are explicity included by allowing chains to twist into free volume created by the vertical displacement of neighbouring chains. The Hamiltonian is expressed in terms of the projection operators, P ij , describing the displacement of chain i relative to a neighbour j, and G ij describing the direction of a twist in chain i. The model is solved both analytically and via Monte Carlo simulations for a one-dimensional system. Single-channel conductance fluctuations are analysed for gramicidin A incorporated into binary-mixed black lipid membranes of charged phosphatidic acid and neutral lecithin in different molar ratios. At very low Ca++ concentrations in the electrolyte (i.e. in the presence of EDTA) homogeneous lipid mixtures are identified through their conductance and life time probability distributions for integral gramicidin pores. As for the pure lipid components, the conductance histograms each show a single maximum with regular width and for all channels a single mean lifetime is found.For Ca++-levels (10-6–10-5 M) that are close to the critical demixing concentration (10-4 M) unusually broad conductance distributions and reduced lifetimes are found provided the PC content, x, of the membrane is close to the critical mixture (x crit0.5). We interpret this as a first example of the coupling of a membrane function (the transport of ions) to a lipid matrix with locally fluctuating composition close to a critical demixing point.For the conductance histogram of gramicidin A in an equimolar mixture of PA and PC shows two well-separated maxima. A correlation analysis between conductance and lifetime of the single pores shows that the two channel populations also differ significantly in their mean channel lifetime, *. Summary Chromosomes were isolated in a preparative scale by synchronisation of CHO cells with a double Thymidine block followed by an arrest in the metaphase by addition of Colcemid. Under proper cultivation conditions a mitotic index of 77% total cells could be routinely achieved. Bulk chromosome preparations free of nuclei and other subcellular particles have been obtained by low speed centrifugation followed by a 60 transfer countercurrent distribution using aqueous two phase systems composed of polyethylenglycol and dextran. The partition of CHO chromosomes previously purified in aqueous two phase systems were studied further to develop a protocol for the separation and isolation of individual chromosomes. Partition experiments with chromosomes changing the electrostatic phase potential by addition of charged PEG-derivatives suggest the existence of relatively highly charged chromosome groups. Most promising results with regard to separation were obtained using two PEG-derivatives, which interact specifically with the bases in DNA. For this affinity partitioning a GC- and AT-specific macroligand were employed. Comparing CCD's using each of these ligands information on the GC and AT content of exposed DNA in the chromosomes groups could be derived, demonstrating that specific sequences of DNA are accessible at the surface of metaphase chromosomes.  相似文献

When the extent of liquid holding recovery (LHR) was measured as a function of the temperature at the time of liquid holding and the Arrhenius plot was made, two distinctive phases for the LHR were demonstrated in UV-irradiated RecA- derivative of E. coli ole28E1, which are unable to synthesize and degrade unsaturated fatty acids. The inflection temperatures were 17-18 degrees C, 23-24 degrees C and 28-30 degrees C for linoleate-, oleate- and elaidate-grown cells, respectively. These temperatures well corresponded to the phase transition temperatures of the cell membrane supplemented with the fatty acid. It is therefore concluded that at least a component involved in in vivo excision repair in E. coli is associated with cell membrane.  相似文献
