Compound ITH33/IQM9.21 (ITH/IQM) belongs to a new family of l-glutamic acid derivatives with antioxidant and neuroprotective properties on in vitro and in vivo models of stroke. Because neuronal damage after brain ischemia is tightly linked to excess Ca2+ entry and neuronal Ca2+ overload, we have investigated whether compound ITH/IQM antagonises the elevations of the cytosolic Ca2+ concentrations ([Ca2+]c) and the ensuing exocytotic responses triggered by depolarisation of bovine chromaffin cells. In fluo-4-loaded cell populations, ITH/IQM reduced the K+-evoked [Ca2+]c transients with an IC50 of 5.31 μM. At 10 μM, the compound decreased the amplitude and area of the Ca2+ transient elicited by challenging single fura-2-loaded cells with high K+, by 40% and 80%, respectively. This concentration also caused a blockade of K+-induced catecholamine release at the single-cell level (78%) and cell populations (55%). These effects are likely due to blockade of the whole-cell inward Ca2+ currents (IC50 = 6.52 μM). At 10 μM, ITH/IQM also inhibited the Ca2+-dependent outward K+ current, leaving untouched the voltage-dependent component of IK. The inward Na+ current was unaffected. Inhibition of depolarisation-elicited Ca2+ entry, [Ca2+]c elevation and exocytosis could contribute to the neuroprotective effects of ITH/IQM in vulnerable neurons undergoing depolarisation during brain ischemia.  相似文献

Oxalate oxidase (OXO) utilizes oxalate to generate hydrogen peroxide, and thereby acts as a source of hydrogen peroxide. The present study was carried out to investigate whether apoplastic OXO modifies the metabolism of cell wall-bound ferulates in wheat seedlings. Histochemical staining of OXO showed that cell walls were strongly stained, indicating the presence of OXO activity in shoot walls. When native cell walls prepared from shoots were incubated with oxalate or hydrogen peroxide, the levels of ester-linked diferulic acid (DFA) isomers were significantly increased. On the other hand, the level of ester-linked ferulic acid (FA) was substantially decreased. The decrease in FA level was accounted neither by the increases in DFA levels nor by the release of FA from cell walls during the incubation. After the extraction of ester-linked ferulates, considerable ultraviolet absorption remained in the hemicellulosic and cellulose fractions, which was increased by the treatment with oxalate or hydrogen peroxide. Therefore, a part of FA esters may form tight linkages within cell wall architecture. These results suggest that cell wall OXO is capable of modifying the metabolism of ester-linked ferulates in cell walls of wheat shoots by promoting the peroxidase action via supply of hydrogen peroxide.  相似文献

Interaction between hyaluronic acid (HA) present at the surface of tubular epithelial cells and calcium oxalate monohydrate (COM) crystals is thought to play an important role in kidney stone formation. AFM-based force spectroscopy, where HA is covalently attached to AFM-probes, was used to quantify the interaction between HA and the surfaces of COM crystals. The work of adhesion of the HA-probe as well as the rupture force of single HA molecules were quantified in order to understand the molecular regulation of HA binding to COM crystals. Our results reveal that HA adsorbs to the crystal surface in physiological conditions. We also observed increased adhesion when the pH is lowered to a value that increases the risk of kidney stone formation. HA adhesion to the COM crystal surface can be suppressed by citrate, a physiological inhibitor of stone retention currently used in the treatment and prevention of kidney stone formation. Interestingly, we also observed preferential binding of HA onto the [100] face versus the [010] face, suggesting a major contribution of the [100] faces in the crystal retention process at the surface of tubular epithelial cells and the promotion of stone formation. Our results clearly establish a direct role for the glycosaminoglycan HA present at the surface of kidney tubular epithelium in the process of COM crystal retention.  相似文献

The X-ray powder diffractograms of racemic citalopram oxalate and (S)-citalopram oxalate are very similar, but the melting point of the racemate is higher than that of the pure enantiomer. The higher melting point indicates that the racemate is a racemic compound, rather than a conglomerate. The crystal structure of the enantiomer contains two molecules of (S)-citalopram in the asymmetric unit. The conformation of the two molecules is different but they approximate mirror images of each other if the aromatic groups are interchanged. The crystal structure of the racemate is essentially isostructural with that of the enantiomer, having almost the same cell parameters but containing a crystallographic inversion centre that is not retained in the enantiomer structure. The closely-comparable crystal structures permit solid solutions to be formed between racemic and enantiomeric citalopram oxalate. Phase diagrams of the (R)-citalopram and (S)-citalopram oxalate system are constructed, and they show that solid solutions are formed at all ratios of the two enantiomers.  相似文献

Plants growing under zinc excess store it in leaves, the organs in charge with carbon assimilation. Therefore, leaf modifications can help to understand the plant responses to environmental stresses. In this study, we tested the hypothesis that high zinc concentration can affect leaf anatomy and physiology of Populus alba cv. Villafranca, a tree species with interesting perspectives of application in phytoremediation.Plants were supplemented or not with 300 mg zinc per kg of soil and grown for six months. Green and macroscopically symptom-less leaves from poplars of each treatment were analyzed by light and electron microscopy (TEM, SEM-EDS) and by Raman spectroscopy. LHCII and D1-D2 protein expression were considered to evaluate PSII functioning. Zinc was histochemically detected in the leaves.Zinc strongly altered leaf morphology and ultrastructure, and negatively affected PSII activity decreasing D1 and D2, but not LHCII, protein expression. Zinc was mostly detected in the cell walls of the xylem and of the parenchyma cells surrounding the bundles of zinc treated plants, but not in the controls. A significant increase in the number of calcium oxalate crystals was observed in the leaves of zinc treated plants, but the latter metal was not incorporated in crystals, suggesting an increase of free calcium following zinc accumulation in the cell walls.  相似文献

Sclerotinia sclerotiorum D-E7 was studied to determine the impact of nutritional supplements and monosaccharides on growth, oxalate accumulation, and culture pH in broth media (initial pH c. 5). Cultures with 0.1% nutritional supplement (tryptone, yeast extract, or soytone) yielded minimal growth, 2-3 mM oxalate, and a final culture pH of 4.2-4.8. In contrast, cultures with 0.1% nutritional supplement and 25 mM glucose yielded significant growth, minimal oxalate (<1 mM), and a final culture pH of 2.8-3.7. Similar trends were observed when glucose in 0.1% soytone cultures was replaced with 25 mM d-mannose, l-arabinose, or d-xylose. With 1% soytone-25 mM glucose cultures, growth and oxalate accumulation ( approximately 21 mM) occurred with little change in initial pH. This was not the case with 1% soytone-250 mM glucose cultures; increased glucose levels resulted in a decrease in oxalate accumulation ( approximately 7 mM) and in final culture pH (3.4). Time-course studies with these cultures revealed that oxalate accumulation was suppressed during growth when the culture pH dropped to <4. Overall, these results indicate that (1) the decrease in external pH (i.e. acidification) was independent of oxalate accumulation and (2) acidification coupled to glucose-dependent growth regulated oxalate accumulation by Sclerotinia sclerotiorum.  相似文献

【背景】草酸钙结石是一种临床常见且易复发的疾病,由于结石质地坚硬,只能通过外科手术的方法治疗,给患者带了很大的痛苦。已有研究证实,肠道菌群可影响草酸钙结石的形成,降低草酸钙结石的发病率。【目的】探究植物乳杆菌对小鼠草酸钙结石的干预效果。【方法】体外实验:在MRS培养基中加入0.02 mol/L草酸钠,制备菌株筛选培养基(MRS-OX)。接种200μL的3.48×1012CFU/L植物乳杆菌悬液至MRS-OX制备含菌培养基(B+MRS-OX)。将等体积MRS-OX和B+MRS-OX于37°C恒温培养2 d,测剩余草酸浓度。体内实验:以10周龄雄性昆明小鼠为实验动物,随机分为对照组、植物乳杆菌组、结石组和植物乳杆菌干预组,每组5只小鼠。通过乙醛酸诱导小鼠建立草酸钙结石模型,并给予200μL的3.48×1012CFU/L植物乳杆菌进行干预治疗以观察其预防小鼠草酸钙结石的效果。实验结束后,绘制各组小鼠平均体重变化趋势图并计算小鼠肾脏脏器指数,检测每只小鼠血液学指标和氧化应激指标总超氧化物歧化酶(superoxide dismutase,SOD)和丙二...  相似文献

Hyperoxaluria‐induced calcium oxalate (CaOx) deposition is the key factor in kidney stone formation, for which adipose‐derived stromal cells (ADSCs) have been used as a therapeutic treatment. Studies revealed that miR‐20b‐3p is down‐regulated in hypercalciuric stone‐forming rat kidney. To investigate whether ADSC‐derived miR‐20b‐3p‐enriched exosomes protect against kidney stones, an ethylene glycol (EG)‐induced hyperoxaluria rat model and an in vitro model of oxalate‐induced NRK‐52E cells were established to explore the protective mechanism of miR‐20b‐3p. The results showed that miR‐20b‐3p levels were decreased following hyperoxaluria in the urine of patients and in kidney tissues from animal models. Furthermore, treatment with miR‐20b‐3p‐enriched exosomes from ADSCs protected EG‐induced hyperoxaluria rats, and cell experiments confirmed that co‐culture with miR‐20b‐3p‐enriched exosomes alleviated oxalate‐induced cell autophagy and the inflammatory response by inhibiting ATG7 and TLR4. In conclusion, ADSC‐derived miR‐20b‐3p‐enriched exosomes protected against kidney stones by suppressing autophagy and inflammatory responses.  相似文献

Flowers commonly face the dilemma of needing to be attractive to pollinators but unattractive to nectar robbers or florivores. When the pollinator has chewing mouthparts, there is also a considerable risk of the pollinator consuming the flowers. Scant field records indicate that the florivorous beetle Erioscelis emarginata (Mannerheim) (Coleoptera: Scarabaeidae: Cyclocephalini), which pollinates Philodendron spp., feeds selectively on the sterile staminate flowers. Here, the hypothesis was tested that E. emarginata prefers sterile staminate flowers of Philodendron bipinnatifidum Schott and Philodendron melinonii Brongn. ex Regel (Araceae) over fertile flowers. This study also examined whether such a preference exists regardless of the proportion of fertile vs. sterile flowers in each Philodendron species. Analysis of nutritional and defensive compounds plus scanning electron microscopy on each flower type were performed. The feeding preference of florivores for either P. bipinnatifidum or P. melinonii was also examined. In both species, sterile flowers were significantly more consumed than fertile ones. The sterile zone of P. bipinnatifidum was significantly larger than that of P. melinonii, and sterile staminate flowers of both plants were consumed at similar rates. Calcium oxalate was markedly low in sterile flowers, which also presented smaller papillae than the other flowers. This study presents evidence that the balance between pollination and florivory has most likely evolved through a strong feeding preference for less‐defended sterile flowers regardless of the size of the sterile zone.  相似文献
