Protein stability is usually characterized calorimetrically by a melting temperature and related thermodynamic parameters. Despite its importance, the microscopic origin of the melting transition and the relationship between thermodynamic stability and dynamics remains a mystery. Here, NMR relaxation parameters were acquired for backbone 15NH groups of the 56 residue immunoglobulin-binding domain of streptococcal protein G over a pre-denaturation temperature range of 5-50 degrees C. Relaxation data were analyzed using three methods: the standard three-Lorentzian model free approach; the F(omega)=2omegaJ(omega) spectral density approach that yields motional correlation time distributions, and a new approach that determines frequency-dependent order parameters. Regardless of the method of analysis, the temperature dependence of internal motional correlation times and order parameters is essentially the same. Nanosecond time-scale internal motions are found for all NHs in the protein, and their temperature dependence yields activation energies ranging up to about 33kJ/mol residue. NH motional barrier heights are structurally correlated, with the largest energy barriers being found for residues in the most "rigid" segments of the fold: beta-strands 1 and 4 and the alpha-helix. Trends in this landscape also parallel the free energy of folding-unfolding derived from hydrogen-deuterium (H-D) exchange measurements, indicating that the energetics for internal motions occurring on the nanosecond time-scale mirror those occurring on the much slower time-scale of H-D exchange. Residual heat capacities, derived from the temperature dependence of order parameters, range from near zero to near 100J/mol K residue and correlate with this energy landscape. These results provide a unique picture of this protein's energy landscape and a relationship between thermodynamic stability and dynamics that suggests thermosensitive regions in the fold that could initiate the melting process. 相似文献

Known high nitrogen utilization efficiency (NUE1, biomass per unit plant N) China lines of sorghum, China 17 and San Chi San, were compared with relatively low NUE1 U.S. lines, CK60 and Tx623, for both their physiological and biochemical adaptations to tolerate an imposed N stress in the greenhouse. Assimilation efficiency indices (ACi) were significantly greater for the China lines than the U.S. lines at both low and high soil nitrogen levels by about two-fold. Chlorophyll levels in leaves of high NUE1 lines were lower at both soil N treatments. Immunoblots of leaf extracts of sorghum subjected to N stress indicated reduced levels of both phosphoenolpyruvate carboxylase (PEPcase) and ribulose 1,5-bisphosphate carboxylase (Rubisco) while NADP-malic enzyme levels, in general, appear not to be affected. However, NUE1 China line, China 17, retained a significantly greater PEPcase activity than the less-NUE1 U.S. lines, and also the NUE1 China line San Chi San, when grown under N stress conditions. This suggests that PEPcase and enzymes associated with phosphoenolpyruvate synthesis, perhaps, are significant factors in maintaining relatively high photosynthesis under N stress. Carbon isotope ratios of leaves from sorghum genotypes, as indicated by 13C values, became less negative when sorghum plants were grown under N stress, but a genotypic variation either at a low or high N was not observed. 相似文献

wild life management. Estimation of maximum harvesting effort has a great impact on the economics of fisheries and other bio-resources.
The present paper deals with the problem of a bioeconomic fishery model under environmental variability. A technique for finding
the maximum harvesting effort in fluctuating environment has been developed in a two-species competitive system, which shows
that under realistic environmental variability the maximum harvesting effort is less than what is estimated in the deterministic
model. This method also enables us to find out the safe regions in the parametric space for which the chance of extinction
of the species is minimized. A real life fishery problem has been considered to obtain the inaccessible parameters of the

Identification of lipophilic flavones and flavonols by comparative HPLC,TLC and UV spectral analysis
The identification of lipophilic flavones and flavonols using a combination of high performance liquid chromatography, thin layer chromatography and UV spectral analysis is discussed. Data are provided for the flavones, apigenin, luteolin and tricetin and twelve of their methyl ethers, 8-hydroxyluteolin, 6-hydroxyluteolin and scutellarein and fourteen of their methyl ethers, and some 6,8-dihydroxyapigenin and 6,8-dihydroxyluteolin derivatives. Data for some forty two flavonols with extra 6- and/or 8-hydroxylation, mostly 6-hydroxykaempferol and quercetagetin derivatives, are also presented. The remaining compounds analysed include fourteen 5-deoxyflavones, four 5-methoxyflavones and five 5-deoxyflavonols plus further 5-hydroxylated flavones and flavonols without B-ring oxidation or with 2'-, 5'- or 6'-hydroxylation. 相似文献

Many ecological studies have used diversity indices to assess the impact of environmental disturbance. In particular, ground beetles have been advocated as a good group for assessing disturbance. Most studies on various organisms have used only one or two indices. For our study of the impact of tillage disturbance on carabid beetles in farm fields in southern Ontario, Canada, we used seven different diversity indices (richness, Shannon–Wiener, Berger–Parker, Q-statistic, Margalef, and evenness). Few studies have used deviation from diversity abundance models as a measure of disturbance; however, we use three that are applicable to our data (geometric, log-normal and log-series). The indices and models were used to test the null hypothesis that there is no change in diversity with increasing tillage disturbance, and that there is no difference in diversity with different crops or years. We were not able to reject the null hypothesis that there is any diversity difference among farms. We also found that there was no single diversity index or model that was better than any other at detecting disturbance. These results are supplemented by a meta-analysis of 45 published data sets for the same taxon but in different habitats. The meta-analysis supports the conclusions from our field research that diversity indices and models are not useful for detecting the possible effect of disturbance on assemblages of carabid beetles. 相似文献

Solid-state spectral editing techniques have been used by others to simplify 13C CPMAS spectra of small organic molecules, synthetic organic polymers, and coals. One approach utilizes experiments such as cross-polarization-with-polarization-inversion and cross-polarization-with-depolarization to generate subspectra. This work shows that this particular methodology is also applicable to natural-abundance 13C CPMAS NMR studies of high-molecular-weight biopolymers. The editing experiments are demonstrated first with model peptides and then with -elastin, a high-molecular-weight peptidyl preparation obtained from the elastic fibers in mammalian tissue. The latter has a predominance of small, nonpolar residues, which is evident in the crowded aliphatic region of typical 13C CPMAS spectra. Spectral editing is particularly useful for simplifying the aliphatic region of the NMR spectrum of this elastin preparation. 相似文献

Blood analyses for rare-earth element (REE)-high background regions in South Jiangxi show that the population averages of
many of the biochemical indices deviate markedly from normal values in the normal region. These deviations are thought to
be caused by prolonged intake of REE through food chains in view of that the toxicity of other harmful metals such as Pb and
Cd can be neglected because of their insignificant amounts in the environment. In comparison with the normal region, blood
biochemical indices abnormal in the REE-high regions are manifested as low total serum protein (TSP), albumin, β-globulin,
glutamic pyruvic transitanase, serium triglycerides, and immunoglobulin, but high cholesterol. These deviations may be related
to the REE concentration and composition of food chains, and are sex dependent. Certain blood indices (such as TSP) of different
age groups in the LREE-high region indicate that the influence of REE on males is a one-way irreversible process, whereas

In this paper, we investigate the abnormalities of electroencephalograph (EEG) signals in the Alzheimer’s disease (AD) by analyzing 16-scalp electrodes EEG signals and make a comparison with the normal controls. The power spectral density (PSD) which represents the power distribution of EEG series in the frequency domain is used to evaluate the abnormalities of AD brain. Spectrum analysis based on autoregressive Burg method shows that the relative PSD of AD group is increased in the theta frequency band while significantly reduced in the alpha2 frequency bands, particularly in parietal, temporal, and occipital areas. Furthermore, the coherence of two EEG series among different electrodes is analyzed in the alpha2 frequency band. It is demonstrated that the pair-wise coherence between different brain areas in AD group are remarkably decreased. Interestingly, this decrease of pair-wise electrodes is much more significant in inter-hemispheric areas than that in intra-hemispheric areas. Moreover, the linear cortico-cortical functional connectivity can be extracted based on coherence matrix, from which it is shown that the functional connections are obviously decreased, the same variation trend as relative PSD. In addition, we combine both features of the relative PSD and the normalized degree of functional network to discriminate AD patients from the normal controls by applying a support vector machine model in the alpha2 frequency band. It is indicated that the two groups can be clearly classified by the combined feature. Importantly, the accuracy of the classification is higher than that of any one feature. The obtained results show that analysis of PSD and coherence-based functional network can be taken as a potential comprehensive measure to distinguish AD patients from the normal, which may benefit our understanding of the disease. 相似文献
