Trichosporon beigelii SBUG 752 was able to transform diphenyl ether. By TLC, HPLC, GC, GC-MS, NMR- and UV-spectroscopy, several oxidation products were identified. The primary attack was initiated by a monooxygenation step, resulting in the formation of 4-hydroxydiphenyl ether, 2-hydroxydiphenyl ether and 3-hydroxydiphenyl ether (48:47:5). Further oxidation led to 3,4-dihydroxydiphenyl ether. As a characteristic product resulting from the cleavage of an aromatic ring, the lactone of 2-hydroxy-4-phenoxymuconic acid was identified. The possible mechanism of ring cleavage to yield this metabolite is discussed. 相似文献

The capacity of chitin (from crab shells) and of fungal cell walls from Trichoderma harzianum to accumulate zinc, cadmium and mercury was studied as well as the effects of adsorbed metals on the enzymatic hydrolysis by Novozym 234 of the two substrates. The total adsorbing capacity with respect to these metals was estimated to be at least 10 mmol kg–1 chitin (dry weight) and 50 mmol kg–1 fungal cell walls (dry weight), respectively, at pH 6.1. Enzymatic digestion of fungal cell walls preloaded with mercury and cadmium was significantly reduced, while zinc did not cause any significant inhibition. The effect of metal complexation by chitin on the enzymatic digestion was not as pronounced as for fungal cell walls. This could reflect the fact that chitin sorbed a lower total amount of metals. The inhibitory effect of metals on the enzymatic hydrolysis was caused by the association of the metals with the two substrates and not by the presence of free metals in solution. 相似文献

The physiological role of VIP in the liver is controversial. VIP receptors are present, but their function in the metabolic regulation is uncertain. The interaction of porcine VIP with isolated cells from pig liver was studied with respect to receptor-binding, degradation and glycogenolytic action. In this model, VIP and liver showed homology of animal species. 1. Receptor-binding was heterogenous with Kd values of 10(-9) mol/l and 4 X 10(-8) mol/l, and a total amount of binding sites of 7 X 10(-11) mol per 10(9) cells. The peptide specificity showed that porcine and chicken VIP were equally potent in inhibiting receptor-bound 125I-VIP; secretin was about 30 times less potent; glucagon and somatostatin were ineffective. 2. Receptor-bound 125I-VIP was degraded since about 70% was released as radioactivity not reacting with VIP-antiserum. 3. Glucose-release was not stimulated by VIP (10(-6) mol/l) whereas the rate was increased two-fold by glucagon (10(-6) mol/l). In conclusion, VIP receptors in pig liver cells are different from other tissues regarding peptide specificity. Fractional rates (% · day–1) of synthesis and degradation were determined by measuring the output of N-methylhistidine (MeHis) in the excreta at 4 and 8 weeks of age in the chicken. At 4 weeks of age, the fractional rate of synthesis of the meat-type stock was twice that of the egg-type stock (White Leghorn), but the fractional rates of synthesis at 8 weeks of age were similar (4.1–5.1% · day–1) among stocks. The fractional rate of degradation (1.3–1.5% · day–1) of the meat-type stock at 8 weeks of age was less than half the rate of the egg-type stock (2.9% · day–1). The fractional rates of synthesis and degradation at 4 weeks of age in the Satsuma native fowl were relatively high compared with those in the other stocks. In particular, the rate of degradation (8.6% · day–1) at 4 weeks of age was approximately twice that of other stocks. These results show that fractional rates of synthesis and degradation of muscle protein in the chicken differ among genetically diverse groups. A new apparatus for electrophoretic transfer of proteins from micro polyacrylamide slab gels has been developed. The apparatus enabled the easy changing of nitrocellulose sheets and was suited for obtaining multiple blots from a gel. Electrophoretic conditions were determined so that all of the blots obtained sequentially from one slab gel were successfully used to visualize specific proteins irrespective of their molecular weight. Combining the transfer technique with the technique of parallel micro two-dimensional electrophoresis, 20 blots could be obtained within 1 h of electroblotting time. The locations of 28 human serum proteins were determined simultaneously on these blots using commercial specific antisera. 相似文献

Andean cultural ecologists have made two claims in recent years: ecological decomposition is absent due to effective indigenous management of communal resources, and agricultural intensification is inversely related to altitude. Drawing on material from the Jukumani Indians of Northern Potosi, Bolivia, these assertions are challenged. First, there is little evidence to prove or disprove ecological degradation. Second, the location of agricultural intensification, as the Jukumani data suggests, is influenced by altitude as well as by the presence of market.Fieldwork in Bolivia was carried out between January 1979 and May 1981. Abstract Escherichia coli was cultured with a low concentration of dihydrostreptomycin (2.5 μg/ml.). Growth was similar to untreated controls for 10 h after which a slow decline in growth rate occurred; growth ceased after 20 h. Intracellular catabolism of pulse-labelled protein synthesised at various points during the antibiotic treatment increased during the first 10 h, but during the second 10 h proteolysis progressively declined to almost control values. New gene selection techniques (Recombinant DNA) are currently available to exploit useful properties of various biological systems hitherto regarded as interesting but of little or no immediate commercial value. The application of genetic engineering techniques to problems in the Pulp and Paper Industry are many. As a first step these techniques are being used to provide much needed fundamental information on the cellular and molecular mechanisms involved in the expression of extra-cellular enzymes that degrade lignocellulosic pulping wastes. The information gleaned from the studies on cellulolytic fungi and bacteria can be used to genetically engineer a yeast or bacterium capable of converting pulping wastes into ethanol and other useful by-products. 相似文献

Summary In the spider Dinopis, retinae of the posterior median eyes synthesise new photoreceptor membrane in bulk at dusk and destroy it at dawn (Blest, 1978). During the dawn period, there is a rapid, anticipatory differentiation of unusual organelles from the rough endoplasmic reticulum (RER) in the intermediate segments of the receptors. This system is classified as GERL. Its products appear to play a role in the autolysis of photoreceptor membrane. Consistent topographical relationship to Golgi bodies has not been determined. Circumscribed regions of RER whorls first reorganise to yield fenestrated profiles; these differentiate further to a number of structures by condensation and loss of ribosomes. Smooth tubular profiles are termed rigid tubules to indicate their probable homology with the rigid lamellae of vertebrate secretory cells. More complex smooth multilocular bodies are also produced. Evidence is discussed which implies that both rigid tubules and multilocular bodies give rise to condensing vacuoles. These, in turn, pinch off coated vesicles assembled as Nebenkerne. Some rigid tubules are transported to the interrhabdomeral cytoplasm of the receptive segments. At late stages of differentiation, rigid tubules, multilocular bodies and Nebenkerne give strong, positive responses to zinc iodide-osmium tetroxide (ZIO) treatment; early stages and both cis and trans Golgi components do not. GERL differentiation is independent of immediate illumination of the retina at dawn. It is suggested to mediate the lysis of membrane degradation products by the production of hydrolases.We thank Professor D.T. Transfer of Lemna minor fronds to culture medium containing 50% (v/v) deuterium oxide induces a large increase in the rate of protein breakdown, which is not due to an increase in the activity of acidic or neutral proteolytic enzymes or peptidases. Biochemical and ultrastructural evidence indicates that deuterium oxide affects the properties of certain membranes, particularly the tonoplast, and allows vacuolar proteolytic enzymes to pass into the cytoplasm and cause the increased protein breakdown.Abbreviations BAPA
