The filamentous fungus Beauveria bassiana is a natural pathogen of the greater wax moth Galleria mellonella. Infection with this fungus triggered systemic immune response in G. mellonella; nevertheless, the infection was lethal if spores entered the insect hemocel. We observed melanin deposition in the insect cuticle and walls of air bags, while the invading fungus interrupted tissue continuity. We have shown colonization of muscles, air bags, and finally colonization and complete destruction of the fat body—the main organ responsible for the synthesis of defense molecules in response to infection. This destruction was probably not caused by simple fungal growth, because the fat body was not destroyed during colonization with a human opportunistic pathogen Candida albicans. This may mean that the infecting fungus is able to destroy actively the insect's fat body as part of its virulence mechanism. Finally, we were unable to reduce the extremely high virulence of B. bassiana against G. mellonella by priming of larvae with thermally inactivated fungal spores. 相似文献

Phylogeographical studies are typically based on haplotype data, occasionally on nuclear markers such as microsatellites, but rarely combine both. This is unfortunate because the use of markers with contrasting modes of inheritance and rates of evolution might provide a more accurate and comprehensive understanding of a species' history. Here we present a detailed study of the phylogeography of the greater horseshoe bat, Rhinolophus ferrumequinum , using 1098 bp of the mitochondrial ND2 gene from 45 localities from across its Palaearctic range to infer population history. In addition, we re-analysed a large microsatellite data set available for this species and compared the results of both markers to infer population relationships and the historical processes influencing them. We show that mtDNA, the most popular marker in phylogeography studies, yielded a misleading result, and would have led us to conclude erroneously that a single expansion had taken place in Europe. Only by combining the mitochondrial and microsatellite data sets are we able to reconstruct the species' history and show two colonization events in Europe, one before the Last Glacial Maximum (LGM) and one after it. Combining markers also revealed the importance of Asia Minor as an ancient refugium for this species and a source population for the expansion of the greater horseshoe bat into Europe before the LGM. 相似文献

ABSTRACT Nest predation is a natural component of greater sage-grouse (Centrocercus urophasianus) reproduction, but changes in nesting habitat and predator communities may adversely affect grouse populations. We used a 2-part approach to investigate sage-grouse nest predation. First, we used information criteria to compare nest survival models that included indices of common raven (Corvus corax) abundance with other survival models that consisted of day of incubation, grouse age, and nest microhabitat covariates using measurements from 77 of 87 sage-grouse nests. Second, we used video monitoring at a subsample of 55 of 87 nests to identify predators of depredated nests (n = 16) and evaluated the influence of microhabitat factors on the probability of predation by each predator species. The most parsimonious model for nest survival consisted of an interaction between day of incubation and abundance of common ravens (wravenXincubation day = 0.67). An estimated increase in one raven per 10-km transect survey was associated with a 7.4% increase in the odds of nest failure. Nest survival was relatively lower in early stages of incubation, and this effect was strengthened with increased raven numbers. Using video monitoring, we found the probability of raven predation increased with reduced shrub canopy cover. Also, we found differences in shrub canopy cover and understory visual obstruction between nests depredated by ravens and nests depredated by American badgers (Taxidea taxus). Increased raven numbers have negative effects on sage-grouse nest survival, especially in areas with relatively low shrub canopy cover. We encourage wildlife managers to reduce interactions between ravens and nesting sage-grouse by managing raven populations and restoring and maintaining shrub canopy cover in sage-grouse nesting areas. 相似文献

Abstract: An assumption of mark-recapture studies is that the marker has no effect on the animal. Neck bands have been used extensively for goose research, but there has long been concern that they may have negative effects on some demographic parameters, and recent studies have yielded contradictory results. We evaluated the effects of neck bands on adult female greater snow geese (Chen caerulescens atlantica) by contrasting breeding propensity and apparent survival of geese marked with both a plastic neck band and a metal leg band and those marked solely with metal leg bands over an 11-year period on Bylot Island, Nunavut Territory, Canada. The use of multistate mark-recapture models also allowed us to estimate neck band loss and to obtain survival and capture probabilities that were not biased by such loss. Finally, we tested the effects of neck bands on other reproductive parameters (laying date, clutch size and nest success) over a 3-year period. Neck-banded females had decreased clutch size and capture probabilities, but their apparent survival rate, nest initiation and hatching dates, and nest survival were not affected compared to leg-banded only or unbanded females. Breeding propensity, indexed by capture probabilities of neck-banded females was, on average, 48% lower that that of leg-banded-only females but clutch size was only 10% lower. Neck band loss of females was low in this population (3% per year). We urge researchers to be cautious in the use of neck bands for estimation of population parameters and that the potential negative effects of neck bands be assessed as it is likely to be species-specific. 相似文献

The phylogeography of the bush habituated African bovid species impala (Aepyceros melampus) and greater kudu (Tragelaphus strepsiceros) is investigated using mitochondrial DNA (mtDNA) markers. Combined analysis of individual lineages, relationships and population genetics suggest a colonization process from Southern Africa toward Eastern regions in the greater kudu. Results are less clear for the impala, although remaining consistent with a similar pattern of historical dispersion. The study reveals a similar pattern, that is a marked divergence of lineages from South-western Africa relative to other regions. This pattern is opposed to previously published findings in other African bovid species. In the impala, the genetically isolated region is consistent with morphology because it is recognized as the subspecies A. m. petersi, the black-faced impala. In contrast, the similar split of South-western mitochondrial lineages was not expected in the greater kudu on the basis of morphology. Both species show a significant population genetic differentiation. Beyond their phylogeographical value, our results should raise conservation concerns about South-western populations of both species. The black-faced impala is categorized as vulnerable and our data show indications of hybridization with common impala A. m. melampus. The previously unrecognized genetic status of the South-western kudus could also imply conservation regulations. 相似文献
