Among elephants, the phylogeographic patterns of mitochondrial (mt) and nuclear markers are often incongruent. One hypothesis attributes this to sex differences in dispersal and in the variance of reproductive success. We tested this hypothesis by examining the coalescent dates of genetic markers within elephantid lineages, predicting that lower dispersal and lower variance in reproductive success among females would have increased mtDNA relative to nuclear coalescent dates. We sequenced the mitochondrial genomes of two forest elephants, aligning them to mitogenomes of African savanna and Asian elephants, and of woolly mammoths, including the most divergent mitogenomes within each lineage. Using fossil calibrations, the divergence between African elephant F and S clade mitochondrial genomes (originating in forest and savanna elephant lineages, respectively) was estimated as 5.5 Ma. We estimated that the (African) ancestor of the mammoth and Asian elephant lineages diverged 6.0 Ma, indicating that four elephantid lineages had differentiated in Africa by the Miocene–Pliocene transition, concurrent with drier climates. The coalescent date for forest elephant mtDNAs was c. 2.4 Ma, suggesting that the decrease in tropical forest cover during the Pleistocene isolated distinct African forest elephant lineages. For all elephantid lineages, the ratio of mtDNA to nuclear coalescent dates was much greater than 0.25. This is consistent with the expectation that sex differences in dispersal and in variance of reproductive success would have increased the effective population size of mtDNA relative to nuclear markers in elephantids, contributing to the persistence of incongruent mtDNA phylogeographic patterns.  相似文献

Breeding chronology, harem structure and changes in male harem dominance were studied at Stranger Point, Isla 25 de Mayo/King George Island, principally by extensive field census work during the 2003 breeding season. Males were individually identified and their size estimated by using a photogrammetric method. Peak female haul out for the population occurred on 31 October, when a total of 276 females were observed along 7 km of coastline, distributed in ten harems with a median size of 16 females. Overall sex ratio and harem sex ratio for the breeding population were 1:6.7 and 1:10.6, respectively. A total of 33 males were identified associated with harems. Male size conferred an advantage in terms of dominance hierarchy, since dominant males (4.91±0.15 m) were significantly longer than subordinate males (4.63±0.19 m). Harems were dominated by an average of 4.5 (range 2–7) different males during the breeding season. Elephant seals at Stranger Point breed in very low density aggregations. The main breeding events in this population occurred later than at other breeding sites, which agrees with previous observations in the area. Male movement among harems suggests that differences in mating success among males could be achieved through their different behaviours.  相似文献

Seventy-seven immobilizations were carried out on adult male southern elephant seals at Stranger Point, Isla 25 de Mayo (King George Island) using a combination of Zoletil® (tiletamine and zolazepam) and ketamine in order to obtain biological samples. During 2006/2007, 22 males were immobilized at the beginning of their breeding period (EB), 19 of which were recaptured at the end of breeding (LB). Four were given only once at an unknown stage of breeding (USB) and 18 males were immobilized at the beginning of molting (BM). During 2007/2008, 14 adult males were immobilized at an USB. Zoletil® was administered using an automatic discharge device, whereas ketamine was injected directly with a syringe, and was used only when the initial sedation was not enough to carry out the programmed sampling. The initial mean dose of Zoletil® was 1,387 ± 304 mg, which represented 0.60 ± 0.14 mg/kg, range 0.36–1.05, n = 77. In 47 procedures, an average dose of 1.04 ± 0.66 mg/kg of ketamine was added. Mean immobilization time was 34 ± 14 min. In 25 out of the 77 procedures, males showed apnea, which lasted 8 ± 4 min (range 2–15 min). The necessary doses of Zoletil® and ketamine to attain immobilization differed between stages. For animals taken twice, doses (mg/kg) of Zoletil® and ketamine were significantly higher at the beginning than at the end of breeding. During molting, the doses of Zoletil® given were significantly lower than those used during breeding, although the proportion of animals that required ketamine during molting was significantly higher than during breeding. Zoletil® proved to be a safe immobilizing agent for field work on adult males of this species, given the wide range of doses used without any serious consequences. Furthermore, the addition of ketamine was useful when the initial sedation was not satisfactory or for prolonging the immobilization period in a practical and reliable way.  相似文献

This study suggests that the fruits of Sacoglottis gabonensis (Baill.) Urb. (Humiriaceae) are a keystone resource for forest elephants ( Loxodonta cyclotis Matschie) in a coastal rain forest, the Réserve de Faune du Petit Loango, Gabon (now part of Loango National Park). Faecal counts demonstrated that forest elephants used Sacoglottis -dominated forest more when Sacoglottis was abundant and electivity indices suggest that Sacoglottis is a preferred food. The flora of Petit Loango is characterized by the absence of herbaceous vegetation such as Marantaceae and Zingiberaceae, and during the prolonged dry season few fleshy fruits are present other than Sacoglottis fruits, which are produced in a glut during this time. While inter-annual fruiting reliability remains to be confirmed, fruit production in 1998 and high stem density relative to other study sites provide indirect evidence that Sacoglottis fruits are a reliable inter-annual resource at Petit Loango. It is thus proposed that Sacoglottis gabonensis fruits fulfil an important role as a keystone 'fallback' resource for forest elephants during the dry season at Petit Loango.  相似文献

We investigated the ranging patterns of elephants in the Marsabit protected area, north eastern Kenya, to ascertain the range of bachelor and female family herds in different seasons, and to identify corridor and noncorridor areas. Data were acquired for five bachelor and four female family herds equipped with satellite‐linked geographical positioning system collars, and monitored from December 2005 to December 2007. Distinct dry (about 260 km2) and wet seasons (about 910 km2) ranges were observed, with connecting corridors (north‐eastern corridor: about 90 km long, about 2‐7 km wide; southern corridors: about 10‐20 km long, about 2‐3 km wide). The dry season range corresponded with Marsabit evergreen forest, while the wet season range matched with dry deciduous lowland shrubs. The ranging elephants moved at speed of about 0.2‐20 kmh?1. Bachelor herds moved faster than female family herds. Elephants moved fast during the intermediate and wet seasons than during the dry season. The speed of ranging elephants was over 1 kmh?1 in the corridor areas and about 0.2 to less than 1 kmh?1 in the non‐corridor areas. Expansion of settlements towards corridor areas needs to be controlled to avoid future blocking of connectivity between wet and dry season elephant ranges.  相似文献

The analysis of linearity is a key aspect of the study of dominance hierarchies. To study the effect of the choice of socio-spatial level of analysis, we calculated linearity in a large set of southern elephant seal (Mirounga leonina) hierarchies from two populations (Valdés Peninsula and Falkland Islands). The socio-spatial level of analysis affects the observational effort, the completeness of matrices, and the frequency of unknown relationships. These factors, in turn, have a notable effect on linearity. We conclude that dominance should be studied at the local level, where the absence of structural zeros and the low incidence of observational zeros produce complete matrices, well rooted in the true spatial and social structure of the population. Depending on the specific social system, the extrapolation of dominance from the local level to higher levels may result in sparse matrices, and in biased estimates of linearity. The variation of the socio-spatial level of analysis may in part explain the contrasting results obtained in different studies of linearity of dominance hierarchies. Electronic Publication  相似文献

The Asian elephant (Elephas maximus) population in the wild has been in decline for several decades and breeding in captivity has not been self-sustaining. The use of artificial insemination (AI) can help overcome many of the difficulties associated with breeding elephants in captivity; however, the ability to store semen for extended periods of time is critical to the successful application of AI to elephants. The objective of the present study was to assess the effects of four different semen extenders and the presence of egg yolk on the viability and motility of Asian elephant semen stored at 4 °C. High quality ejaculates (n=4) were collected from two Asian elephant bulls by rectal massage. Aliquots of each ejaculate were extended in four different diluents (Beltsville thawing solution (BTS); Tris–citric acid (TCA)/fructose-based; Beltsville F5 (BF5); dextrose-supplemented phosphate-buffered saline (PBS)) with or without egg yolk then cooled and stored at 4 °C. The percentages of viable (viability) and motile (motility) sperm were evaluated at 8, 24 and 48 h following collection. The addition of egg yolk significantly reduced the percentage loss in viability from initial collection to 48 h compared to extenders without egg yolk (17.0±8.2 versus 32.6±8.9 decline in percent viable sperm in the population, respectively; P<0.05). Extender and egg yolk affected (P<0.005) total motility and percent progressively motile sperm at all evaluation times during incubation. TCA + egg yolk maintained higher (P<0.05) levels of progressive motility compared to other extenders supplemented with egg yolk. These results indicate that Asian elephant semen extended in TCA diluent supplemented with egg yolk can maintain at least 50% viability and motility when stored at 4 °C for 48 h.  相似文献

The duct system of the lactating mammary gland of the African elephant (Loxodonta africana) was investigated with histochemical and immunohistochemical techniques and with the transmission electron microscope in order to detect specific cell biological differentiations in the ductal epithelia of this species, which is marked by an unusually long lactation period. General histology and electron microscopy allow to distinguish several segments in the entire duct system. The apical membranes of the epithelia have binding sites for several lectins [Canavalia ensiformis agglutinin (ConA), Ricinus communis agglutinin (RCA I), Wisteria floribunda agglutinin (WFA), peanut agglutinin (PNA)] and also stain with alcian blue indicating the presence of a highly differentiated negatively charged glycocalyx forming an effective barrier between lumen and epithelium. Cytokeratins, actin, tubulin and vinculin show different expression intensities in the proximal and distal portion of the duct system. Lysozyme, lactoferrin, the secretory component of IgA and human beta defensin-2 are expressed in the epithelium of the entire duct system. In the distal portion of the ducts the staining intensity is stronger than in the proximal portion. We conclude that the duct system of the elephant mammary gland has specific secretory functions and that the secretory products are part of the defensive mechanisms against invading microorganisms.  相似文献

Some ecological factors that might potentially influence intestinal parasite loads in the Asian elephant (Elephas maximus Linn.) were investigated in the Nilgiris, southern India. Fresh dung samples from identified animals were analysed, and the number of eggs/g of dung used as an index of parasite load. Comparisons across seasons and habitats revealed that parasite loads were significantly higher during the dry season than the wet season, but were not different between the dry-deciduous and dry-thorn forests in either season. After accounting for the effect of age on body condition, there was no correlation between body condition, assessed visually using morphological criteria, and parasite load in either season. Individuals of different elephant herds were not characterized by distinct parasite communities in either season. When intra-individual variation was examined, samples collected from the same individual within a day differed significantly in egg densities, while the temporal variation over several weeks or months (within a season) was much less. Egg densities within dung piles were uniform, enabling a simpler collection method henceforth.  相似文献
