cDNA clones encoding a novel proline-rich protein (NaPRP4) have been isolated from a Nicotiana alata stylar cDNA library. The N-terminal part of the derived protein is highly rich in proline (32.2%) and contains several repeats such as Lys-Pro-Pro (7 times) and Pro-Thr-Lys-Pro-Pro-Thr-Tyr-Ser-Pro-Ser-Lys-Pro-Pro (twice); the C-terminal part, on the other hand, has a lower proline content (9.9%) and contains two potential N-glycosylation sites and all the six cysteine residues. Northern blot and in situ hybridisation analyses indicate that expression of the NaPRP4 gene is restricted to cells of the transmitting tract of the style.  相似文献

Summary Changes in the plasma membrane surface and in the cortical cytoplasm during wound healing in giant green algal cells ofErnodesmis verticillata (Kützing) Brgesen were followed using scanning and transmission electron microscopy. Microvillus-like structures that contain cytoplasmic and cytoskeletal constituents were observed emanating from the surface of the plasma membrane at the retracting/cut end of wounded cells. These delicate structures seem to be remnants of cell wall-plasmalemma connections that draw out the plasma membrane and cortical components from the contracting cytoplasm as it pulls away from the cell wall. Most of these connections break during wound healing and, when contraction stops, the microvillus-like protrusions become progressively shorter. In cells treated with a calmodulin antagonist (W-7), a number of distinctive bodies accumulate that are of unknown composition, are oblong in shape, and have a diameter slightly smaller than the protoplasmic protrusions. Ultrastructural and other data indicate that these bodies result from retrieved constituents of the plasma-membrane protrusions, as they do not accumulate in unwounded drugtreated cells or in cells treated in W-5. These findings suggest that the protoplasmic protrusions accumulate membrane and cytoplasmic components that are retrieved and recycled during wound healing inErnodesmis by a novel mechanism. In Arabidopsis, phosphate starvation (-Pi)-induced responses of primary root and lateral root growth are documented to be correlated with ambient iron (Fe) status. However, whether and how Fe participates in -Pi-induced root hair growth (RHG) remains unclear. Here, responses of RHG to different Fe concentrations under Pi sufficiency/deficiency were verified. Generally, distinct dosage effects of Fe on RHG appeared at both Pi levels, due to the generation of reactive oxygen species. Following analyses using auxin mutants and the phr1 mutant revealed that auxin and the central regulator PHR1 are required for Fe-triggered RHG under −Pi. A further proteomic study indicated that processes of vesicle trafficking and auxin synthesis and transport were affected by Fe under −Pi, which were subsequently validated by using a vesicle trafficking inhibitor, brefeldin A, and an auxin reporter, R2D2. Moreover, vesicle trafficking-mediated recycling of PIN2, an auxin efflux transporter, was notably affected by Fe under -Pi. Correspondingly, root hairs of pin2 mutant displayed attenuated responses to Fe under -Pi. Together, we propose that Fe affects auxin signalling probably by modulating vesicle trafficking, chiefly the PIN2 recycling, which might work jointly with PHR1 on modulating -Pi-induced RHG.  相似文献

Recycling of cellulases should lower the overall cost of lignocellulosiic bioconversion processes. In this study, three recycling strategies were evaluated to determine their efficiencies over five successive rounds of hydrolysis. The effect of lignin on recycling was examined by comparing water-washed, steam-exploded birch (WB; 32% lignin) and WB which had been further extracted with alkali and peroxide (PB; 4% lignin). When the cellulases were recovered from the residual substrates after partial hydrolysis of both substrates, the recovered cellulase activity toward the mixture of fresh and residual substrates decreased after each recycling step. When the cellulases in the supernatants were also recycled, up to 20% more activity could be recovered. In both of these cases, the recovered activities did not correspond to the activities expected from the amount of cellulase protein recovered during recycling. The best recovery was obtained when the cellulases were recovered from both the residue and the supernatant after complete hydrolysis of the PB substrate. In this case, all of the originally added cellulase activity could be recovered for four consecutive hydrolysis rounds. However, when the same recycling strategy was carried out using the WB substrate, the recovered cellulase activity declined quickly with each recycling round. An enclosure experiment was conducted to assess the effects of a zooplankton elimination on the structure of a phytoplankton community. Phytoplankton biomass and production were higher in grazer-free enclosures, while the productivity per unit biovolume was lower. Exclusion of zooplankton favoured the majority of algal species, especially chrysophyceans (Dinobryon spp.) and the diatom Rhizosolenia, while mucilagineous green-algae were disfavoured. Middle sized algae (ESD 15–50 µm) and those with the largest Surface Area/Volume ratio were proportionally most favoured by the elimination of grazers.These differences in phytoplankton community structure are discussed in relation to effects of direct selective grazing and nutrient recycling by zooplankton. Some differences, as the immediate positive response of Dinobryon and Rhizosolenia, are probably caused by grazing release, while others, e.g. the response of mucilagineous species, might be caused by changed competitive relationships between the algae.  相似文献

Aerobic mineralization, i.e. seston respiration, microbial breakdown of detritus and microbial assimilation-dissimilation of photosynthetically derived D(issolved) O(rganic) C(arbon) was measured in concentrated samples from the pelagic zone of Lake Vechten. The samples were described by cell numbers of dominant algae prior to concentrating by centrifuge with continuous rotor. The concentrated samples were incubated in the laboratory at in situ temperature and a light intensity of 30 W · m–2 for measuring primary production and photosynthetically derived DOC. After filtration the particulate fraction was incubated in unlabelled lake water far measuring respiratory production of DI14C. Portions of the same particulate fraction were sterilized and incubated in unlabelled sample concentrate for measuring microbial breakdown of detritus. The seston respiration amounted to 19–30% loss of the particulate fraction over 15–45 h. Microbial breakdown of detritus amounted to 28–40% loss of the particulate fraction over 24–168 h. In both cases P(articulate) O(rganic) 14C(arbon) was transformed to mainly DI14C. Microbial assimilation-dissimilatioh of photosynthetically derived DOC could not be measured reliably.  相似文献

The relative importance of respiration and organic carbon release to the efficiency of carbon specific growth of Skeletonema costatum (Grev.) Clave was evaluated over a light range from 1500–15 μE · m?2· s?1. Net growth efficiency ranged from 0.45–0.69 with a maximum at 130 μE · m?2· s?1. Respiration was 93% or more of the variations in growth efficiency. Organic carbon release ranged from 0–7% of gross production and increased with light intensity. Carbon specific particulate production was a hyperbolic function of incident light intensity and was related exponentially to particulate carbon production per unit chlorophyll a. Full sunlight conditions, 1500 μE · m?2· s?1, did not induce photoinhibition of gross production. Variations in the efficiency of growth of S. costatum were minimized over a wide range of light intensities mainly because of variations in cellular pigments which permitted the efficient utilization of available light energy, and a reduction in the losses of carbon which increases the growth rate, possibly as a consequence of the recycling of respired carbon within the cell.  相似文献
