Phosphoenolpyruvate (PEP) carboxylase (EC 4.1.1.31) and ribulose-1,5-bisphospate (RuBP) carboxylase (EC 4.1.1.39) activities in leaves of different maize hybrids grown under field conditions (high light intensity) and in a growth chamber (low light intensity) were determined. Light intensity and leaf age affected PEP carboxylase activity, whereas RuBP carboxylase was affected by leaf age only at low light intensity. PEP carboxylase/RuBP carboxylase activity ratio decreased according to light intensity and leaf age. Results demonstrate that Zea mays grown under field conditions is a typical C4 species in all leaves independently from their position on the stem, whereas it may be a C3 plant when it is grown in a growth chamber at low light intensityAbbreviations PEP phosphoenolpyruvate - RuBP ribulose-1,5-bisphosphate  相似文献

A full accounting of net greenhouse gas balance (NGHGB) and greenhouse gas intensity (GHGI) was examined in an annual coastal reclaimed saline Jerusalem artichoke-fallow cropping system under various soil practices including soil tillage, soil ameliorant, and crop residue amendments. Seasonal fluxes of soil carbon dioxide (CO2), methane (CH4), and nitrous oxide (N2O) were measured using static chamber method, and the net ecosystem exchange of CO2 (NEE) was determined by the difference between soil heterotrophic respiration (RH) and net primary production (NPP). Relative to no-tillage, rotary tillage significantly decreased the NPP of Jerusalem artichoke while it had no significant effects on the annual RH. Rotary tillage increased CH4 emissions, while seasonal or annual soil N2O emissions did not statistically differ between the two tillage treatments. Compared with the control plots, soil ameliorant or straw amendment enhanced RH, soil CH4, and N2O emissions under the both tillage regimes. Annual NGHGB was negative for all the field treatments, as a consequence of net ecosystem CO2 sequestration exceeding the CO2-equivalents released as CH4 and N2O emissions, which indicates that Jerusalem artichoke-fallow cropping system served as a net sink of GHGs. The annual net NGHGB and GHGI were estimated to be 11–21% and 4–8% lower in the NT than in RT cropping systems, respectively. Soil ameliorant and straw amendments greatly increased NPP and thus significantly decreased the negative annual net NGHGB. Overall, higher NPP but lower climatic impacts of coastal saline bioenergy production would be simultaneously achieved by Jerusalem artichoke cultivation under no-tillage with improved saline soil conditions in southeast China.  相似文献

We present a protocol and calculation methods for the determination of zooplankton ingestion and assimilation rates with stable isotope tracers. These methods have been developed from experiments with the rotifer Brachionus calyciflorus that had been fed 13C-labelled Scenedesmus obliquus. Stable isotope tracers offer the same advantages as radioisotopes. These include the possibility for direct and accurate quantification of ingestion and assimilation rates, short sample analysis times and low animal densities requirements. However, the use of stable isotope tracers requires relatively long sample preparation times and specialist equipment and is, thus, relatively costly for most laboratories. The application of stable isotope tracers in zooplankton feeding studies offers several advantages in comparison with radioisotopes. Firstly, they do not emit harmful radiation and can therefore be applied safely both in the laboratory and in the field. Secondly, the samples can be dried for safe storage and easy transportation. Thirdly, no aggressive chemicals are required for sample analysis.  相似文献

Cellulose samples treated with sodium hydroxide (NaOH) and carbon dioxide in dimethylacetamide (DMAc) were analyzed by FTIR spectroscopy. Absorbance of hydrogen-bonded OH stretching was considerably decreased by the treatment of NaOH and carbon dioxide. The relative absorbance ratio (A(4000-2995)/A(993)) represented the decrease of absorbance as a criterion of hydrogen-bond intensity (HBI). The absorbance of the band at 1430cm(-1) due to a crystalline absorption was also decreased by NaOH treatment. The absorbance ratio of the bands at 1430 and 987-893cm(-1) (A(1430)/A(900)), adopted as crystallinity index (CI), was closely related to the portion of cellulose I structure. With the help of FTIR equipped with an on-line evacuation apparatus, broad OH bending due to bound water could be eliminated. FTIR spectra of the carbon dioxide-treated cellulose samples at 1700-1525cm(-1) were divided into some bands including 1663, 1635, 1616, and 1593cm(-1). The broad OH bending due to bound water at 1641-1645cm(-1) was resolved to two bands at 1663 and 1635cm(-1). As a trace of DMAc, the band at 1616cm(-1) is disappeared by washing for the cellulose treated with carbon dioxide (Cell 1-C and Cell 2/60-C). The decrease of HBI, the easy removal of DMAc, and the band at 1593cm(-1) supported the introduction of new chemical structure in cellulose. The bands shown at 1593 and 1470cm(-1) was assigned as hydrogen-bonded carbonyl stretching and O-C-O stretching of the carbonate ion.  相似文献

  • 2 The data showed population dynamics similar to other small freshwater bodies; diatoms were numerous during the spring, chlorophytes dominated during the summer months, and mixotrophs, in particular Gymnodinium, dominated during the autumn and winter.
  • 3 The mixotrophic dinoflagellates were responsible for a high chlorophyll concentration during the autumn and winter. Mixotrophs were important consumers of bacteria, particularly during the autumn when population densities of pure heterotrophs were low. Mixotrophs were an important component of the microbial food web in this pond.

  • 1 Planktivorous fish were hypothesised to influence the abundance of algal biomass in lakes by changing zooplankton grazing, affecting zooplankton nutrient recycling and by direct recycling of nutrients to phytoplankton. The relative roles of direct fish effects vs. zooplankton grazing were tested in mesocosm experiments by adding to natural communities large grazing zooplankton (Daphnia carinata) and small planktivorous fish (mosquitofish or juveniles of Australian golden perch).
  • 2 The addition of Daphnia to natural communities reduced the numbers of all phytoplankton less than 30 µm in size, but did not affect total biomass of phytoplankton as large Volvox colonies predominated.
  • 3 The addition of Daphnia also reduced the abundance of some small (Moina, Bosmina, Keratella) and large (adult Boeckella) zooplankton, suggesting competitive interactions within zooplankton.
  • 4 The addition of mosquitofish to communities containing Daphnia further reduced the abundance of some small zooplankton (Moina, Keratella), but increased the numbers of Daphnia and adult Boeckella. In spite of the likely increase in grazing due to Daphnia, the abundance of total phytoplankton and dominant alga Volvox did not decline in the presence of mosquitofish but was maintained at a significantly higher level than in control.
  • 5 The addition of juveniles of golden perch to communities containing Daphnia reduced the abundance of small zooplankton (Moina), increased the abundance of large zooplankton (adult Boeckella) but had no significant effect on Daphnia and total phytoplankton abundance.
  • 6 The results of the present study suggest that some planktivorous fish can promote the growth of phytoplankton in a direct way, probably by recycling nutrients, and even in the presence of large grazers. However, the manifestation of the direct effect of fish can vary with fish species.

Pasture lands are an important facet of land use in the northeast United States, yet little is known about their recent diversity. To answer some fundamental questions about the diversity of these pasture lands, we designed a broad survey to document plant species richness using an intensive, multi scale sampling method. We also wanted to learn whether environmental (soils or climate) or land management variables could help explain patterns of species richness. A total of 17 farms, encompassing 37 pastures, were sampled in New York, Pennsylvania, Vermont, Maryland, Massachusetts and Connecticut during July and August 1998. We positively identified a total of 161 different plant species across the study region. Species richness averaged 31.7±1.1 on pastures. Infrequent, transient species that were mostly perennial and annual forbs accounted for 90% of the species richness. Except for a subjective rating of grazing intensity, land management methods were not good predictors of species richness. Over time, it appears that grazing neither reduces nor increases species richness in pastures. Of the environmental variables measured, only soil P explained a significant amount of the variation in species richness. Soil P was inversely related to species richness at the 1m2 scale. Percent SOM was positively associated with species richness at this scale, although weakly. At larger spatial scales, we suggest that patterns of species richness are best explained by the species diversity of soil seed banks, or seed rain, and stochastic recruitment of these species into existing vegetation.  相似文献

The effects of patches formed by a deciduous shrub (Salix reinii) and an evergreen shrub (Gaultheria miqueliana) on cohabitants were examined on an active volcano Mt. Koma, northern Japan, using bareground as a comparative baseline (control). Species richness increased with increasing the areas of three habitats (Salix patches, Gaultheria patches and bareground), but significantly slowed in Gaultheria patches where the shoot densities of cohabitants also decreased. Calamagrostis hakonensis and Spiranthes sinensis were 2–3 times more likely to establish in Salix patches than in bareground, Anaphalis margaritacea var. angustior and Campanula lasiocarpa were 4–5 times more likely to establish on bareground than in Salix patches. Cohabitants was fewer with Gaultheria. Light intensities of the two patch types were ca 1/3 of that on bareground. Light intensity on Gaultheria was ca 88% of that on Salix patch and decreased with increasing patch size, because the differences in stem and leaf architectures and phenology between the two patch-forming species. Moisture and total nitrogen in volcanic deposits were 2–3 times higher in the patches than in bareground. In the patches, low light intensity restricted seed germination, in particular, for the plants characteristic of bareground, e.g., Anaphalis, while high soil moisture increased the survival rates of Calamagrostis. In Gaultheria patches, despite high moisture, the reduced light intensity influenced seed germination and inhibited the establishment of most species. Seedbanks estimated by a lab-experiment indicated that Salix patches trapped more seeds. As a consequence, Salix patches act as facilitator, in particular, for Calamagrostis, by improving moisture and nutrients status of volcanic deposits, moderating light intensity and seed trapping. Deciduous shrub patches, such as Salix reinii, have an improving role in accelerating rates of succession in harsh environments, while evergreen shrub patches slow plant establishment.
