The complicated routes by which organic material is channelled up to higher trophic levels via bacteria and protozoans is a major issue in aquatic microbial ecology. Because of the fragile nature of protists it is not straightforward to perform experimental studies of prey–predator interactions. Here we present an approach for the assessment of ciliate grazing on living heterotrophic nanoflagellates. Stationary phase cultures of a heterotrophic nanoflagellate (Cafeteria sp.) were live-stained by allowing them to take up fluorescently labelled macromolecules. Controls revealed that this label persisted for several hours. Fluorescently labelled living flagellates (FLLF) were added into enriched natural assemblages of marine oligotrich ciliates and uptake of FLLF was monitored over time. Oligotrich ciliates did not incorporate fluorescent-labelled macromolecules but a linear FLLF uptake over time was observed for 20–30 min at 20°C. Ingestion rates were 21–46 FLLF h–1 at a concentration of about 2×104 FLLF ml–1, which corresponded to clearance rates of 0.7–0.8 l ciliate–1 h–1. These results are in the same order of reported ciliate grazing on phytoplankton of similar size. This method represents a direct approach to measure ciliate grazing specifically on living heterotrophic nanoflagellates.  相似文献

异养硝化细菌的筛选、鉴定及其氨氮转化特性的研究   总被引:1,自引:0,他引:1  
从巢湖湖底泥床中分离筛选出一株具有氨氮转化活性的异养型硝化菌株X5.经生理生化分析、16S rDNA序列测定及系统发育学比较,菌株X5为枯草芽孢杆菌属(Bacillus sp.).菌株X5对氨氮的转化能力受温度、pH值以及接种量等条件的影响.实验结果表明,菌株X5的最适氨氮转化条件为:25~35℃,16 h,pH值7.0~8.0以及6%的接种量.将菌株X5扩大培养后接种于含蓝藻的发酵培养基中,分别测定总氮(TN)、氨氮(NH4+-N)及硝态氮(NO-3-N)的含量.结果显示,菌株X5对蓝藻中的氮素具有一定的转化作用.  相似文献

Colourless, nonscaled chrysophytes comprise morphologically similar or even indistinguishable flagellates which are important bacterivors in water and soil crucial for ecosystem functioning. However, phylogenetic analyses indicate a multiple origin of such colourless, nonscaled flagellate lineages. These flagellates are often referred to as “Spumella‐like flagellates” in ecological and biogeographic studies. Although this denomination reflects an assumed polyphyly, it obscures the phylogenetic and taxonomic diversity of this important flagellate group and, thus, hinders progress in lineage‐ and taxon‐specific ecological surveys. The smallest representatives of colourless chrysophytes have been addressed in very few taxonomic studies although they are among the dominant flagellates in field communities. To overcome the blurred picture and set the field for further investigation in biogeography and ecology of the organisms in question, we studied a set of strains of specifically small, colourless, nonscaled chrysomonad flagellates by means of electron microscopy and molecular analyses. They were isolated by a filtration‐acclimatisation approach focusing on flagellates of around 5 μm. We present the phylogenetic position of eight different lineages on both the ordinal and the generic level. Accordingly, we describe the new genera Apoikiospumella, Chromulinospumella, Segregatospumella, Cornospumella and Acrispumella Boenigk et Grossmann n. g. and different species within them.  相似文献

生物淋滤技术主要应用于低品位矿石金属选矿、煤气脱硫、废弃物中金属回收和污染介质中金属离子毒性的去除等方面。作为生物淋滤技术中的主体微生物之一,异养微生物可通过其产生的酸性代谢物还原、酸化及络合,提取或者溶解非硫化矿、固体废弃物、污水污泥及土壤中的金属,有助于解决目前的资源短缺问题,还可对污染环境治理提供技术支持,具有重要的理论意义和实践价值。应用于异养微生物淋滤技术中的常见微生物包括细菌(以假单胞菌为主)和真菌(以曲霉菌和青霉菌应用最为广泛)。淋滤过程涉及酸解、络合、还原及碱化等。目前,异养微生物淋滤技术主要应用于生物冶金、固体废弃物处理、污水处理和污染土壤修复等。本文分析了异养微生物金属淋滤过程中的问题,并提出了未来研究的发展方向。  相似文献

利用Li-8100土壤碳通量测量系统,研究了2013年4月—2014年3月浙江临安市毛竹人工林土壤呼吸、异养呼吸和自养呼吸速率的动态变化规律.结果表明:毛竹人工林土壤总呼吸速率、异养呼吸速率和自养呼吸速率均呈现出明显的季节变化特征,最高值出现在7月,最低值出现在1月,年平均值分别为2.93、1.92和1.01 μmol CO2·m-2·s-1.毛竹林土壤总呼吸、异养呼吸和自养呼吸年累积CO2排放量分别为37.25、24.61和12.64 t CO2·hm-2·a-1.土壤呼吸各组分均与土壤5 cm温度呈显著指数相关,土壤总呼吸、异养呼吸和自养呼吸的温度敏感系数Q10值分别为2.05、1.95和2.34.土壤总呼吸速率、异养呼吸速率与土壤水溶性有机碳(WSOC)含量均呈显著相关,而自养呼吸与WSOC无显著相关性;土壤呼吸各组分与土壤含水〖JP2〗量以及微生物生物量碳均无显著相关性.土壤温度是影响毛竹人工林土壤呼吸及其组分季节变化的主要驱动因子,土壤WSOC含量是影响土壤总呼吸和异养呼吸的重要环境因子.  相似文献

Abstract Sea water and sediment samples taken near the coasts of Hyeres bay (France) were used for anionic surfactant titrations with surface and bottom waters and the finest part of sediments. The capacity for surfactant degradation by the 'in situ' microflora was evaluated. By using a selective plating technique 26 strains able to utilize anionic surfactant were isolated from the selected bacterial communities. Their ability to degrade anionic surfactant was verified according to the biodegradation standard method. Isolated strains were characterized by morphological and physiological properties using API 20 NE micro-method. All tested strains were Gram negative, strictly aerobic, rod or helical shaped. Their weak utilization of phenolic substrates suggests that they degrade preferentially the alkyl chain of the surfactant molecule. Biodegradation was more efficient with bacterial communities rather than with any isolated strains. Such observations indicate that complete mineralization involves several other so far non-isolated strains which complete the degradation initiated by the isolated strains.  相似文献

The influence of inorganic nutrient concentrations on the ability of bacterioplankton to produce and degrade polysaccharidic transparent exopolymer particles (TEPs) and proteinaceous Coomassie-stained particles (CSPs) was investigated in an 11-day experiment. The dynamics of these particles were followed in prefiltered (1 microm) northern Adriatic seawater enclosures enriched either with 1 microM orthophosphate (main limiting nutrient in this area), 10 microM ammonium or both orthophosphate and ammonium. These enclosures were referenced to a nonenriched control. A high potential for bacterial TEP and CSP production was observed (10(4) - 10(5) L(-1) for particles larger than 4 microm). In conditions of high orthophosphate concentration (either orthophosphate enriched or both orthophosphate and ammonium enriched), lower abundances and surface areas of CSPs were obtained, whereas TEP dynamics were more affected by unbalanced enrichments where only orthophosphate or ammonium was added. The impact of unbalanced nutrient ratios on TEPs was indicated by their higher abundance but low capacity for Alcian blue absorption, implying a change in their structure. Inorganic nutrient availability was thus proven to affect the bacterial potential for producing and degrading bacterially derived TEPs and CSPs.  相似文献

研究了不同氮源下好氧反硝化菌Defluvibacter lusatiensis str.DN7的脱氮特性。结果表明:菌株均能以硝酸盐和亚硝酸盐为唯一氮源进行好氧反硝化作用。反应4 h,NO-3-N和NO-2-N的去除率分别达83.35%和85.72%。亚硝酸盐完全还原比硝酸盐提前42 h。硝酸盐还原过程中基本无亚硝酸盐积累,而亚硝酸盐还原过程中则检测到明显的硝酸盐积累,反应4 h,NO-3-N积累量达到21.83 mg/L。培养液中同时存在硝酸盐和亚硝酸盐时,菌株优先选择硝酸盐作电子受体。亚硝酸盐共存对硝酸盐还原无显著影响,但培养液中残留的NO-2-N随亚硝酸盐比例上升而增加,当亚硝酸盐比例从10%升至50%时,NO-2-N残留量由3.38 mg/L增至7.60 mg/L。少量硝酸盐的加入对亚硝酸盐的还原产生抑制作用。当硝酸盐比例为10%时,72 h NO-2-N的去除率仅为74.79%,远低于以亚硝酸盐为唯一氮源情况(去除率100%)。以氨氮为唯一氮源时,菌株同时进行异养硝化和好氧反硝化反应,72 h,NH+4-N去除率达85.66%,且基本无硝酸盐或亚硝酸盐积累。少量氨氮共存(氨氮比例<30%)有利于促进菌株的好氧反硝化作用,反之亦然。  相似文献

The objective of this work was to analyse the distribution of the planktonic communities involved in the functioning of a 255-km river stretch and to get a better understanding of the influence of the river morphology on the diversity and dynamics of the micro-organisms. The planktonic communities (phytoplankton, bacterioplankton, proto- and metazooplankton) scarcely considered together in fluvial systems, were analysed at four sites of the Middle Loire during the low water period, in parallel to physical and chemical analyses. Physical and chemical variables such as turbidity, pH, suspended matter and chlorophyll a concentration were high, illustrating the classical, productive summer period. The algae played a major role in the water oxygenation until end-summer, then the algal drop concomitant to the bacterial sustained abundance appeared responsible for oxygen depletion. The downstream site enriched by nutrients inputs of two tributaries, carried the highest algal and bacterial densities. Situated in a meanders zone, the Middle Loire is characterised by a high habitat heterogeneity, the up- and downstream sites were wide and spread of vast standing zones and vegetated islands, whereas the two intermediate ones were narrower and more uniform. This morphological variability strongly impacted the micro-organisms diversity and distribution. Indeed, the algae and zooplankton composition were clearly influenced by the physical habitats of the river, the Cyanophyta were favoured by the lentic conditions and the Bacillariophyta by the turbulent ones, while the young stage of copepod and the large rotifer predators were indicator of a lentic origin. Thereafter, the river heterogeneity interfered with the zooplankton dynamics, the standing conditions enhancing the rotifer predation. In that way, we hypothesise that two opposite patterns characterised the wide sites spread of lentic water and the more uniform channels. In the first case, the zooplankton could prey on the ciliates protozoan, which in return favoured the flagellate ones; conversely in the second situation the zooplankton limited by the physical constraints did not impact the ciliates which could depress the flagellates. Thus, the similar geomorphology of the distant upstream and downstream sites (255-km apart) induced relatively close organisms distribution. Hence, disagreeing with the river continuum concept, this assertion shows the strong influence played by the local morphological characteristics of the Middle Loire in potamoplankton composition and dynamics.  相似文献
