Grass cell walls have hydroxycinnamic acids attached to arabinosyl residues of arabinoxylan (AX), and certain BAHD acyltransferases are involved in their addition. In this study, we characterized one of these BAHD genes in the cell wall of the model grass Setaria viridis. RNAi silenced lines of S. viridis (SvBAHD05) presented a decrease of up to 42% of ester-linked p-coumarate (pCA) and 50% of pCA-arabinofuranosyl, across three generations. Biomass from SvBAHD05 silenced plants exhibited up to 32% increase in biomass saccharification after acid pre-treatment, with no change in total lignin. Molecular dynamics simulations suggested that SvBAHD05 is a p-coumaroyl coenzyme A transferase (PAT) mainly involved in the addition of pCA to the arabinofuranosyl residues of AX in Setaria. Thus, our results provide evidence of p-coumaroylation of AX promoted by SvBAHD05 acyltransferase in the cell wall of the model grass S. viridis. Furthermore, SvBAHD05 is a promising biotechnological target to engineer crops for improved biomass digestibility for biofuels, biorefineries and animal feeding. 相似文献

We tested whether the host species identity in grass-Epichloë symbioses affected soil chemical and microbial properties. We grew endophyte infected (E+) and endophyte free (E−) Lolium perenne, Hordeum brevisubulatum and Achnatherum inebrians for 18 months in field plots. In E+ soil of all three grasses, available phosphorus was lower whereas total soil nitrogen was higher. Endophyte effects on soil pH, microbial biomass nitrogen, total carbon and organic carbon as well as bacteria and fungi abundance were host species dependent. Ammonia oxidizing bacteria abundance was higher in E+ soils for all species. Bacterial community composition of E+ and E− soils were different only for Lolium perenne with soil pH being the key factor. Fungal community composition of E+ and E− soils was not different for the three grasses. This study confirmed that the effects of foliar Epichloë infection on belowground properties depended on host species identity. 相似文献

The vegetation and the associated germinable seedbank of a range of heath and grass heath communities in Breckland, eastern England, were compared. The composition of vegetation and seedbank were similar for heather-dominated communities due to the predominance of Calluna vulgaris . Recent rotovation of previously heather-dominated communities invaded by Deschampsia flexuosa reduced the number of emerging C. vulgaris seedlings by more than 80%. The composition of seedbanks of calcicolous grasslands differed considerably from the vegetation, with a significant contribution from many ruderal species not present in the vegetation and an absence of important species such as Pilosella officinarum. Acidic grasslands were intermediate between calcicolous grasslands and heather-dominated heathlands, with seedbanks characterized by high densities of Rumex acetosella and a number of ruderal species that were absent or rare in the vegetation. The composition of the seedbank of more mesotrophic grasslands suggests that the imposition of some form of disturbance may recreate more open grasslands typical of Breckland. 相似文献

The composition of the essential oil of Khavi grass, Cymbopogon jawarancusa, was investigated by glass capillary gas chromatography in combination with mass spectrometry. Sixty-four compounds were identified, 55 of which are reported for the first time. Alternative inexpensive complex nitrogen- and phosphate sources from agriculture, green grass juice (GGJ) and silage juice (SJ), were added to cultivation medium in order to investigate their impact on growth of the well-known polyhydroxyalkanoate (PHA) accumulating strain Wautersia eutropha. The influence of these additives was directly compared with cultivations on defined minimal mineral medium (M) as well as on the same medium supplemented with more expensive complex additives: corn steep liquor (CSL) and casamino acids (CA). It turned out that the supplementation with most complex additives results in shortening of lag-phases of bacterial growth and in higher end-concentrations of residual biomass compared with M-medium. Finally, higher volumetric productivities for poly(3-hydroxybutyrate) (3-PHB) were achieved. The effect of the inexpensive additive SJ on volumetric productivity was similar to the result for the expensive CA (0.653 vs. 0.619 g L-1 h-1). The same was found for the biomass concentration (7.00 vs. 7.44 g L-1 respectively). Together with an economic appraisal presented in this study, the results suggest it is possible to make the sustainable process of microbial PHA-production more economically feasible. A survey of the thermal characteristics and molecular mass properties of the isolated polymers completes this work. 相似文献

The role of sewage sludge as an immobilising agent in the phytostabilization of metal-contaminated soil was evaluated using five grass species viz., Dactylis glomerata L., Festuca arundinacea Schreb., F. rubra L., Lolium perenne L., L. westerwoldicum L. The function of metal immobilization was investigated by monitoring pH, Eh and Cd, Pb, and Zn levels in column experiment over a period of 5-months. Grasses grown on sewage sludge-amendments produced high biomass in comparison to controls. A significant reduction in metal uptake by plants was also observed as a result of sewage sludge application, which was attributed to decreased bioavailability through soil stabilisation. We have observed that the sludge amendment decreased metal bioavailability and concentrations in soil at a depth of 25 cm, in contrast to untreated columns, where metal concentrations in the soil solution were very high. 相似文献
