The minimum sampling areas (MSAs) for the shrub communities in the arid valley in the upper reach of the Minjiang River, China, were studied by fitting community species-area relationships using 3 types of equations. The MSAs were determined at the proportional factor (ρ) 0.6, 0.7, 0.8 and 0.9. The proportional factors represent the proportion of the number of species within a sampling plot in the total number of species. The MSAs of the shrub communities at different elevations and on different slope faces for ρ = 0.6, 0.7 and 0.8 were all around 100 m2. Hence, the MSAs could be set to be 100 m2 (10 m × 10 m) at 60%–80% precision levels. For ρ = 0.9, that is, for a 90% precision level, the MSAs were less than 200 m2 (10 m × 20 m). The MSAs and species richness increased gradually with the rising elevation. At the elevation below 2000 m, the MSAs and species richness on the north-facing slope were larger than those on the south-facing slope. However, at the elevation around 2200 m, there was no difference amongst different facing slopes. For the shrub communities in the arid valley in the upper reach of the Minjiang River, the species-area curves by fitting the first two equations are better than that by fitting the third equation.

A small library of antiplasmodial methoxy-thiazinoquinones, rationally designed on the model of the previously identified hit 1, has been prepared by a simple and inexpensive procedure. The synthetic derivatives have been subjected to in vitro pharmacological screening, including antiplasmodial and toxicity assays. These studies afforded a new lead candidate, compound 9, endowed with higher antiplasmodial potency compared to 1, a good selectivity index when tested against a panel of mammalian cells, no toxicity against RBCs, a synergistic antiplasmodial action in combination with dihydroartemisinin, and a promising inhibitory activity on stage V gametocyte growth. Computational studies provided useful insights into the structural requirements needed for the antiplasmodial activity of thiazinoquinone compounds and on their putative mechanism of action.

The marine dinoflagellate Gyrodinium resplendens Hulburt is a mixotroph. It possesses chloroplasts and is photosynthetic, and it also feeds phagotrophically on another dinoflagellate, Prorocentrum minimum (Pavillard) Schiller. The species could be cultivated only in food-replete cultures. When kept in cultures without food, cellular chl a content and photosynthetic activity of G. resplendens decreased and growth ceased after a few days. In food-replete cultures, G. resplendens could grow strictly heterotrophically in darkness, but growth rate was then three times lower than in food-replete cultures kept in light. It is suggested that the main importance of phagotrophy is to acquire a growth factor essential to photosynthetic growth. The addition of soil extract or amino acids to the growth medium induced enhanced photosynthetic growth of the species even without the presence of particulate food, but only for approximately 2 weeks. Long-term starvation of G. resplendens led to loss of the ability to feed, and therefore starved cells eventually reached a point of no return where neither photosynthesis nor phagotrophy could sustain further growth. Light microscopical observations on G. resplendens revealed new morphological and behavioral details of the species.

Assessment of arbovirus vector infection rates using variable size pooling
Pool testing of vector samples for arboviruses is widely used in surveillance programmes. The proportion of infected mosquitoes (Diptera: Culicidae) is often estimated from the minimum infection rate (MIR), based on the assumption of only one infected mosquito per positive pool. This assumption becomes problematic when pool size is large and/or infection rate is high. By relaxing this constraint, maximum likelihood estimation (MLE) is more useful for a wide range of infection levels that may be encountered in the field. We demonstrate the difference between these two estimation approaches using West Nile virus (WNV) surveillance data from vectors collected by gravid traps in Chicago during 2002. MLE of infection rates of Culex mosquitoes was as high as 60 per 1000 at the peak of transmission in August, whereas MIR was less than 30 per 1000. More importantly, we demonstrate roles of various pooling strategies for better estimation of infection rates based on simulation studies with hypothetical mosquito samples of 18 pools. Variable size pooling (with a serial pool sizes of 5, 10, 20, 30, 40 and 50 individuals) performed consistently better than a constant size pooling of 50 individuals. We conclude that variable pool size coupled with MLE is critical for accurate estimates of mosquito infection rates in WNV epidemic seasons.

Vivipary with precocious seedlings in mangrove plants was thought to be a hindrance to long-range dispersal. To examine the extent of seedling dispersal across oceans, we investigated the phylogeny and genetic structure among East Asiatic populations of Kandelia candel based on organelle DNAs. In total, three, 28 and seven haplotypes of the chloroplast DNA (cpDNA) atpB-rbcL spacer, cpDNA trnL-trnF spacer, and mitochondrial DNA (mtDNA) internal transcribed spacer (ITS) were identified, respectively, from 202 individuals. Three data sets suggested consistent phylogenies recovering two differentiated lineages corresponding to geographical regions, i.e. northern South-China-Sea + East-China-Sea region and southern South-China-Sea region (Sarawak). Phylogenetically, the Sarawak population was closely related to the Ranong population of western Peninsula Malaysia instead of other South-China-Sea populations, indicating its possible origin from the Indian Ocean Rim. No geographical subdivision was detected within the northern geographical region. An analysis of molecular variance (AMOVA) revealed low levels of genetic differentiation between and within mainland and island populations (phiCT = 0.015, phiSC = 0.037), indicating conspicuous long-distance seedling dispersal across oceans. Significant linkage disequilibrium excluded the possibility of recurrent homoplasious mutations as the major force causing phylogenetic discrepancy between mtDNA and the trnL-trnF spacer within the northern region. Instead, relative ages of alleles contributed to non-random chlorotype-mitotype associations and tree inconsistency. Widespread distribution and random associations (chi2 = 0.822, P = 0.189) of eight hypothetical ancestral cytotypes indicated the panmixis of populations of the northern geographical region as a whole. In contrast, rare and recently evolved alleles were restricted to marginal populations, revealing some preferential directional migration.

Humans and other animals can temporarily store mechanical energy in elastic oscillations, f(el), of body parts and in pendulum oscillations, f(p) = const sq.rt (g/L), of legs, length L, or other appendages, and thereby reduce the energy consumption of locomotion. However, energy saving only occurs if these oscillations are tuned to the leg propagation frequency f. It has long been known that f is tuned to the pendulum frequency of the free-swinging leg of walkers. During running the leg frequency increases to some new value f = f(r). We propose that in order to maintain resonance the animal, mass M, actively increases its leg pendulum frequency to the new value f(p,r) =const sq.rt (a(y)/L)=f(r), by giving its hips a vertical acceleration a(y)= F(y)/M. The pendulum frequency is increased if the impact force F(y) of the stance foot is larger than Mg, explaining the observation by Alexander and Bennet-Clark (1976) that F(v) becomes larger than Mg when animals start to run. Our model predictions of the running velocity U(r) as function of L, F(v), are in agreement with measurements of these quantities (Farley et al. 1993). The leg's longitudinal elastic oscillation frequency scales as f(el) = const sq.rt (k/M). Experiments by Ferris et al., (1998) show that runners adjust their leg's stiffness, k, when running on surfaces of different elasticity so that the total stiffness k remains constant. Our analysis of their data suggests that the longitudinal oscillations of the stance leg are indeed kept in tune with the running frequency. Therefore we conclude that humans, and by extension all animals, maintain resonance during running. Our model also predicts the Froude number of walking-running transitions, Fr = U(2)/gL approximately 0.5 in good agreement with measurements.

We present fast new algorithms for evaluating trees with respectto least squares and minimum evolution (ME), the most commonlyused criteria for inferring phylogenetic trees from distancedata. The new algorithms include an optimal O(N2) time algorithmfor calculating the edge (branch or internode) lengths on atree according to ordinary or unweighted least squares (OLS);an O(N3) time algorithm for edge lengths under weighted leastsquares (WLS) including the Fitch-Margoliash method; and anoptimal O(N4) time algorithm for generalized least-squares (GLS)edge lengths (where N is the number of taxa in the tree). TheME criterion is based on the sum of edge lengths. Consequently, the edge lengths algorithms presented here lead directly toO(N2), O(N3), and O(N4) time algorithms for ME under OLS, WLS,and GLS, respectively. All of these algorithms are as fast asor faster than any of those previously published, and the algorithmsfor OLS and GLS are the fastest possible (with respect to orderof computational complexity). A major advantage of our new methodsis that they are as well adapted to multifurcating trees asthey are to binary trees. An optimal algorithm for determiningpath lengths from a tree with given edge lengths is also developed. This leads to an optimal O(N2) algorithm for OLS sums of squaresevaluation and corresponding O(N3) and O(N4) time algorithmsfor WLS and GLS sums of squares, respectively. The GLS algorithmis time-optimal if the covariance matrix is already inverted. The speed of each algorithm is assessed analytically—thespeed increases we calculate are confirmed by the dramatic speedincreases resulting from their implementation in PAUP* 4.0.The new algorithms enable far more extensive tree searches andstatistical evaluations (e.g., bootstrap, parametric bootstrap,or jackknife) in the same amount of time. Hopefully, the fastalgorithms for WLS and GLS will encourage the use of these criteriafor evaluating trees and their edge lengths (e.g., for approximatedivergence time estimates), since they should be more statisticallyefficient than OLS.

The terrestrial isopod species Armadillo tuberculatus Vogl, 1876 (Crustacea, Isopoda, Oniscidea) is a widely polymorphic species distributed in the south-central Aegean region (Greece) with a different morph on each island. Variation consists in coloration, size of cuticular tubercules, shape of telson and the shape of the male first pleopod exopodite (secondary sexual character of taxonomic importance). We studied the allometric growth of a cuticular tubercule in 17 populations (for both male and female individuals) and the shape variation of the first male pleopod exopodite in 10 populations using Elliptic Fourier Analysis, in order to test for patterns of intraspecific variation and possible relationships between morphs. In addition, Thin Plate Spline analysis was used for the calculation of the minimum bending energy between different exopodite shapes, which was then used for estimating the minimum spanning network (MSN) connecting them. The different allometric growth rates of the tubercule among island groups were significantly related to island latitude and climatic factors. On the other hand, the clustering of islands and the MSN based on male exopodite shape differences were not related to the palaeogeography of the Aegean region or to the present geographic distances of islands. These results are interpreted as evidence for non-adaptive radiation of the morphs.
