Although selective pesticides are recommended to use in integrated pest management (IPM) programmes, they could adversely affect biological agents. So, the aim of the current study was to investigate the LC30 of pirimicarb and thiamethoxam on Hippodamia variegata. Two pesticides, pirimicarb and thiamethoxam, when applied at concentrations of 1522.8 and 251.3?mg (ai)/L, respectively, on the third instar larvae of the insect, produced 30% mortality. Also, results showed that neither pirimicarb nor thiamethoxam affect stage age distribution (cx), fecundity (eggs/female) and adult development time. These two pesticides extended preadult duration significantly (p?<?0.0001, F?=?31.22, df?=?122). Also, survival rate (lx) and age-specific reproductive value (vx) decreased and some changes in age-specific life expectancy (ex) happened. Generally, more adverse effects were found in the population treated by thiamethoxam. The results showed that pirimicarb and thiamethoxam have potential to harm the predatory ladybird in IPM programme, though in sublethal doses. 相似文献

ABSTRACTIn Portugal, the European clam (Ruditapes decussatus) is an important commercial resource. Óbidos Lagoon is a strong candidate as a cultivation area to increase European clam exploitation. However, the reproductive biology of this population has not been described. In this work, the reproductive cycle of R. decussatus was characterized by determining gonadal development stages, gonad index, condition index, and biochemical composition. The relationship between reproduction and environmental parameters (sea surface temperature, chlorophyll a, and particulate organic matter) was assessed. Ruditapes decussatus had an annual reproductive cycle. The gametogenic cycle started in late winter, and the ripe stage in spring was followed by spawning that began at the end of spring/early summer and extended until early autumn. The subsequent period of sexual rest occurred during the winter. Condition index showed seasonal variations related to food availability (chlorophyll a). The European clams in Óbidos Lagoon recovered rapidly after their reproductive period, most likely owing to the availability of food. This study will help to improve sustainable management of this wild stock and is important for future aquaculture development of this species. 相似文献

An accurate solvation model is essential for computer modeling of protein folding and other biomolecular self-assembly processes. Compared to explicit solvent models, implicit solvent models, such as the Poisson-Boltzmann (PB) with solvent accessible surface area model (PB/SA), offer a much faster speed—the most compelling reason for the popularity of these implicit solvent models. Since these implicit solvent models typically use empirical parameters, such as atomic radii and the surface tensions, an optimal fit of these parameters is crucial for the final accuracy of properties such as solvation free energy and folding free energy. In this paper, we proposed a combined approach, namely SD/GA, which takes the advantage of both local optimization with the steepest descent (SD), and global optimization with the genetic algorithm (GA), for parameters optimization in multi-dimensional space. The SD/GA method is then applied to the optimization of solvation parameters in the non-polar cavity term of the PB/SA model. The results show that the newly optimized parameters from SD/GA not only increase the accuracy in the solvation free energies for ~200 organic molecules, but also significantly improve the free energy landscape of a β-hairpin folding. The current SD/GA method can be readily applied to other multi-dimensional parameter space optimization as well. 相似文献

AbstractThe emergence of the biopharmaceutical industry represented a major revolution for modern medicine, through the development of recombinant therapeutic proteins that brought new hope for many patients with previously untreatable diseases. There is a ever-growing demand for these therapeutics that forces a constant technological evolution to increase product yields while simultaneously reducing costs. However, the process changes made for this purpose may also affect the quality of the product, a factor that was initially overlooked but which is now a major focus of concern. Of the many properties determining product quality, glycosylation is regarded as one of the most important, influencing, for example, the biological activity, serum half-life and immunogenicity of the protein. Consequently, monitoring and control of glycosylation is now critical in biopharmaceutical manufacturing and a requirement of regulatory agencies. A rapid evolution is being observed in this context, concerning the influence of glycosylation in the efficacy of different therapeutic proteins, the impact on glycosylation of a diversity of parameters/processes involved in therapeutic protein production, the analytical methodologies employed for glycosylation monitoring and control, as well as strategies that are being explored to use this property to improve therapeutic protein efficacy (glycoengineering). Abstract A series of potential energy calculations have been carried out to estimate base sequence dependent structural differences in B-DNA. Attention has been focused on the simplest dimeric fragments that can be used to build long chains, computing the energy as a function of the orientation and displacement of the 16 possible base pair combinations within the double helix. Calculations have been performed, for simplicity, on free base pairs rather than complete nucleotide units. Conformational preferences and relative flexibilities are reported for various combinations of the roll, tilt, twist, lateral displacement, and propeller twist of individual residues. The predictions are compared with relevant experimental measures of conformation and flexibility, where available. The energy surfaces are found to fit into two distinct categories, some dimer duplexes preferring to bend in a symmetric fashion and others in a skewed manner. The effects of common chemical substitutions (uracil for thymine, 5-methyl cytosine for cytosine, and hypoxanthine for guanine) on the preferred arrangements of neighboring residues are also examined, and the interactions of the sugar-phosphate backbone are included in selected cases. As a first approximation, long range interactions between more distant neighbors, which may affect the local chain configuration, are ignored. A rotational isomeric state scheme is developed to describe the average configurations of individual dimers and is used to develop a static picture of overall double helical structure. The ability of the energetic scheme to account for documented examples of intrinsic B-DNA curvature is presented, and some new predictions of sequence directed chain bending are offered. 相似文献

Abstract Plasmid pEJ4, which is a derivative of pUC19 containing an insert with 60-bp-long · homopurine homopyrimidine tract from sea urchin P. miliaris histone gene spacer, was studied by chemical probes of the DNA structure osmium tetroxide and glyoxal. The former probe reacts with pyrimidine bases, while the latter forms a stable product only with guanine residues. These probes can thus be applied as specific probes for the homopyrimidine and homopurine strands, respectively. At pH 6.0 the site-specific modification of the homopurine · homopyrimidine tract by both probes was observed at native superhelical density of the plasmid. In the linear plasmid under the same conditions this modification was absent; it appeared, however, at more acid pH values. In supercoiled DNA the hypersensitivity of the homopurine homopyrimidine tract to osmium tetroxide did not substantially change when pH was decreased from 6.0 to 4.0. Changes in NaCl concentration at pH 4.5 did not influence the hypersensitivity to osmium tetroxide; at pH 6.0 this hypersensitivity decreased with increasing NaCl concentration. These results thus show that the chemical probes recognize an unusual protonated structure containing unpaired bases or non-Watson-Crick base pairs. At pH 5.6 the site-specific modification occurred at or near to the middle of the homopurine · homopyrimidine tract, suggesting that a hairpin may be involved in the unusual structure under the given conditions. In the present work, the adsorption kinetics of extended ligands on DNA duplexes at small fillings when molecules of DNA duplexes are on the underlayer within diffusion layer has been investigated. Both diffusion of ligands in solution (diffusion stage) and adsorption of ligands (kinetic stage) are taken into consideration at adsorption of ligands on DNA duplexes. Nonlinear system of differential equations describing adsorption of ligands where not only diffusion stage but also kinetic stage is taken into account, is obtained, moreover the equations allow localizing duplexes in arbitrary place within diffusion layer. Numeric solution of the equations makes possible to investigate the filling kinetics of DNA duplexes by ligands depending on parameters controlling adsorption process. It has been shown that depending on relation between adsorption parameters different kinetic regimes of adsorption – kinetic, complex, and diffusion regimes may be realized. 相似文献

Sleep-deprived people, or those performing extended monotonous tasks, can exhibit brief episodes in which they suspend performance and appear to fall asleep momentarily—behavioral microsleeps (“microsleeps”). In this study, microsleeps were identified using eye video and tracking response during a 20-min continuous tracking task undertaken by 16 healthy volunteers (mean age 24.9?yrs; 8 females, 8 males) in the early afternoon following a normally rested night and a night of restricted sleep (time-in-bed restricted to 4?h). Sessions were 1 wk apart and counterbalanced. Wrist actigraphy, self-reported sleepiness, and sleep quality were also recorded. We hypothesized that high microsleep rates when normally rested or after a night of sleep restriction would be related to poor sleep quality, sleep disturbance, circadian type, irregular sleep patterns, low daily sleep duration, or poor sleep efficiency. We also hypothesized that prior performance on a 10-min psychomotor vigilance task (PVT) (mean reaction time or number of PVT lapses) would be related to the number of microsleeps during the tracking task and that PVT performance could, therefore, be used as a fitness-for-duty indicator. The number of microsleeps during the tracking task increased following sleep restriction (mean 11.4 versus 27.9; p?=?0.03). There were no correlations between the number of microsleeps in the normally rested session and any of the actigraphically measured or self-reported sleep measures. However, the number of microsleeps following sleep restriction was correlated with sleep efficiency (r?=?0.73, p?=?0.001), sleep onset latency (r?=??0.57, p?=?0.02), and sleep onset time-of-day standard deviation (r?=??0.54, p?=?0.03) over 11 normally rested nights. There was no correlation between PVT performance and the subsequent number of microsleeps during the tracking task in either session. Attributes usually associated with beneficial nighttime sleep patterns—going to sleep at a similar time each night, falling asleep quickly, and infrequent arousals—were related to greater vulnerability to microsleeps following sleep restriction. There were intercorrelations between all the sleep measures associated with microsleep rate following sleep restriction, indicating that the measures form a pattern of behaviors and are not independently related to microsleep rate. Perhaps some people maintain a regular sleep pattern because they experience sleepiness the following day when their pattern is disrupted. Conversely, people with more variation in their sleep pattern may do so because this does not substantially increase sleepiness the following day. We conclude that people with consistent sleep patterns and efficient sleep may be more prone to microsleeps than other people when their usual regular pattern is disrupted by sleep restriction. 相似文献

Daily rhythms in different biochemical and hematological variables have been widely described in either diurnal or nocturnal species, but so far no studies in the rhythms of these variables have been conducted in a dual-phasing species such as the degus. The Octodon degus is a rodent that has the ability to switch from diurnal to nocturnal activity under laboratory conditions in response to wheel-running availability. This species may help us discover whether a complete temporal order inversion occurs parallel to the inversion that has been observed in this rodent's activity pattern. The aim of the present study is to determine the phase relationships among 26 variables, including behavioral, physiological, biochemical, and hematological variables, during the day and at night, in diurnal and nocturnal degus chronotypes induced under controlled laboratory conditions through the availability of wheel running. A total of 39 male degus were individually housed under a 12:12 light-dark (LD) cycle, with free wheel-running access. Wheel-running activity (WRA) and body temperature (Tb) rhythms were recorded throughout the experiment. Melatonin, hematological, and biochemical variables were determined by means of blood samples obtained every 6?h (ZT1, ZT7, ZT13, and ZT19). In spite of great differences in WRA and Tb rhythms between nocturnal and diurnal degus, no such differences were observed in the temporal patterns of most of the biological variables analyzed for the two chronotypes. Variation was only found in plasma urea level and lymphocyte number. A slight delay in the phase of the melatonin rhythm was also observed. This study shows the internal temporal order of a dual-phasing mammal does not show a complete inversion in accordance with its activity and body temperature pattern; it would appear that the switching mechanism involved in the degu's nocturnalism is located downstream from the pacemaker. The effects of experimental parameters including soil type, curing duration, inoculum size, and biomass and nutrients concentration on soil strengthening due to calcite precipitation by Sporosarcina pasteurii PTCC 1645 were investigated. The laboratory-scale mixing experiments on remolded samples were designed by the Taguchi method. Soil type proved to be the most incorporating factor, followed by curing time and nutrient concentration. The main effect and the interactions of the parameters were presented and the optimal conditions were obtained. This suggests the importance of local conditions including soil type on any future large-scale, in situ application. 相似文献
