Abstract Fire frequency is the number of fires experienced by a particular community within a given time period. This variable can potentially be resolved into a number of interacting components, including time since the most recent fire, and the length of the inter-fire intervals. We estimated the effects of inter-fire intervals and sequences of inter-fire intervals, independently of time-since-fire, on the floristic composition of 26 samples from dry sclerophyll vegetation with different fire histories in Brisbane Water National Park near Sydney. In particular, we examined the effects in the recent fire history of: the length of the shortest inter-fire interval (1–26 years); repetition of very short (1–3 years) and medium (7–14 years) inter-fire intervals; and recovery from very short (1–3 years) and short (4–6 years) inter-fire intervals. Our analyses suggest that inter-fire interval may account for about 55% of the floristic variation among our samples, which were taken at a uniform 18 months after the most recent fire. There appear to be at least three general and unrelated effects of the recent history of inter-fire intervals on the floristic composition of this vegetation: (i) shorter inter-fire intervals were associated with a reduction in the number of species present in the community, along with a more unequal abundance of the remaining species, particularly of those large Proteaceae shrubs that often dominate the community biomass in dry sclerophyll shrublands of southeastern Australia; (ii) the presence of an inter-fire interval that is 1–6 years long was associated with an additional reversible reduction in the number of fire-sensitive species present in the community, particularly those common shrub species with a canopy-stored seed-bank and non-leguminous species with a soil-stored seed-bank; and (iii) repetition of inter-fire intervals that are 1–5 years long was associated with an increase in the abundance of herbaceous fire-tolerant species. This means that variation in these characteristics is associated with changes in abundance of different plant species, and that the floristic composition of any one area can be influenced in three different ways by these variables. Management of fire for plant species conservation must therefore be based on both length of time-since-fire and length of inter-fire intervals if it is to be effective.  相似文献

The terminal DNA sequence requirements for piggyBac transposable element excision were explored using a plasmid-based assay in transfected, cultured insect cells. A donor plasmid containing duplicate 3′piggyBac terminal inverted repeats was constructed that allowed individual nucleotides or groups of nucleotides within one of the 3′ repeats to be mutated. The relative extent of excision using the mutated end versus the wild-type end was then assayed. Removal of even one of the terminal 3′ G nucleotides from the piggyBac inverted repeat, or removal of the dinucleotide AA from the flanking TTAA target site prevents excision of piggyBac at the mutated terminus. Incorporation of an asymmetric TTAC target site at the 3′ end does not prevent excision from the mutated end. Summary We used various microscopic and labeling techniques to examine events occurring during the programmed cell death (PCD) of plant tracheary elements (TEs) developing in vitro. TEs differentiating in vitro synthesize a secondary cell wall which is complex in composition and pattern at approximately 72 h after hormone manipulation. The timing of PCD events was established relative to this developmental marker. Cytoplasmic streaming continues throughout secondary wall synthesis, which takes 6 h to complete in a typical cell. Vital dye staining and ultrastructural analysis show that the vacuole and plasma membrane are intact during secondary cell wall synthesis, but the cytoplasm becomes less dense in appearance, most likely through the action of confined hydrolysis by small vacuoles which are seen throughout the cell at this time. The final, preeminent step of TE PCD is a rapid collapse of the vacuole occurring after completion of secondary cell wall synthesis. Vacuole collapse is an irreversible commitment to death which results in the immediate cessation of cytoplasmic streaming and leads to the complete degradation of cellular contents, which is probably accomplished by release of hydrolytic enzymes sequestered in the vacuole. This event represents a novel form of PCD. The degradation of nuclear DNA is detectable by TUNEL, an in situ labeling method, and appears to occur near or after vacuole collapse. Our observations indicate that the process of cellular degradation that produces the hollow TE cell corpse is an active and cell-autonomous process which is distinguishable morphologically and kinetically from necrosis. Although TE PCD does not resemble apoptosis morphologically, we describe the production of spherical protoplast fragments by cultured cells that resemble apoptotic bodies but which are not involved in TE PCD. We also present evidence that, unlike the hypersensitive response (HR), TE PCD does not involve an oxidative burst. In Drosophila melanogaster, crossing males carrying autonomous P elements with females devoid of P copies results in hybrid dysgenesis in the germline of progeny. The reciprocal cross produces non-dysgenic progeny due to a maternally inherited state non-permissive for P transposition. The capacity of a P copy to repress transposition depends on both its structure and its chromosomal location. Naturally occuring regulatory P elements inserted at the telomere of the X chromosome have been genetically isolated in a genomic context devoid of other P elements. One or two copies of autonomous P elements at this site (1A) are sufficient to elicit a strong P repression in the germline. These elements are flanked by Telomeric Associated Sequences, previously identified and described by Karpen and Spradling (1992) as having heterochromatic properties. The regulatory properties of P elements at 1A are strongly impaired by mutations affecting Su(var)205, which encodes Heterochromatin Protein 1, a non-histone heterochromatin protein. The regulatory properties of classical P strains are not sensitive to Su(var)205. Models based on chromatin structure or on nuclear localisation of the telomeres are discussed in order to explain both the strong regulatory properties of P elements at the X chromosome telomere and their sensitivity to Su(var)205.
