The amino reagent 2,4,6-trinitrobenzenesulfonate (TNBS) was found to inactivate mitochondrial F1-ATPase through covalent labeling, which was not reversed by dithiothreitol. The observed rate of inactivation was retarded by inorganic phosphate, but enhanced by prior labeling of F1 with 7-chloro-4-nitrobenzo-2-oxa-1,3-diazole (NBD-C1). These observations are consistent with the presence of an essential amino group near the bound inorganic phosphate at the catalytic site of F1. A comparison of the observed protection of F1 from NBD-C1 and 5′-p-fluorosulfonyl-benzoyladenosine (FSBA) respectively by inorganic phosphate and by 2′,3′-O-(2,4,6-trinitrophenyl)adenosine 5′-monophosphate (TNP-AMP) suggests that NBD-C1 labels an essential Tyr residue in the positively charged locus for binding the polyphosphate end of ATP, and that FSBA labels an essential Tyr residue in the more hydrophobic locus for binding the adenosine moiety of ATP at the catalytic site of F1.  相似文献

The titration of metal-freed bovine α-lactalbumin with Mg2+ ions causes a two-stepped decrease in the tryptophan fluorescence quantum yield and a pronounced spectral shift towards shorter wavelengths, which seems to be a result of the binding of two magnesium ions to the protein molecule. The magnesium binding constants evaluated from the fluorimetric Mg2+-titration are 2·103 and 2·102 M?1. Mg2+ ions in millimolar concentrations almost do not influence the binding of Ca2+ ions to the protein.  相似文献

There have been numerous reports that liver of birds contain only isoenzymes of the low KM hexokinases, but lack the high KM glucokinase. We describe here the presence of glucokinase in livers of chicken and Japanese quail. The enzyme is membrane bound and is solubilized by vigorous mechanical disruption of the tissue. With gentle homogenization the glucokinase is recovered upon centrifugation in the 1000g pellet, from which it may be liberated by prolonged sonication. It appears to be localized in the cell plasma membrane. The activities of hexokinase and glucokinase appear to be about equal in liver parenchyma of fed chicken, but in that of Japanese quail the activity of glucokinase exceeds greatly that of hexokinase.  相似文献

The regulation of nucleotide excision repair and base excision repair by normal and repair deficient human cells was determined. Synchronous cultures of WI-38 normal diploid fibroblasts and Xeroderma pigmentosum fibroblasts (complementation group D) (XP-D) were used to investigate whether DNA repair pathways were modulated during the cell cycle. Two criteria were used: (1) unscheduled DNA synthesis (UDS) in the presence of hydroxyurea (HU) after exposure to UV light or after exposure to N-acetoxy-acetylaminofluorene (N-AcO-AAF) to quantitate nucleotide excision repair or UDS after exposure to methylmethane sulfonate (MMS) to measure base excision repair; (2) repair replication into parental DNA in the absence of HU after exposure to UV light. Nucleotide excision repair after UV irradiation was induced in WI-38 fibroblasts during the cell cycle reaching a maximum in cultures exposed 14–15 h after cell stimulation. Similar results were observed after exposure to N-AcO-AAF. DNA repair was increased 2–4-fold after UV exposure and was increased 3-fold after N-AcO-AAF exposure. In either instance nucleotide excision repair was sequentially stimulated prior to the enhancement of base excision repair which was stimulated prior to the induction of DNA replication. In contrast XP-D failed to induce nucleotide excision repair after UV irradiation at any interval in the cell cycle. However, base excision repair and DNA replication were stimulated comparable to that enhancement observed in WI-38 cells. The distinctive induction of nucleotide excision repair and base excision repair prior to the onset of DNA replication suggests that separate DNA repair complexes may be formed during the eucaryotic cell cycle.  相似文献

Guanylate cyclase activities are present in both soluble and particulate fractions of rat myometrial extract. Triton, slightly stimulated the soluble (50%) while markedly increasing (1000%) the particulate activity. Both fractions appear to be regulated independently. Predominantly, the soluble form was activated by sodium nitroprusside, involving interactions with SH-groups. On the other hand, the particulate form was stimulated by a series of unsaturated fatty acids and their hydroperoxides. The latter activation appears to result from direct hydrophobic effects rather than peroxide or free radical generation.  相似文献

Influx and efflux measurements of carnitine with submitochondrial particles lead to the conclusion that carnitine can cross the inner mitochondrial membrane by either facilitated diffusion or more rapidly by a carnitine-carnitine exchange. Both, the facilitated diffusion and the exchange are inhibited by N-ethylmaleimide or mersalyl at low concentrations. Reconstituted particles prepared from liposomes and either submitochondrial particles or an octyl β-glucoside-solubilized preparation were active in catalyzing carnitine-carnitine exchange.  相似文献
