A mutant Had nl was induced in Drosophila melanogaster and found to be deficient in -hydroxy acid dehydrogenase. This mutation was utilized to study the genetics and physiological expression of Had +. Had+ was mapped to the X chromosome at 54.4 and seems to be the structural gene for the enzyme. Enzyme activity in male and female flies indicates that the gene shows both dosage compensation independent from dose effect and differential activity during ontogeny. Electrophoretic mobility data indicate that the enzyme is a dimer which forms by random association of subunits. The fact that the mutant shows no detrimental effect implies that the enzyme is dispensable, at least under laboratory conditions. The biological and technical implications of this gene-enzyme system are discussed.This research was sponsored by the Energy Research and Development Administration under contract with the Union Carbide Corporation. Mature seeds of diploid and tetraploidHibiscus species were analyzed for enzyme activity (alcohol dehydrogenase, malate dehydrogenase, leucine aminopeptidase), total protein content, DNA amount and dry weight. The recently formed tetraploid,H. radiatus, generally had enzyme and protein levels very similar to the sum of its progenitors, while the more ancient speciesH. acetosella had several lower levels. In Drosophila, the ratio of the number of X chromosomes to sets of other chromosomes initiates a series of events which result in sexual differentiation. In addition, this ratio establishes dosage compensation, a mechanism which equalizes the products of X-linked genes in males and females. Many plants react to pollutants in the atmosphere andcan consequentely be used to examine air quality.A distinction is made between bioindicator,bioaccumulator and hiperaccumulator and biomonitorplants.Given the encouraging results of a study to monitorstress caused by pollution in the city of Palermo(Sicily), we decided to carry out a survey ofpollutants, by examining some passive bioindicatorplants, which grow naturally or which had long agobeen planted in the city. The species chosen were:Eucaliptus camaldulensis Dehnh.; Pinuspinea L.; Platanus hybrida Brot, examples ofwhich are found all over the city.Biostructural and spectrophotometry analyses werecarried out on the leaf and pollen samples, payingparticular attention to lead. AIMS: The aerosolization and collection of submicrometre and ultrafine virus particles were studied with the objective of developing robust and accurate methodologies to study airborne viruses. METHODS AND RESULTS: The collection efficiencies of three sampling devices used to sample airborne biological particles - the All Glass Impinger 30, the SKC BioSampler and a frit bubbler - were evaluated for submicrometre and ultrafine virus particles. Test virus aerosol particles were produced by atomizing suspensions of single-stranded RNA and double-stranded DNA bacteriophages. Size distribution results show that the fraction of viruses present in typical aqueous virus suspensions is extremely low such that the presence of viruses has little effect on the particle size distribution of atomized suspensions. It has been found that none of the tested samplers are adequate in collecting submicrometre and ultrafine virus particles, with collection efficiencies for all samplers below 10% in the 30-100 nm size range. Plaque assays and particle counting measurements showed that all tested samplers have time-varying virus particle collection efficiencies. A method to determine the size distribution function of viable virus containing particles utilizing differential mobility selection was also developed. CONCLUSIONS: A combination of differential mobility analysis and traditional plaque assay techniques can be used to fully characterize airborne viruses. SIGNIFICANCE AND IMPACT OF THE STUDY: The data and methods presented here provide a fundamental basis for future studies of submicrometre and ultrafine airborne virus particles.  相似文献

Induced mutagenesis in callus tissues was studied in the medicinal plant Scilla indica irradiated with different doses of -radiation ranging from 2.5 to 20 Gy. Low doses accelerated the cell division and growth rate of the tissues whereas high doses repressed growth rate and resulted in lethality of tissues. Various cytological and chromosomal abnormalities were observed in the irradiated calli, the degree of which depended upon the dosage. Low doses of irradiation also promoted the regenerating capacity of the calli tissues and plants regenerating from them exhibited better growth and vigour compared to normal plants. High doses led to loss of regenerating capacity and promoted formation of malformed and stunted plants. Cytological study of regenerants revealed both diploid and mixoploid plants but no tetraploids were obtained.  相似文献

Recombinant glutathione S-transferase (agGST1-6) from the malaria vector mosquito Anopheles gambiae Giles (Diptera: Culicidae) was expressed in Escherichia coli using a pET3a vector system. The expressed enzyme was biochemically active with reduced glutathione (GSH) and 1-chloro-2,4-dinitrobenzene (CDNB). Activity of agGST1-6 with GSH and CDNB was inhibited to different degrees by both alpha-cyano and non-alpha-cyano pyrethroid insecticides. This inhibition was used to develop an assay for quantification of pyrethroids. Standard curves of insecticide concentration against percentage of enzyme inhibition or volume of iodine solution were established by spectrophotometry and iodine volumetric titration, respectively, for permethrin and deltamethrin. These assays allowed estimation of pyrethroid concentrations both spectrophotometrically and visually. For the residue assay of each insecticide, a cut-off point of 50% of the initial pyrethroid impregnation concentration was used, which should differentiate between biologically active and inactive treated bednets. The cross-reactivity of the primary permethrin photodegradants (3-phenoxyalcohol and 3-phenoxybenzoic acid) with the recombinant agGST1-6 was assayed in the same system. No agGST1-6 inhibition by the insecticide metabolites was observed, suggesting that the system is unaffected by primary permethrin metabolites and will accurately measure insecticide parent compound concentrations. The estimated pyrethroid insecticide concentrations, given spectrophotometrically and by iodine titration assay, were comparable to those obtained by direct HPLC quantification of residues extracted from bednets. Hence, it should be relatively easy to adapt this method to produce a test kit for residue quantification in the field.  相似文献

Although there are currently more than 30 antibody-drug conjugates (ADC) in clinical development for the treatment of blood cancers and solid tumors, comparison of their clinical pharmacokinetics (PK) is challenging because of the large number of, and differences between, the targets, ADC constructs, dosing regimens, and patient populations. In this review, we standardized the evaluation, using non-compartmental PK data reported at Cycle 1, i.e., following the first drug administration of what is usually a repeated-dose treatment, in monotherapy. We report ADC clinical PK properties, dosing regimen, determination of doses ranges and associated maximum tolerated doses. We also evaluated the effect of structural characteristics and target types (hematological vs. solid tumors) on PK. In addition, we discuss how integration of PK/pharmacodynamics approaches on top of classical dose escalation in first-in-human studies may improve dosing regimen determination for subsequent phases of clinical development.  相似文献
