Plodia interpunctella is a major economic pest that commonly infests most stored and processed agricultural products. Currently, heating at 50–60°C for at least 48 h is applied in facilities for disinfestation. However, this condition requires a great deal of time and expense. To improve the control efficiency of this system, we conducted combined treatments with heating and diatomaceous earth (DE), which is known to be toxic to pest insects. The DE effect was compared to heating at 25°C or 40°C to wandering fifth instar larvae, which is the stage most tolerant to heat. When larvae were brushed with DE powder, mortality was only 15.0–18.3% at 25°C for 10 days, but rapidly increased to 100% at 40°C within 4 h post‐treatment. In addition, when larvae were kept in a plastic cage with DE [4 mg/L (w/v)], their mortality was 100% in 24 h at 40°C post‐treatment; otherwise mortality was only 8.8% without DE. Thus, the control efficiency of heating significantly improved with the combination of DE. These effects increased further at higher temperatures and with longer exposure. Our results clearly showed that DE treatment showed synergistic effects with heating systems for the control of P. interpunctella.  相似文献

采用响应面分析法(RSM)对低温豆粕大豆分离蛋白(soybean protein isolated,SPI)的碱提工艺进行优化。在单因素实验的基础上,选取了影响SPI提取率的4个关键因素(pH、温度、时间和液料比)进行四因素五水平的中心组合旋转实验设计(CCRD)。通过RSM建立了响应值(SPI提取率)与各影响因素之间的回归方程,并获得了SPI的最优提取条件:pH 8.5,提取温度55℃,提取时间42.8 min,料水比1∶9.7(g/mL),此条件下SPI提取率预测值为37.12%,与实验值(36.69%)的误差为1.16%。将一次碱提后残渣进行二次碱提,二次提取率为10.16%。2次碱提上清液等电点沉淀的蛋白沉淀率分别为84.03%和85.84%。  相似文献

A meta-analysis was conducted (i) to evaluate broiler response to partial or total substitution of corn by sorghum and millet and (ii) to determine the effect of soybean meal replacement by cottonseed meal in broiler diet. The database included 190 treatments from 29 experiments published from 1990 to 2013. Bird responses to an experimental diet were calculated relative to the control (Experimental−Control), and were submitted to mixed-effect models. Results showed that diets containing millet led to similar performance as the corn-based ones for all parameters, whereas sorghum-based diets decreased growth performance. No major effect of the level of substitution was observed with millet or cottonseed meal. No effect of the level of substitution of sorghum on feed intake was found; however, growth performance decreased when the level of substitution of corn by sorghum increased. Cottonseed meal was substituted to soybean meal up to 40% and found to increase feed intake while reducing growth performance. Young birds were not more sensitive to these ingredients than older birds since there was no negative effect of these ingredients on performance in the starter phase. Results obtained for sorghum pointed out the necessity to find technological improvements that will increase the utilization of these feedstuffs in broiler diet. An additional work is scheduled to validate these statistical results in vivo and to evaluate the interactions induced with the simultaneous inclusions of sorghum, millet and cottonseed meal in broiler feeding.  相似文献

The variety of compounds present in chemically defined media as well as media supplements makes it difficult to use a mechanistic approach to study the effect of supplement composition on culture functionality. Typical supplements, such as soy protein hydrolysates contain peptides, amino acids, carbohydrates, isoflavones, and saponins. To study the relative contribution of these compound classes, a set of hydrolysates were produced, containing 58‐83% proteinaceous material and 5‐21% carbohydrates. While the content of the different compounds classes varied, the composition (e.g., peptide profiles, carbohydrate composition) did not vary in hydrolysates. The hydrolysates were supplemented to a chemically defined medium in cell culture, based on equal weight and on equal protein levels. The latter showed that an increase in the carbohydrate concentration significantly (P value < 0.004) increased integral viable cell density (IVCD) (R = 0.7) and decreased total IgG (R = ?0.7) and specific IgG production (R = ?0.9). The extrapolation of effects of protein concentration showed that an increase in protein concentration increased total and specific IgG production and suppressed IVCD. In addition to proteins and carbohydrates, the functionality of soy protein hydrolysates may be modulated by the presence of other minor compounds. In the current study, the large differences in the balance between total proteins and total carbohydrates in the supplemented media seem to be a main factor influencing the balance between the viable cell density, total IgG, and specific IgG production. © 2015 American Institute of Chemical Engineers Biotechnol. The purpose of the current research was to determine the impact of various biochar supplements on nutrient digestibility and growth performance of Catla catla fingerlings fed on Moringa oleifera seed meal (MOSM) based diet. An experiment with 90 days of feeding was conducted to investigate the efficacy of biochar obtained from different sources such as parthenium, farmyard manure, poultry waste, vegetable waste, and corncob waste at 2 mg/kg. There were15 fingerlings in each tank in the triplicate set of tanks. Fingerlings were fed at the rate of 5 % of their live wet weight. From each tank, feces were collected twice daily and stored to determine nutrient digestibility. Results showed that poultry waste biochar (test diet-IV) at 2 mg/kg was the best source to significantly (P < 0.05) improve weight gain % (256.58 %) and feed conversion ratio (1.19) than the other biochar sources and control diet. Furthermore, in terms of nutrient digestibility, optimum fat (81.90 %), protein (75.92 %), and gross energy (74.84 kcalg?1) values were obtained by using the same type of biochar (poultry waste). Conclusively, among all the five biochar sources, poultry waste biochar proved to be the best one, improving fish body performance.  相似文献

A laboratory strain of Aedes aegypti (L) was subjected repeatedly to larval selection pressure with two bacterial insecticides, spinosad (Saccharopolyspora spinosa) and bacilod (Bacillus thuringiensis israelensis). The results indicated that the mosquito Ae. aegypti acquired low resistance to spinosad and bacilod by about 3.1 and 2.4-fold, respectively, due to selection pressure for fifteen successive generations. The slope values of the selected strains were increased gradually from one generation to the next, indicating moderate homogeneity between individuals in their response to the test bio-insecticide. Moreover, larval selection with current bacterial bioinsecticides prolonged the time required to digest a blood meal. It showed an evident decrease in the reproductive potential of adult mosquitoes surviving selected larvae.  相似文献

In fall 1997, the toxic cyanobacterium Microcystis aeruginosa was documented in Lake Sammamish (western Washington, U.S.A.) for the first time. Cyanobacterial activity and environmental conditions that may promote toxic cyanobacteria were investigated during summer and fall 1999. Development of toxic Microcystis was hypothesized to be due to runoff of nutrients from the watershed (external loading hypothesis) or from vertical migration of dormant cyanobacteria from the nutrient-rich sediments into the water column (cyanobacterial migration hypothesis). Microcystins were detected using an enzyme-linked immunosorbent assay during late August and early September 1999 despite low cyanobacterial abundance. Microcystin concentrations ranged between 0.19–3.8 g l–1 throughout the lake and at all depths with the exception of the boat launch where concentrations reached 43 g l–1. Comparison of the conditions associated with the toxic episodes in 1997 and 1999 indicate that Microcystis is associated with a stable water column, increased surface total phosphorus concentrations (> 10 g l–1), surface temperatures greater than 22°C, high total nitrogen to phosphorus ratios (> 30), and increased water column transparency (up to 5.5 m). Migration of the cyanobacteria, Microcystis and Anabaena, occurred in both the deep and shallow portions of the lake. Microcystis dominated (89–99%) the migrating cyanobacteria with greater migration from the shallow station. External loading of nutrients due to the large rainfall preceding the 1997 toxic episode may have provided the nutrients needed to fuel that bloom. However, toxic Microcystis occurred in 1999 despite the lack of rain and subsequent external runoff. The migration of Microcystis from the nutrient-rich sediments may have been the inoculum for the toxic population detected in 1999.  相似文献

Raisins were a better source of carbohydrate than sucrose for reproduction by autogenous Culex pipiens. A blood meal increased the number of eggs per raft from 49 autogenously to 114. Eggs of aposymbiotic females produced autogenously did not hatch, but 34% of the eggs were viable if the mosquitoes fed on chickens. With repeated blood meals the number of eggs per raft and the rate of embryonation and hatching declined in each successive gonotrophic cycle. In about 14 of the unhatched eggs of normal females there were no fully developed embryos, while many more of the unhatched eggs of aposymbiotic females contained no evidence of embryonic development. After the fifth blood meal, neither normal nor aposymbiotic insects oviposited. The ovaries of the nulliparous females contained approximately 10% of the potential number of mature terminal oocytes. Proximally in the ovarioles there were dilatations and coiled tracheoles indicating egg resorption. There were fewer parous follicles in aposymbiotic than in normal females. Larval rearing water, i.e., infusion in which larvae had been reared, was more attractive than fresh infusion for oviposition by normal, autogenous mosquitoes. The degree of embryonation of the eggs was lower and the hatching success rate was poorer in fresh infusion than in larval rearing water.  相似文献

The molting process and body growth in Rhodnius prolixus (Hemiptera: Reduviidae) (Ståhl, 1859) are significantly influenced by the availability and quality of food. Based on the body weight of each stage, the present study provides estimates of a potential critical weight threshold required for molt initiation in R. prolixus. In addition, a new measure given by the area under the weight curve is proposed, which encapsulates both body weight and time. It is shown that this measure is consistent with the data, and allows the estimation of a pre‐refractory period (i.e. the time interval between the moment at which the critical weight threshold is reached and the moment when no further meals are accepted). The present analysis estimates the critical weight threshold as 1.6, 5.3, 12.9, 42.0 and 97.0 mg for stages 1–5, respectively, whereas the values of the area under the curve threshold as 5, 16, 31.2, 159.7 and 329.9 mg days for stages 1–5, respectively. The results of the present study confirm the existence of a weight‐dependent mechanism for the initiation of molting in R. prolixus.  相似文献
