The contextual and cued fear conditioning test is one of the behavioral tests that assesses the ability of mice to learn and remember an association between environmental cues and aversive experiences. In this test, mice are placed into a conditioning chamber and are given parings of a conditioned stimulus (an auditory cue) and an aversive unconditioned stimulus (an electric footshock). After a delay time, the mice are exposed to the same conditioning chamber and a differently shaped chamber with presentation of the auditory cue. Freezing behavior during the test is measured as an index of fear memory. To analyze the behavior automatically, we have developed a video analyzing system using the ImageFZ application software program, which is available as a free download at http://www.mouse-phenotype.org/. Here, to show the details of our protocol, we demonstrate our procedure for the contextual and cued fear conditioning test in C57BL/6J mice using the ImageFZ system. In addition, we validated our protocol and the video analyzing system performance by comparing freezing time measured by the ImageFZ system or a photobeam-based computer measurement system with that scored by a human observer. As shown in our representative results, the data obtained by ImageFZ were similar to those analyzed by a human observer, indicating that the behavioral analysis using the ImageFZ system is highly reliable. Fear of certain threat and anxiety about uncertain threat are distinct emotions with unique behavioral, cognitive-attentional, and neuroanatomical components. Both anxiety and fear can be studied in the laboratory by measuring the potentiation of the startle reflex. The startle reflex is a defensive reflex that is potentiated when an organism is threatened and the need for defense is high. The startle reflex is assessed via electromyography (EMG) in the orbicularis oculi muscle elicited by brief, intense, bursts of acoustic white noise (i.e., “startle probes”). Startle potentiation is calculated as the increase in startle response magnitude during presentation of sets of visual threat cues that signal delivery of mild electric shock relative to sets of matched cues that signal the absence of shock (no-threat cues). In the Threat Probability Task, fear is measured via startle potentiation to high probability (100% cue-contingent shock; certain) threat cues whereas anxiety is measured via startle potentiation to low probability (20% cue-contingent shock; uncertain) threat cues. Measurement of startle potentiation during the Threat Probability Task provides an objective and easily implemented alternative to assessment of negative affect via self-report or other methods (e.g., neuroimaging) that may be inappropriate or impractical for some researchers. Startle potentiation has been studied rigorously in both animals (e.g., rodents, non-human primates) and humans which facilitates animal-to-human translational research. Startle potentiation during certain and uncertain threat provides an objective measure of negative affective and distinct emotional states (fear, anxiety) to use in research on psychopathology, substance use/abuse and broadly in affective science. As such, it has been used extensively by clinical scientists interested in psychopathology etiology and by affective scientists interested in individual differences in emotion. 相似文献

In mountain gorillas (Gorilla g. beringei), male immigration in bi-sexual units is rare. This paper presents the case of a nearly weaned male infant gorilla who followed
his mother in her transfer. This case was recorded in the study population at the Kariske Research Center in 1988. The data
come from observation in Group B (on 12 days just prior to the transfer and on 54 days after the transfer over a period of
6 months). The situation of the infant did match the conditions in which infanticide occurs in gorillas, but he was not killed,
despite receiving male aggression and being wounded twice. In fact, both the mother and the infant received male aggression
more frequently than the long term residents in the group. The aggression received by the mother decreased after she mated
with the males and after she weaned the infant. The aggression received by the infant, however, did not decrease after his
mother mated with the males, and increased in intensity. The infant reacted fearfully to male aggression, in marked contrast
to his mother, who reacted either with indifference, or by simply avoiding the males. The aggression eventually stopped, and
the infant became a blackback in Group B. Evolutionarily, the death of the infant would not have markedly accelerated the
mother's return to estrus, but the death of the infant could still have benefitted the males, by decreasing the reproductive
output of a competitor. Adult male gorillas are also presumably selected to resist male immigration. Proximately, the aggression
directed towards the infant was not related to mating access to the mother. The sex of the infant may contribute to explain
the post-transfer male aggression, but data on the integration of old female infants in a new group is needed to test whether
the sex of the infant has an effect on their vulnerability to infanticide. Also, the intense fear displayed by the infant

On the resilience of remote traumatic memories against exposure therapy‐mediated attenuation 下载免费PDF全文
How to attenuate traumatic memories has long been the focus of intensive research efforts, as traumatic memories are extremely persistent and heavily impinge on the quality of life. Despite the fact that traumatic memories are often not readily amenable to immediate intervention, surprisingly few studies have investigated treatment options for remote traumata in animal models. The few that have unanimously concluded that exposure therapy‐based approaches, the most successful behavioral intervention for the attenuation of recent forms of traumata in humans, fail to effectively reduce remote fear memories. Here, we provide an overview of these animal studies with an emphasis on why remote traumatic memories might be refractory to behavioral interventions: A lack of neuroplasticity in brain areas relevant for learning and memory emerges as a common denominator of such resilience. We then outline the findings of a recent study in mice showing that by combining exposure therapy‐like approaches with small molecule inhibitors of histone deacetylases (HDACis), even remote memories can be persistently attenuated. This pharmacological intervention reinstated neuroplasticity to levels comparable to those found upon successful attenuation of recent memories. Thus, HDACis—or any other agent capable of heightening neuroplasticity—in conjunction with exposure therapy‐based treatments might constitute a promising approach to overcome remote traumata. 相似文献

We describe the behavioral repertoire of captive-born black tufted-eared marmosets (Callithrix penicillata) elicited by brief exposures to three potential mounted taxidermized predators (caracara hawk, Polyborus plancus; rattlesnake, Crotalus durissus; oncilla, Leopardus tigrina), and a stuffed toy. For each of the four stimuli, we submitted the subjects to a 9-min trial divided into three consecutive intervals: a 4-min pre-exposure baseline observation, a 1-min stimulus exposure, and a 4-min postexposure observation period. We positioned stimuli in front of each subject's home cage, and video-taped trials for behavioral analysis. During exposures to the potential taxidermized predators, we heard tsik-tsik vocalization and alarm behavior. After exposures, only the cat induced these reactions. All stimuli elicited observational reaction, albeit only during exposure intervals. Further comparisons between the three trial intervals indicated a decrease in the time spent in proximity to the cat during exposures, while an increase in proximity occurred when subjects were exposed to either the hawk or snake for the same period. Taken together, the behavioral responses during and after exposures to the taxidermized oncilla suggest that this stimulus is capable of inducing strong and persistent emotional reactions in Callithrix penicillata. 相似文献

Male rats (Rattus novergicus) infected with protozoan Toxoplasma gondii relinquish their innate aversion to the cat odours. This behavioural change is postulated to increase transmission of the parasite to its definitive felid hosts. Here, we show that the Toxoplasma gondii infection institutes an epigenetic change in the DNA methylation of the arginine vasopressin promoter in the medial amygdala of male rats. Infected animals exhibit hypomethylation of arginine vasopressin promoter, leading to greater expression of this nonapeptide. The infection also results in the greater activation of the vasopressinergic neurons after exposure to the cat odour. Furthermore, we show that loss of fear in the infected animals can be rescued by the systemic hypermethylation and recapitulated by directed hypomethylation in the medial amygdala. These results demonstrate an epigenetic proximate mechanism underlying the extended phenotype in the Rattus novergicus–Toxoplasma gondii association. 相似文献

ABSTRACTScientists are often reluctant to attribute emotions to nonhuman animals that are similar to human emotions. When the author published her early studies, reviewers prohibited the word fear. Fearful behavior had to be described as agitated. The core emotional systems described by Panksepp may provide a useful framework for people who work hands-on with animals. The core systems are fear, rage, panic (separation distress), seek, lust, nurture, and play. Some scientists who deny that animals have real emotions often fail to review important areas of the literature. The areas that are sometimes left out are the effects of psychiatric medications on animals and genetic influences on differences in animal behavior. In both people and animals, genetics has an influence on both fearfulness and novelty seeking. Visualizing the seven core emotional systems as separate volume controls on a music mixing board may help zoo professionals determine the motivation of both normal and abnormal behavior. It may also help them to design more effective environmental enrichments. 相似文献
