【目的】为了分析水产品腐败菌群体感应的新型信号分子二酮哌嗪(DKPs)化合物,建立一种简便、灵敏的气相色谱-质谱(GC-MS)定量检测方法。【方法】通过优化气相色谱和质谱条件、培养基和提取溶剂建立定量检测方法,确定Cyclo-(L-Pro-L-Gly)、Cyclo-(L-Pro-L-Leu)、Cyclo-(L-Leu-L-Leu)和Cyclo-(L-Pro-L-Phe)4种DKPs标准品的特征离子,并检测水产品腐败菌荧光假单胞菌(Pseudomonas fluorescens)和波罗的海希瓦氏菌(Shewanella baltica)中的DKPs。【结果】4种DKPs化合物在1-200 mg/L范围内线性良好,检测限为0.06、0.10、0.06和0.04 mg/L,定量限为0.16、0.18、0.14和0.12 mg/L,回收率为51.8%-88.5%,标准偏差为1.4%-8.3%。以LB培养基为细菌培养基,氯仿作为萃取溶剂,检测的DKPs含量较高。两种水产品腐败菌都检测到DKPs活性,主要种类为Cyclo-(L-Pro-L-Leu)和Cyclo-(L-Pro-L-Phe)。随着两种细菌的生长,培养上清中DKPs含量显著增加,在12 h达到最高。【结论】建立了检测细菌DKPs的GC-MS定量方法,具有较高精密度、准确度,能够准确定量分析4种DKPs的含量。为探究水产品特定腐败菌DKPs的调控机制奠定基础。 相似文献

Increased contamination of the environment with toxic pollutants has paved the way for efficient strategies which can be implemented for environmental restoration. The major problem with conventional methods used for cleaning of pollutants is inefficiency and high economic costs. Bioremediation is a growing technology having advanced potential of cleaning pollutants. Biofilm formed by various micro-organisms potentially provide a suitable microenvironment for efficient bioremediation processes. High cell density and stress resistance properties of the biofilm environment provide opportunities for efficient metabolism of number of hydrophobic and toxic compounds. Bacterial biofilm formation is often regulated by quorum sensing (QS) which is a population density-based cell–cell communication process via signaling molecules. Numerous signaling molecules such as acyl homoserine lactones, peptides, autoinducer-2, diffusion signaling factors, and α-hydroxyketones have been studied in bacteria. Genetic alteration of QS machinery can be useful to modulate vital characters valuable for environmental applications such as biofilm formation, biosurfactant production, exopolysaccharide synthesis, horizontal gene transfer, catabolic gene expression, motility, and chemotaxis. These qualities are imperative for bacteria during degradation or detoxification of any pollutant. QS signals can be used for the fabrication of engineered biofilms with enhanced degradation kinetics. Xanthomonas albilineans, the causal agent of sugarcane leaf scald, is a bacterial plant pathogen that is mainly spread by infected cuttings and contaminated harvesting tools. However, some strains of this pathogen are known to be spread by aerial means and are able to colonize the phyllosphere of sugarcane before entering the host plant and causing disease. The objective of this study was to identify the molecular factors involved in the survival or growth of X. albilineans on sugarcane leaves. We developed a bioassay to test for the attachment of X. albilineans on sugarcane leaves using tissue‐cultured plantlets grown in vitro. Six mutants of strain XaFL07‐1 affected in surface polysaccharide production completely lost their capacity to survive on the sugarcane leaf surface. These mutants produced more biofilm in vitro and accumulated more cellular poly‐β‐hydroxybutyrate than the wild‐type strain. A mutant affected in the production of small molecules (including potential biosurfactants) synthesized by non‐ribosomal peptide synthetases (NRPSs) attached to the sugarcane leaves as well as the wild‐type strain. Surprisingly, the attachment of bacteria on sugarcane leaves varied among mutants of the rpf gene cluster involved in bacterial quorum sensing. Therefore, quorum sensing may affect polysaccharide production, or both polysaccharides and quorum sensing may be involved in the survival or growth of X. albilineans on sugarcane leaves. 相似文献

Soil salinity is recognized worldwide as a major threat to agriculture, particularly in arid and semi-arid regions. Producers and decision makers need updated and accurate maps of salinity in agronomically and environmentally relevant ranges (i.e., <20 dS m−1, when salinity is measured as electrical conductivity of the saturation extract, ECe). State-of-the-art approaches for creating accurate ECe maps beyond field scale (i.e., 1 km2) include: (i) Analysis Of Covariance (ANOCOVA) of near-ground measurements of apparent soil electrical conductivity (ECa) and (ii) regression modeling of multi-year remote sensing canopy reflectance and other co-variates (e.g., crop type, annual rainfall). This study presents a comparison of the two approaches to establish their viability and utility. The approaches were tested using 22 fields (total 542 ha) located in California’s western San Joaquin Valley. In 2013 ECa-directed soil sampling resulted in the collection of 267 soil samples across the 22 fields, which were analyzed for ECe, ranging from 0 to 38.6 dS m−1. The ANOCOVA ECa-ECe model returned a coefficient of determination (R2) of 0.87 and root mean square prediction error (RMSPE) of 3.05 dS m−1. For the remote sensing approach seven years (2007–2013) of Landsat 7 reflectance were considered. The remote sensing salinity model had R2 = 0.73 and RMSPE = 3.63 dS m−1. The robustness of the models was tested with a leave-one-field-out (lofo) cross-validation to assure maximum independence between training and validation datasets. For the ANOCOVA model, lofo cross-validation provided a range of scenarios in terms of RMSPE. The worst, median, and best fit scenarios provided global cross-validation R2 of 0.52, 0.80, and 0.81, respectively. The lofo cross-validation for the remote sensing approach returned a R2 of 0.65. The ANOCOVA approach performs particularly well at ECe values <10 dS m−1, but requires extensive field work. Field work is reduced considerably with the remote sensing approach, but due to the larger errors at low ECe values, the methodology is less suitable for crop selection, and other practices that require accurate knowledge of salinity variation within a field, making it more useful for assessing trends in salinity across a regional scale. The two models proved to be viable solutions at large spatial scales, with the ANOCOVA approach more appropriate for multiple-field to landscape scales (1–10 km2) and the remote sensing approach best for landscape to regional scales (>10 km2). 相似文献

Upland moors in the UK have been managed for centuries using rotational prescribed-burning, but in recent years there has been contentious debate over its continuing use due to varying effects on moorland ecosystem services. Prescribed-burning should only be carried out using good-practice codes, which include restrictions on the size, location and frequency of burns. Good burning practice is an indicator of management standards and habitat condition in moorland landscapes. However, there has been little attempt to assess management performance with respect to these restrictions. We investigated prescribed-burning on a case-study estate (Howden Moor) in the Peak District National Park from 1988 to 2009 using management maps and aerial photography. The annual area burned (0.9%) was far below recommendations (10%) and patches were in keeping with the target sizes specified (mean ± se: 2370 ± 70 m2). The risk of a large or escaped fire was very low, with less than 1% of fires greater than 15,000 m2. However, only 28.9% of the total burnable area was burned, leaving the rest unmanaged and accumulating fuel. Future guidelines might recommend the application of prescribed-burning across the range of Calluna vulgaris growth phases, to reduce fuel load and promote biodiversity at the landscape scale. We show that vegetation mapping and aerial photography are an effective method for monitoring prescribed-burning practice on moorlands. The information derived from such monitoring studies should lead to greater confidence in the standard of prescribed-burning and adherence to good-practice guidelines and requirements imposed by statutory authorities. 相似文献

Due to deforestation, intact tropical forest areas are increasingly transformed into a mixture of remaining forest patches and human modified areas. These forest fragments suffer from edge effects, which cause changes in ecological and ecosystem processes, undermining habitat quality and the offer of ecosystem services. Even though detailed and long term studies were developed on the topic of edge effects at local scale, understanding edge effect characteristics in fragmented forests on larger scales and finding indicators for its impact is crucial for predicting habitat loss and developing management options. Here we evaluate the spatial and temporal dimensions of edge effects in large areas using remote sensing. First we executed a neighborhood pixel analysis in 11 LANDSAT Tree Cover (LTC) scenes (180 × 185 km each, 8 in the tropics and 3 in temperate forested areas) using tree cover as an indicator of habitat quality and in relation to edge distance. Second, we executed a temporal analysis of LTC in a smaller area in the Brazilian Amazon forest where one larger forest fragment (25,890 ha) became completely fragmented in 5 years. Our results show that for all 11 scenes pixel neighborhood variation of LTC is much higher in the vicinity of forest edges, becoming lower towards the forest interior. This analysis suggests a maximum distance for edge effects and can indicate the location of unaffected core areas. However, LTC patterns in relation to fragment edge distance vary according to the analyzed region, and maximum edge distance may differ according to local conditions. Our temporal analysis illustrates the change in tree cover patterns after 5 years of fragmentation, becoming on average lower close to the edge (between 50 and 100 m). Although it is still unclear which are the main causes of LTC edge variability within and between regions, LANDSAT Tree Cover could be used as an accessible and efficient discriminator of edge and interior forest habitats in fragmented landscapes, and become invaluable for deriving qualitative spatial and temporal information of ecological and ecosystem processes. 相似文献

Aims: Shrub-encroached grassland has become an important vegetation type in China's arid and semi-arid region. Our study objective is to explore the spectral features of shrub and grass communities, as well as their empirical relationships with shrub coverage. The quantitative estimation of shrub cover based on medium-resolution Landsat satellite imagery provides the practical basis for long term retrieval of large areas of shrub expansion in the grassland region. Methods: Linear models and Multiple Endmember Spectral Analysis Model (MESMA) based on medium resolution Landsat satellite imagery were developed to quantify the shrub coverage in a shrub-encroached grassland region in Xianghuang Banner, Nei Mongol using the spectral features and their seasonal differences between the shrub and grass communities. Important findings: Compared to Leymus chinensis and Stipa krylovii dominated grass communities, Caragana microphylla community had a higher normalized difference vegetation index (NDVI), modified red edge normalized difference vegetation index (mNDVI705), and red edge slope. The red edge position of C. microphylla community shifted to longer wavelengths. The average and the maximum shrub coverage was 13% and 25%, respectively, in the shrub-encroached grassland based on both models. The correlation coefficient of determination (R2) and root mean square error (RMSE) of the linear model was 0.31 and 0.05, respectively. We found that the linear model based on seasonal differences of shrub and grass community was more suitable for retrieving shrub coverage in the study area from medium resolution imagery than the MESMA model that is based on mid-summer images. 相似文献

The research in the field of design and synthesis of unnatural amino acids is growing at a fast space for the increasing demand of proteins of potential therapeutics and many other diversified novel functional applications. Thus, we report herein the design and synthesis of microenvironment sensitive fluorescent triazolyl unnatural amino acids (UNAA) decorated with donor and/or acceptor aromatic chromophores via click chemistry. The synthesized fluorescent amino acids show interesting solvatochromic characteristic and/or intramolecular charge transfer (ICT) feature as is revealed from the UV–visible, fluorescence photophysical properties and DFT/TDDFT calculation. HOMO–LUMO distribution shows that the emissive states of some of the amino acids are characterized with more significant electron redistribution between the triazolyl moiety and the aromatic chromophores linked to it leading to modulated emission property. A pair of donor–acceptor amino acid shows interesting photophysical interaction property indicating a FRET quenching event. Furthermore, one of the amino acid, triazolyl-perylene amino acid, has been exploited for studying interaction with BSA and found that it is able to sense BSA with an enhancement of fluorescence intensity. Finally, we incorporated a pair of donor/acceptor amino acids into a Leu-enkephalin analogue pentapeptide which was found to adopt predominantly type II β-turn conformation. We envisage that our investigation is of importance for the development of new fluorescent donor–acceptor unnatural amino acids a pair of which can be exploited for generating fluorescent peptidomimetic probe of interesting photophysical property for applications in studying peptide–protein interaction. 相似文献
