Vacuolar sorting receptors (VSRs) are type‐I integral membrane proteins that mediate biosynthetic protein traffic in the secretory pathway to the vacuole, whereas secretory carrier membrane proteins (SCAMPs) are type‐IV membrane proteins localizing to the plasma membrane and early endosome (EE) or trans‐Golgi network (TGN) in the plant endocytic pathway. As pollen tube growth is an extremely polarized and highly dynamic process, with intense anterograde and retrograde membrane trafficking, we have studied the dynamics and functional roles of VSR and SCAMP in pollen tube growth using lily (Lilium longiflorum) pollen as a model. Using newly cloned lily VSR and SCAMP cDNA (termed LIVSR and LISCAMP, respectively), as well as specific antibodies against VSR and SCAMP1 as tools, we have demonstrated that in growing lily pollen tubes: (i) transiently expressed GFP‐VSR/GFP‐LIVSR is located throughout the pollen tubes, excepting the apical clear‐zone region, whereas GFP‐LISCAMP is mainly concentrated in the tip region; (ii) VSRs are localized to the multivesicular body (MVB) and vacuole, whereas SCAMPs are localized to apical endocytic vesicles, TGN and vacuole; and (iii) microinjection of VSR or SCAMP antibodies and LlVSR small interfering RNAs (siRNAs) significantly reduced the growth rate of the lily pollen tubes. Taken together, both VSR and SCAMP are required for pollen tube growth, probably working together in regulating protein trafficking in the secretory and endocytic pathways, which need to be coordinated in order to support pollen tube elongation. 相似文献

Lily pollen was developed to express HBsAg by Agrobacterium-mediated transformation. A double prime-boost strategy combining parenteral and oral HBsAg boosters was found to increase antibody titer levels 10-fold, as compared to a single process of priming and boosting. Twelve polymorphic microsatellite loci were isolated and characterized from the threatened toad lily Tricyrtis flava. The number of alleles ranged from three to 17, and the observed and expected heterozygosities ranged from 0.35 to 0.96 and from 0.50 to 0.93, respectively, in a population of T. flava. We also tested cross‐species amplifications of the markers in three other species of Trycyrtis sect. Flavae. Most of 12 markers developed for T. flava were successfully amplified in the species of sect. Flavae and will be useful for population genetic studies of these species. 相似文献

Endemic Muscari muscarimi Medikus is the most fragrant plant among Muscari species and has a high ornamental potential. The natural populations of M. muscarimi, are severely affected by increased environmental pollution and urbanization. There is a need to develop a micropropagation method that should serve effectively for commercial propagation and conservation. Therefore, the study targeted to set up a strategy for efficient in vitro bulblet regeneration system of M. muscarimi using twin scale bulb explants on 1.0 × MS medium containing 4.44, 8.88, 17.76 μM BAP (6-Benzylaminopurine) plus 2.685, 5.37, 10.74 μM NAA (α-Naphthalene acetic acid). Maximum number of 19 daughter axillary bulblets and 16 daughter adventitious bulblets per twin bulb scale explant was regenerated on 1.0 × MS medium containing 17.76 μM BAP plus 10.74 μM NAA and 17.76 μM BAP plus 2.685 μM NAA respectively. The daughter bulblets regenerated on twin bulb scales on 8 out of 9 regeneration treatment could be easily rooted on 1.0 × MS medium containing 4.9 μM IBA (Indole-3-butyric acid). The daughter bulblets regenerated on 9th treatment (1.0 × MS medium containing 17.76 μM BAP plus 10.74 μM NAA) were transferred to 1.0 × MS medium containing 30 g/l sucrose to break negative carry over effect of this dose of BAP–NAA, where they grew 2–3 roots of variable length. Daughter bulblet diameter was increased by culturing them on 1.0 × MS medium containing 4.44 μM BAP plus 5.37 μM NAA. The results verified that both age and the source of explants had significant effect on regeneration. In another set of experiments, twin scales were obtained from in vitro regenerated daughter bulblets, although they induced bulblets, yet their bulblet regeneration percentage, mean number of bulblets per explant and their diameter were significantly reduced. In vitro regenerated bulblets were acclimatized in growth chamber under ambient conditions of temperature and humidity on peat moss, where they flowered. Chronic manganese (Mn) exposure in rodents, non-human primates and humans has been linked to Parkinson's disease like condition known as Manganism. Mn being a cofactor for many enzymes in brain has been known to be accumulated in various regions differentially and thus exert toxic effect upon chronic overexposure. In present study, neuropathology of Manganism was investigated by evaluating regional neuronal and astroglial metabolism in mice under chronic Mn exposure. Male C57BL6 mice were treated with MnCl(2) (25 mg/kg, i.p.) for 21 days. Cerebral metabolism was studied by co-infusing [U-(13)C(6)]glucose and [2-(13)C]acetate, and monitoring (13)C labeling of amino acids in brain tissue extract using (1)H-[(13)C] and (13)C-[(1)H]-NMR spectroscopy. Glutamate, choline, N-acetyl aspartate and myo-inositol were found to be reduced in thalamus and hypothalamus indicating a loss in neuronal and astroglial cells due to Mn neurotoxicity. Reduced labeling of Glu(C4) from [U-(13)C(6)]glucose and [2-(13)C]acetate indicates an impairment of glucose oxidation by glutamatergic neurons and glutamate-glutamine neurotransmitter cycle in cortex, striatum, thalamus-hypothalamus and olfactory bulb with chronic Mn exposure. Additionally, reduced labeling of Gln(C4) from [2-(13)C]acetate indicates a decrease in acetate oxidation by astroglia in the same regions. However, GABAergic function was alleviated only in thalamus-hypothalamus. Our findings indicate that chronic Mn impairs excitatory (glutamatergic) function in the majority of regions of brain while inhibitory (GABAergic) activity is perturbed only in basal ganglia. 相似文献

Leaf samples were collected from four spider lily and tobacco plants with tospovirus‐like symptoms in Yunnan province. The nucleocapsid ORFs of the four isolates were obtained by RT‐PCR, cloning and sequencing of the amplified products. Nucleotide sequences of the obtained amplicons were more than 90% identical to that of Calla lily chlorotic spot virus (CCSV) isolates deposited in the GenBank database. The virus induced systematic leaf necrosis, chlorotic lesions, rugosity and deformation on mechanically inoculated experimental hosts. This is the first report of CCSV in mainland China, and also on spider lily and tobacco. 相似文献

BACKGROUND AND AIMS: Success during the early stages of the life-history of alien plants is essential for invasion to occur. The reproductive components of plant invaders have mostly been studied in species reproducing sexually but little is known about invaders that depend exclusively on vegetative reproduction. In this paper, the importance of the different recruitment stages on population growth is quantified and, thus, the invasion potential of the South African annual geophyte Oxalis pes-caprae invading Mediterranean ecosystems is assessed. METHODS: Tests and experiments were conducted across Menorca (Balearic Islands) to analyse the spatial variability of Oxalis pes-caprae reproductive components (i.e. bulb production, bulb bank, bulb predation, bulb mortality, bulb dormancy, bulb germination, plant establishment and survival). KEY RESULTS: Oxalis pes-caprae has a transient bulb bank that remains dormant in the soil during summer. High levels of bulb predation after dispersal, followed by bulb mortality during summer or a failure to germinate in autumn were the most critical factors limiting plant establishment. Bulb germination was high. However, plant establishment and bulb production is constrained by intraspecific competition, but is not affected by soil disturbance. No symptoms of spatial discordance could be found between recruitment stages because the spatial variability of the life cycle was extremely low at all the scales examined (i.e. among populations, habitats and microsites). It was estimated that, on average, 4 % of bulbs can become plants the following year and the field rate of population increase (lambda) to be 0.08. CONCLUSIONS: The results suggest that invasion is constrained by post-dispersal bulb predation, loss of viability of the propagule bank due to summer drought and high intraspecific competition. However, a high spatial concordance between recruitment stages and probably a high propagule pressure due to human and livestock bulb dispersal determine the success of this invader across Menorca Island. 相似文献

Long-term potentiation (LTP) of synaptic transmission is considered a cellular mechanism for neural plasticity and memory
formation. Previously, we showed that in the carp olfactory bulb, LTP occurs at the dendrodendritic mitral-to-granule cell
synapse following tetanic electrical stimulation applied to the olfactory tract, and suggested that it is involved in the
process of olfactory memory formation. As a first step towards understanding mechanisms underlying plasticity at this synapse,
we examined the effects of various drugs (glutamate and GABA receptor agonists and antagonists, noradrenaline, and drugs affecting
cAMP signaling) on dendrodendritic mitral-to-granule cell synaptic transmission in an in vitro preparation. Two forms of LTP
are involved: a postsynaptic form (tetanus-evoked LTP) and a presynaptic form. The postsynaptic form is evoked at the granule
cell dendrite following tetanic olfactory tract stimulation and is suppressed by the NMDA receptor antagonist, D-AP5, enhanced
