The value of biotin-avidin (B-A) ELISA for the detection of grapevine fanleaf virus (GFLV) in Xiphinema was estimated with field populations and greenhouse subpopulations. Samples consisted of increasing numbers of adults ranging from 1 to 64 in multiples of two. Tests with virus-free X. index populations reared on grapevine and fig plants as negative controls did not reveal a noticeable effect of the host plant. ELISA absorbances of virus-free X. index samples were greater than corresponding absorbances of X. pachtaicum samples. Differences occurred between two X. index field populations from GFLV-infected grapevines in Champagne and Languedoc. In most tests, 1-, 2-, 4-, and 8-nematode samples of virus-free and virus-infected populations, respectively, could not be separated. Consequently, B-A ELISA was not a reliable method for GFLV detection in samples of less than 10 X. index adults, but comparison of the absorbances obtained with increasing numbers may allow differentiation of the viral infectious potential of several populations.  相似文献

The effects of fatty acids, extracted during prefermentation grape skin-contact on Saccharomyces cerevisiae and Oenococcus oeni, were studied. The influence of skin-contact on total fatty acid content was evaluated both in Chardonnay must and in synthetic medium. Prior to alcoholic fermentation, the skin-contact contributes to a large enrichment of long-chain fatty acids (C16 to C18:3). These results induced a positive effect on yeast growth and particularly on cell viability. In the skin-contact fermented media, levels of C12 and especially C10 are lower and macromolecules content higher than in controls. This production of extracellular mannoproteins and the reduction of medium-chain fatty acids in media by S. cerevisiae increased growth of O. oeni. The influence of fatty acids (C10 to C18:3), in their free and esterified forms, on bacterial growth and on malolactic activity was also examined. Only C10 and C12, especially in their esterified forms, always appeared to be toxic to O. oeni. Received 15 May 1997/ Accepted in revised form 02 December 1997  相似文献

Cultured grape cells accumulate anthocyanins in vacuoles rather than secreting them into the nutrient medium. Therefore, grape cells that contain tonoplast segments in their plasmalemma should be capable of excreting anthocyanins rather than sequestering them in their vacuoles. In initial attempts to construct such novel cells, small vacuoles were fused with the plasmalemma of cultured plant cells. Protoplasts were isolated from grape calluses that produce and accumulate anthocyanins. Small vacuoles were formed by gently rupturing vacuoles isolated from grape protoplasts. Although small vacuoles and protoplasts became aligned in an AC field, the tonoplast and plasmalemma did not readily fuse when subjected to 3 DC pulses of 1200 V cm–1 for 50 s each. Changes in the intensity, number and/or duration of the DC pulses had no effect on the fusion process. When 1.0% polyethylene glycol was added to the electrofusion buffer, however, small vacuoles and protoplasts fused within a few minutes after the DC pulses were applied. Adventitious shoots developed from in vitro-grown leaves of Vitis vinifera cultivars Cabernet Sauvignon, French Colombard, Grenache, Thompson Seedless (syn. Sultana) and White Riesling, V. rupestris cv. St. George (syn. du Lot) and V. vinifera × rupestris cv. Ganzin 1. Leaf explants less than 15 mm long were excised from nodal cultures and cultured on Murashige and Skoog or Nitsch and Nitsch-based regeneration media with 0, 1, 2 or 4 mgl-1 6-benzylaminopurine (BAP). Adventitious shoots developed within 4 weeks at the petiolar stub and occasionally from wounded lamina tissues. Shoot organogenesis occurred only on media containing BAP and at a higher frequency with 2 mgl-1 than with 1 or 4 mgl-1. On media containing 2 mgl-1 BAP, 47, 67, 60, and 42%, respectively, of leaf explants of Cabernet Sauvignon, French Colombard, Thompson Seedless, and White Riesling produced adventitious shoots compared to 14, 14, and 29%, respectively, for Grenache, St. George, and Ganzin 1. Solid culture medium was superior to liquid medium and transfer frequency on solid medium did not affect the regeneration frequency. Further shoot growth was promoted by the transfer of regenerating tissues to fresh regeneration medium. More than 80% of explants initially producing adventitious buds exhibited further shoot growth, developing an average of more than 6 shoots each. Shoots rooted easily and the resulting plants appeared morphologically identical to parent vines.  相似文献

The initial velocity of the oxidation of 4-methylcatechol by grape catechol oxidase was determined. The kinetic analysis indicates that first there is random binding of an oxygen and a 4-methylcatechol molecule to the enzyme. Then one product molecule is released prior to the binding of second 4-methylcatechol molecule which is followed by the release of a second product molecule. The true Km values were determined; they were found to be 0.5 mM for oxygen and 17 mM for 4-methylcatechol.  相似文献

以不同葡萄组织为材料对目前常用的2种总RNA提取方法一改良SDS法和CTAB-LiCl法进行研究。2种RNA提取方法中均不使用酚。采用这两种方法从葡萄不同组织中均成功地提取到RNA,琼脂糖凝胶电泳结果显示28s和18SrRNA条带完整清晰。检测A260/A280 值分布在1.7-2.0之间,A260/A230值分布于1.9-2.3之间,说明RNA质量较高。管家基Actin和ACS5基因的检测表明2种方法所得RNA能够满足RT-PCR和基因克隆等研究需要。改良SDS法的RNA得率是CTAB-LiCl法的RNA得率的2~3倍,而CTAB-LiCl法获得的RNA纯度高。可以根据原料的数量和对RNA质量的要求来选取最佳提取方法。  相似文献

以12个葡萄品种1年生枝条为试材,通过0、-15、-18、-21、-24、-27、-30、-133℃8个低温处理24h后,测定不同低温胁迫下葡萄枝条的,6-含水量、束缚水含量、自由水含量和束缚水与自由水的比值。结果表明:随着处理温度的降低,同一品种枝条的总含水量基本恒定,自由水含量呈先下降再升高的趋势,束缚水含量和束缚水与自由水的比值均呈现为先升高再下降的趋势;束缚水与自由水的比值随低温胁迫的变化呈Cubic方程,不同品种束缚水与自由水的比值达到最高点的温度不同,‘双红’、‘北冰红’、‘左优红’的拐点温度在-25℃以下,‘775’、‘巨峰’的拐点温度在-20℃左右,‘雷司令’、‘黑比诺’、‘霞多丽’的拐点温度在-18℃左右,‘赤霞珠’、‘梅鹿特’、‘白比诺’、‘红地球’拐点温度在-16~17℃。对葡萄休眠枝条进行低温处理后,通过测定束缚水与自由水含水量变化鉴定葡萄品种的抗寒性是可行的。  相似文献

本文研究了高温与不同光强结合处理对‘赤霞珠’葡萄叶片PSII活性及恢复的影响。结果表明,高温黑暗处理(40℃,0μmaol·m-2.s-1)导致叶片PSII最大光化学效率(Fv/Fm)、反应中心吸收的光能用于电子传递的量子产额(ψEo)与单位反应中心光能的传递(ETo/RC)降低明显,且无恢复趋势,K点相对荧光(Vk)、单位反应中心光能的吸收(ABS/RC)与捕获(TRo/RC)显著升高。高温弱光处理(40℃,200μmol·m-2.s-1)后的叶片PSII活性明显恢复,ETo/RC降低明显,TRo/RC无显著变化。高温强光(40℃,1600μmol·m-2.S-1)处理导致单位面积有活性反应中心数量(RC/CSm)抑制程度最大,恢复程度较低。实验结果说明,高温处理下黑暗对葡萄PSII功能活性及恢复均会造成抑制,而弱光可以显著缓解高温对葡萄叶片的胁迫作用,并促进PSII的恢复,强光导致胁迫下的PSII功能抑制最明显。  相似文献

选取广西、湖南等地野生葡萄,与经典酿酒葡萄比较,研究抗氧化活性和活性物质,同时监测葡萄酒发酵过程中各指标的动态变化,并对不同品种葡萄酒的抗菌性进行研究。结果表明:赤霞珠的酚类含量和抗氧化活性高于野生葡萄和玫瑰香葡萄,但野生葡萄酒的抗菌性能显著优于赤霞珠和玫瑰香葡萄酒。葡萄酒在发酵过程中其抗氧化活性和酚类物质含量均随发酵过程的进行而升高;总抗氧化活性与总酚含量、氧自由基清除能力与原花青素含量成显著正相关,相关系数均大于0.989;总花色苷含量在发酵初期上升,后期下降,葡萄酒颜色变浅。  相似文献
