In 76 healthy persons (right-handed men and women), we recorded background EEG and event-related potentials from the C3 and C4 sites; tests were performed within the framework of an experimental situation requiring internal measuring-off of the time intervals. To limit the interval, the tested person had to push a button; he/she did not know the precise value of the interval, which was preset by the experimenter, and was informed only of the lower and upper limits of this interval, 17 to 23 sec. The person obtained information about the coincidence/noncoincidence of the measured-off and preset intervals via visual feedback; the respective signal was presented 2 sec after measuring-off had been completed. In the case where the intervals coincided with each other, the person should confirm this by pushing the button next time (confirming push). We characterized the parameters of the measured-off time interval by the following indices: (i) measuring-off efficacy (accuracy of fitting the preset interval), (ii) estimation tendency (measured-off interval/preset standard interval ratio), and (iii) coefficient of variation (CV) of the measured-off interval. Features of the subject's personality were estimated using Eysenck's (PEN) and Cattell's (16PF) questionnaires. We found correlations of the powers of the background EEG rhythms (beta1, beta2, and alpha/theta ratio) and characteristics of the measured-off time interval. In addition, we observed significant positive correlations between the estimation tendency and extraversion index and between CV of the interval and urge toward domination and protension indices. Negative correlations were observed between the measuring-off efficacy and protension (suspiciousness), between the estimation tendency and anxiety, and between the CV of the interval and age of the subjects. We support the conclusion that correlations between the patterns of EEG potentials, peculiarities of measuring-off of the time interval, and psychological features of the personality are to a noticeable extent mediated by the individual specificity of the neurodynamics.  相似文献

Callus was induced from hypocotyl and primary leaf explants of cumin (Cuminum cyminum L.) seedlings on a medium with 4 M 2,4-D alone or plus 2 or 4 M kinetin. An embryogenic callus developed within 2 weeks after transferring the callus to medium lacking plant growth regulators (PGR). The presence of kinetin in the callus induction medium with 2,4-D enhanced both the callus proliferation and the subsequent differentiation of the embryoids on the PGR-free medium. Plumules with or without simultaneously developed roots were observed 3–4 weeks after subculturing the embryogenic callus on medium containing 0.5 or 1.0 M kinetin. Subsequently, they were transferred onto half-strength medium supplemented with 1 M indole-3-butyric acid (IBA) and 2% polyethylene glycol (PEG, 6000) for root induction and/or proliferation, and in vitro hardening of the regenerated plants. The survival rate ex vitro was 70%. No plants developed from the embryogenic callus continuously incubated on medium lacking kinetin. We concluded that kinetin is crucial for plant regeneration from the induced embryoids of cumin.  相似文献

Somatic embryogenesis and whole plant regeneration were achieved in callus cultures derived from hypocotyl, cotyledon and leaf explants excised from seedlings of Gymnema sylvestre. Embryogenic callus was induced on Murashige and Skoog (MS) medium containing 2,4-D (0.5–5.0 M) +BA (0.5–2.0 M) and 2% (w/v) sucrose in 6–8 weeks of culture. Globular/heart stage embryos developed on induction medium. These embryos produced torpedo and cotyledon stage embryos upon sub-culturing on embryo maturation medium EM8 (medium containing MS salts, B5 vitamins, 0.5 M BA and 2% sucrose). Embryo germination and plantlet formation was achieved by sub-culturing mature embryos on fresh EM8 medium. The plantlets were acclimatized in the greenhouse.  相似文献

Yields and retention of dissolved inorganic nitrogen (DIN: NO3 + NH4 +) and nitrate concentrations in surface runoff are summarized for 28 high elevation watersheds in the Sierra Nevada of California and Rocky Mountains of Wyoming and Colorado. Catchments ranged in elevation from 2475 to 3603 m and from 15 to 1908 ha in area. Soil cover varied from 5% to nearly 97% of total catchment area. Runoff from these snow-dominated catchments ranged from 315 to 1265 mm per year. In the Sierra Nevada, annual volume-weighted mean (AVWM) nitrate concentrations ranged from 0.5 to 13 M (overall average 5.4 M), and peak concentrations measured during snowmelt ranged from 1.0 to 38 M. Nitrate levels in the Rocky Mountain watersheds were about twice those in the Sierra Nevada; average AVWM NO3 was 9.4 M and snowmelt peaks ranged from 15 to 50 M. Mean DIN loading to Rocky Mountain watersheds, 3.6 kg ha–1 yr–1, was double the average measured for Sierra Nevada watersheds, 1.8 kg ha–1 yr–1. DIN yield in the Sierra Nevada, 0.69 kg ha–1 yr–1, was about 60% that measured in the Rocky Mountains, 1.1 kg ha–1 yr–1. Net inorganic N retention in Sierra Nevada catchments was 1.2 kg ha–1 yr–1 and represented about 55% of annual DIN loading. DIN retention in the Rocky Mountain catchments was greater in absolute terms, 2.5 kg ha–1 yr–1, and as a percentage of DIN loading, 72%.A correlation analysis using DIN yield, DIN retention and surface water nitrate concentrations as dependent variables and eight environmental features (catchment elevation, slope, aspect, roughness, area, runoff, soil cover and DIN loading) as independent variables was conducted. For the Sierra Nevada, elevation and soil cover had significant (p > 0.1) Pearson product moment correlations with catchment DIN yield, AVWM and peak snowmelt nitrate concentrations and DIN retention rates. Log-linear regression models using soil cover as the independent variable explained 82% of the variation in catchment DIN retention, 92% of the variability in AVWM nitrate and 85% of snowmelt peak NO3 . In the Rocky Mountains, soil cover was significantly (p < 0.05) correlated with DIN yield, AVWM NO3 and DIN retention expressed as a percentage of DIN loading (%DIN retention). Catchment mean slope and terrain roughness were positively correlated with steam nitrate concentrations and negatively related to %DIN retention. About 91% of the variation in DIN yield and 79% of the variability in AVWM NO3 were explained by log-linear models based on soil cover. A log-linear regression based on soil cover explained 90% of the variation of %DIN retention in the Rocky Mountains.  相似文献

This paper describes rapid propagation of Sesbania drummondii using nodal explants isolated from seedlings and young plants. The nodal segments proliferated into multiple shoots on Murashige and Skoog's (MS) medium supplemented with 22.2 M benzyladenine. MS medium containing 2.2 and 4.5 M thidiazuron induced 5 – 6 shoots per stem node from 3-month-old plants. Nodal explants when cultured on MS medium containing combinations of benzyladenine (8.8 and 11.1 M) and indole-3-butyric acid (0.24 – 2.46 M) or indole-3-acetic acid (0.28 – 2.85 M) gave lesser number of shoots. Callus induced on cotyledonary explants when subcultured on 2.2 M thidiazuron containing medium resulted in its mass proliferation having numerous embryoid-like structures. Indole-3-butyric acid (0.24 – 2.46 M) was found suitable for root induction. In vitro regenerated plants were acclimatized in greenhouse conditions.  相似文献

An efficient in vitro plant regeneration system via hypocotyl segments of tetraploid Isatis indigotica Fort. was established. Murashige and Skoog's (MS) and Gamborg's (GB5) media were found to be superior to White medium for promoting shoot regeneration. The highest shoot regeneration (92 %) was achieved from hypocotyls cultured on MS medium containing 8.9 M benzyladenine (BA) and 2.7 M naphthaleneacetic acid (NAA), with an average of 4.2 shoots developed per explant. Plant regeneration was also improved when the explants were cultured in MS basal medium containing 3 % (m/v) sucrose and grown under a 12-h photoperiod. The developed shoots were well rooted in a half-strength MS medium supplemented with 0.5 M indole-3-butyric acid (IBA) and were morphologically normal after transfer to soil.  相似文献

Summary A micropropagation method for Orthosiphon stamineus, using stem nodal segments, has been developed. The highest number of regenerated shoots was obtained on Murashige and Skoog (MS) medium supplemented with 6.7 μM benzyladenine with the formation of an average of 6.1 shoots per explant over a period of 4 wk. The number of shoots increased with longer culture duration on proliferation medium. Multiple shoots which were maintained on the proliferation medium for 6 wk had the highest proliferation rate. Separation of multiple shoots and culturing in larger flasks significantly promoted the growth and formation of plantlets. All the in vitro plantlets survived when transferred to the field and showed no significant morphological differences from the mother plants.  相似文献

Summary Shi-hu (Dendrobium spp. or Dendrobii Herba) is one of the important traditional Chinese medicines. The commercially available crude drug in the traditional medicine market is composed mainly of three species: Dendrobium tosaense, D. nobile, and D. moniliforme. An efficient method of propagation has been developed via asymbiotic germination of seeds in vitro for the medicinally important D. tosaense. Seeds from capsules of D. tosaense collected 8–14 wk after artificial pollination germinated after being cultured on full-strength or half-strength Murashige and Skoog (MS) medium devoid of plant growth regulators and with 3% sucrose. Germination of seeds varied with the medium type and seed maturity. Germinated seedlings after transfer to MS medium with 1.5% sucrose and 8% banana homogenate or potato juice or coconut water and 20 wk of incubation developed into healthy plantlets. Well-developed plantlets were transplanted to moss or moss and tree fern or tree fern as substrates in plastic trays and transferred to a greenhouse for hardening. All plants survived, attained maturity, and developed normal flower and capsule after one and a half years. This protocol of successful plant regeneration by asymbiotic seed germination should permit rapid propagation and conservation of this medicinally important Dendrobium species.  相似文献

The amounts of colchicine present in two Jordanian species of Colchicum, namely, C. steveni Kunth and C. hierosolymitanum Feibrun (Colchicaceae), have been determined. An HPLC-UV (photodiode array) method employing gradient elution was developed and the results compared with those obtained using a simple TLC-spectrophotometric method. The levels of colchicine as measured by these methods were not significantly different (p < 0.05) indicating that the spectrophotometric method is an acceptable alternative to HPLC. With respect to C. steveni, the leaves contained the largest amount of colchicine (0.204/100 g), whilst in C. hierosolymitanum corms showed the highest colchicine content (0.126/100 g). As a source of colchicine, the two investigated species showed levels comparable with those found in C. autumnale, the traditional source of colchicine.  相似文献
