为探究中国沙棘对土壤镉(Cd)胁迫的性别响应差异,该研究以中国沙棘2年生幼苗为材料,利用盆栽试验研究在不同浓度Cd处理下(0(CK)、25、50、100和200 mg·kg-1)雌、雄株幼苗的生长、叶片生理特性以及Cd富集特征的差异。结果表明:(1)Cd处理下中国沙棘幼苗雌、雄株的株高和基径生长以及各器官生物量均表现出低浓度(<50 mg·kg-1)促进,高浓度(> 100 mg·kg-1)抑制的现象;低浓度Cd处理下雌株的株高、基径增长率和生物量的增幅均高于雄株;高浓度Cd处理下(200 mg·kg-1)雄株株高增长率、叶生物量和总生物量分别较CK显著降低,而雌株均未显著下降。(2)随着Cd浓度升高,雌、雄株叶片光合色素含量和抗氧化酶活性呈先升后降的变化趋势,丙二醛(MDA)和渗透调节物质含量呈上升趋势;Cd浓度为50~200 mg·kg-1时,雌株叶片的光合色素含量、抗氧化酶活性和渗透调节物质含量均高于雄株,而MDA含量始终低于雄株。(3)随着Cd浓度升高,雌、... 相似文献

陕西佛坪国家级自然保护区位于秦岭南坡的中段 ,1998~ 1999年冬季通过分层抽样的方法 ,测定了保护区内羚牛秦岭亚种 (Budorcastaxicolorbedfordi) 10种栖息地类型的单位面积食物资源量和含有的能量 ,估计了越冬区可以利用的总能量。结果表明 ,羚牛越冬区提供的可供羚牛和其它草食动物利用的能量为 (9 877±5 173)× 10 12 J,每只羚牛平均每天需要的能量为 2 6 4 8± 11 88kJ ,整个冬季需要的能量为 4 82 0± 2 16 3× 10 6J ,保护区的羚牛容纳量为 2 0 4 9± 10 37只。考虑到羚牛对空间的需求、保持最佳繁殖状态、其它草食动物种群的需求和保护当地植被等因素 ,建议保护区将羚牛种群的数量控制在 10 0 0只左右. 相似文献

The effects of spatial structure in terms of local capacity, or the maximum number of larvae surviving competition at resource patches, and temporal structure in terms of the period vulnerable to parasitoid attack in host populations on the persistence of host-parasitoid systems were quantitatively evaluated by laboratory experiments and well-parameterized model analyses. One of two bruchid beetles,Callosobruchus maculatus andC. phaseoli, were used as a host with Heterospilus prosopidis used as the parasitoid.C. maculatus, in which few larvae survive competition to become adults in each bean, andC. phaseoli, in which many larvae become adults in each bean, along with two kinds of beans, the mung and the azuki, were combined to construct four (2×2) resource-herbivorous host-parasitoid systems that differed in local capacity and vulnerable period. The mung-C. maculatus system with the parasitoid was the most persistent, i.e., took the longest time for extinction of either the host or parasitoid to occur. Since this resource-herbivorous host combination exhibited the lowest local capacity and the shortest vulnerable period, these two conditions possibly promoted the persistence of the system. A model incorporating the host population structure supported the observed persistence. Manipulation of quality and mineralization of tropical legume tree prunings by varying nitrogen supply 总被引:5,自引:1,他引:4
The effect of N supply on the quality of Calliandra calothyrsus and Gliricidia sepium prunings was studied in a glasshouse over a 7-month growing period. Increasing the concentration of N supplied from 0.625 to 10.0 mM NO3-N resulted in increased N concentration but decreased polyphenol concentration, protein-binding capacity and C:N ratio of prunings from both species. Lignin concentration was not consistently altered by the N treatment. Mineralization of N from the prunings was measured over a 14-week period under controlled leaching and non-leaching conditions. The results indicated a strong interaction between legume species and concentration of N supply in their influence on N mineralization of the prunings applied to the soil. Differences in the %N mineralized were dictated by the quality of the prunings. The (lignin + polyphenol):N ratio was the pruning quality factor which could be used most consistently and accurately to predict N mineralization of the legume prunings incubated under leaching conditions, and the relationship was best described by a linear regression. Under non-leaching conditions, however, the protein-binding capacity appeared to be the most important parameter in determining the patterns of N release from the prunings studied. The relationship between the N mineralization rate constant and the protein-binding capacity was best described by a negative exponential function, y=0.078 exp(–0.0083x). The present study also indicated that the release of N from legume prunings containing a relatively high amount of polyphenol could be enhanced by governing the N availability conditions under which the plant is grown, for example whether or not it is actively fixing nitrogen. Estimates of pruning N mineralization after 14 weeks with the difference method averaged 6% (leaching conditions) and 22% (nonleaching conditions) more than with the 15N method for all legume prunings studied. The recovery of pruning by maize (4–38%) was well correlated with the % pruning N mineralized suggesting that incubation data closely reflect the pruning N value for a given catch crop under non-leaching conditions. 相似文献

The function of the supra-orbital salt gland was studied in the common eider duck (Somateria mollissima). The maximum salt-secreting capacity was determined in (1) wild ducks which had been living in a marine environment, (2) ducks reared in captivity on fresh water, and (3) ducks from group 2 adapted to salt water. The maximum secreting capacity was found by infusing a solution of NaCl (1000 mosmol·kg-1) at increasing rates, from 0.691 to 1.671 mosmol·min-1. Freshwater-adapted ducks secreted at a maximum rate of 0.785 mosmol·min-1 (1500 mosmol·kg-1). Adapted to salt water they increased their capacity, and the best duck secreted at a rate of 1215 mosmol·min-1 (1600 mosmol·kg-1). The best wild duck secreted at a rate of 1516 mosmol·min-1. Ducks in group 3 were used to examine the response to a hyperosmotic or an isoosmotic infusion. The amount of salt (NaCl) given per unit time was the same. Given a hyperosmotic solution their salt glands secreted at a high rate: 30 min after the infusion had stopped the ducks had excreted 94% of the sodium infused, 92.9% via the salt gland. Given an isoosmotic solution they secreted at a rate about half the infusion rate: 30 min after cessation of infusion they had excreted 73% of the sodium, 42.9% via the salt gland and the rest by the kidneys.Abbreviations

The maltose binding protein (MBP or MalE) of Escherichia coli is the periplasmic component of the transport system for malto-oligosaccharides. It is used widely as a carrier protein for the production of recombinant fusion proteins. The melting of recombinant MBP was studied by differential scanning and titration calorimetry and fluorescence spectroscopy under different solvent conditions. MBP exhibits a single peak of heat absorption with a delta(Hcal)/delta(HvH) ratio in the range of 1.3-1.5, suggesting that the protein comprises two strongly interacting thermodynamic domains. Binding of maltose resulted in elevation of the Tm by 8-15 degrees C, depending of pH. The presence of ligand at neutral pH, in addition to shifting the melting process to higher temperature, caused it to become more cooperative. The delta(Hcal)/delta(HvH) ratio decreased to unity, indicating that the two domains melt together in a single two-state transition. This ligand-induced merging of the two domains appears to occur only at neutral pH, because at low pH maltose simply stabilized MBP and did not cause a decrease of the delta(Hcal)/delta(HvH) ratio. Binding of maltose to MBP is characterized by very low enthalpy changes, approximately -1 kcal/mol. The melting of MBP is accompanied by an exceptionally large change in heat capacity. 0.16 cal/K-g, which is consistent with the high amount of nonpolar surface--0.72 A2/g--that becomes accessible to solvent in the unfolded state. The high value of delta Cp determines a very steep delta G versus T profile for this protein and predicts that cold denaturation should occur above freezing temperatures. Evidence for this was provided by changes in fluorescence intensity upon cooling the protein. A sigmoidal cooperative transition with a midpoint near 5 degrees C was observed when MBP was cooled at low pH. Analysis of the melting of several fusion proteins containing MBP illustrated the feasibility of assessing the folding integrity of recombinant products prior to separating them from the MBP carrier protein. 相似文献

We examined the effect of differences in exercise intensity on the time constant (t
c) of phosphocreatine (PCr) resynthesis after exercise and the relationships betweent
c and maximal oxygen uptake (VO2max) in endurance-trained runners (n = 5) and untrained controls (n = 7) (average VO2max = 66.2 and 52.0 ml · min–1 · kg–1, respectively). To measure the metabolism of the quadriceps muscle using phosphorus nuclear magnetic resonance spectroscopy, we developed a device which allowed knee extension exercise inside a magnet. All the subjects performed four types of exercise: light, moderate, severe and exhausting. The end-exercise PCr: [PCr + inorganic phosphate (Pi)] ratio decreased significantly with the increase in the exercise intensity (P < 0.01). Although there was little difference in the end-exercise pH, adenosine diphosphate concentration ([ADP]) and the lowest intracellular pH during recovery between light and moderate exercise, significant changes were found at the two higher intensities (P < 0.01). These changes for runners were smaller than those for the controls (P < 0.05). The
c remained constant after light and moderate exercise and then lengthened in proportion to the increase in intensity (P < 0.05). The runners had a lowert
c at the same PCr and pH than the controls, particularly at the higher intensity (P < 0.05). There was a significant correlation betweent

The biodiversity sciences represent the disciplines of whole-organism biology, including systematics, ecology, population biology, behaviour and the fields of comparative biology. The biodiversity sciences are critically important to society because it is knowledge of whole-organisms that is essential for managing and conserving the world's species. Because of an acceleration in environmental degradation and global biodiversity loss in recent decades, the need for the biodiversity sciences has never been more urgent. Yet, biodiversity science is not well supported relative to other fields of science, and thus the need for knowledge about organisms and their environment is far outstripping biologists' ability to provide it. National and international capacity for biodiversity science must therefore be increased substantially. Each nation should establish a national biodiversity research programme coordinated across all government agencies. An international biodiversity research programme should also be established, perhaps with an organizational structure that parallels the International Geosphere-Biosphere Programme. Biodiversity scientists must assume a leadership role in educating the public and bringing about policy changes that will enhance our understanding of the world's species and their ecosystems. 相似文献

The dielectric dispersion of isolated intact mitochondria in suspension has been measured between 10 kHz and 500 MHz. In isotonic KCI media at 4°C, the mitochondria maintained their characteristic ‘double membrane’ structure as examined by electron microscopy, and the observed dispersion curves were successfully simulated in terms of a superposition of two sub-dispersions having different characteristic frequencies and different permittivity magnitudes. Taking these observations into account we analyzed the dispersion data on the basis of a ‘double-shell’ model in which two concentric shells are meant to represent the mitochondrial outer and inner membranes. The analyses by a computerized curve-fitting method revealed that: (i) electric capacities for the outer and the inner membrane are 1.7 and 0.5 μF/cm2, respectively, (ii) relative permittivity for the inner compartment (or the equivalent homogeneous matrical space) = 50–60, (iii) outer compartment-to-external conductivity ratio = 0.4–0.6, and (iv) inner compartment-to-external conductivity ratio = 0.14. The implications of these parameter values are discussed with due attention paid to the limitations inherent in our ‘double-shell’ model approach. 相似文献
