 A procedure for producing transgenic Chinese cabbage plants by inoculating cotyledonary explants with Agrobacterium tumefaciens strain EHA101 carrying a binary vector pIG121Hm, which contains kanamycin-resistance and hygromycin-resistance genes and the GUS reporter gene, is described. Infection was most effective (highest infection frequency) when explants were infected with Agrobacterium for 15 min and co-cultivated for 3 days in co-cultivation medium at pH 5.2 supplemented with 10 mg/l acetosyringone. Transgenic plants of all three cultivars used were obtained with frequencies of 1.6–2.7% when the explants were regenerated in shoot regeneration medium solidified with 1.6% agar. A histochemical GUS assay and PCR and Southern blot analyses confirmed that transformation had occurred. Cauliflower curd is composed structurally of a number of shortened shoots on which there are tremendous amount of naked apical meristems. The authors isolated four pure lines of different curd types from cauliflower (Brassica oleracea L. var. botrytis) accessions: smooth curd (curd-s), haired curd (curd-h), crystal curd (curd-c) and thorned curd (curd-t). BobCAL genes was cloned separately from each pure line by PCR amplification and a termination mutation from AAG to TAG was found in the fifth exon of all four sequences, whereas DNA sequences of 638 bp upstream of the stop codons were identical, demonstrating that BobCAL is not the sole regulator of flower buds and cauline on curd. After crosses of cauliflower with cabbage (B.oleracea var. capitata) or broccoli (B.oleracea var. italica), the resultant F1 plants failed to form curd and had the diversity of inflorescence which implicates the relatedness of CAL to other regulators. One homologous gene of CAL, BcpCAL was isolated from Chinese cabbage (B.campestris L. ssp. pekinensis) and it was found not to have the termination mutation in the fifth exon, and thus the structure of BcpCAL gene was analyzed. The results offer molecular and genetic evidence for the study of biological function of CAL and morphological genetics of curd.  相似文献

The spatio-temporal distribution of Ceutorhynchus assimilis Payk. (Coleoptera: Curculionidae) adults caught in a rectangular grid of flight traps in a crop of winter oilseed rape (Brassica napus L.) was mapped and was analysed using Spatial Analysis by Distance IndicEs (SADIE). Their distribution was compared to that of their larvae and that of their parasitoid Trichomalus perfectus (Walker) (Hymenoptera: Pteromalidae) in pods. The distribution of immigrating C. assimilis adults was consistent with their arrival at the crop boundaries and movement within the crop towards its centre. Adult C. assimilis were aggregated at all times, invasion being on two fronts, leading to the formation of two major clusters within the crop. Large areas of the crop remained relatively unpopulated. During the emigration phase, numbers declined simultaneously in all parts of the crop. The distributions of adult and larval C. assimilis and of larval T. perfectus were spatially associated. The distribution of the parasitoid did not show a density dependent relationship with that of its host. We discuss the movements of insects which underlie their population distributions, the value of integrating spatial information into improved management strategies for C. assimilis and the potential for the spatial targeting of insecticides to reduce the amount applied and to conserve T. perfectus.  相似文献

Cross-mating was performed between subspecies of cabbage butterfly Pieris rapae crucivora from Japan and P. rapae rapae from New Zealand, whose females are known to reflect and absorb ultraviolet (UV) light, respectively. In the female F 1 hybrids UV reflectance was intermediate between the two parent females. Further, the UV reflectance was significantly higher in the female F 1 hybrids derived from females of the New Zealand subspecies than in those derived from females of the Japanese subspecies.  相似文献

We studied the effect of different host plants and rearing atmosphere on life cycle of cabbage butterfly, Pieris brassicae. Insects were reared in the field (fluctuating weather) as well as in the laboratory (constant rearing conditions) on four host plants, viz. cabbage, cauliflower, knol-khol and broccoli. Significant differences were not found in the incubation and pupal period of butterfly reared on different host plants. However, larval period was found to be significantly lower on cabbage followed by knol-khol and highest on broccoli. Therefore, the developmental period (from eggs to adult) was reasonably lower on cabbage. Furthermore, inverse relationship was found in the body weight of butterfly and developmental period, where weight of full grown caterpillar and pupae was significantly higher on cauliflower and lowest on cabbage. Besides, significant differences were not found in the body weight of P. brassicae caterpillar reared under field and in the laboratory. Nevertheless, pupal weight of butterfly was significantly higher under field conditions than the laboratory conditions. Overall, the development of P. brassicae was much faster on cabbage than other hosts; but its body weight was considerably higher on cauliflower.  相似文献

Abstract. Drought avoidance due to cuticular control increases with leaf number to a maximum in the intermediate leaves, decreasing to a minimum in the upper leaves. Dehydrated intermediate leaves do not rehydrate detectably when floated on water for several days. Excision of their petioles when submerged, permits full rehydration, presumably via the xylem. Stressing the plant by withholding water for 1–3 weeks fails to increase this already high resistance to water movement through the leaf surface. It does, however, markedly decrease the loss of water from the fully rehydrated (100% RWC) leaves during the first hour of dehydration, presumably due to a more rapid stomatal closure than in the non-stressed leaves. Dehydration tolerance increases with leaf number, without an intermediate maximum. The intermediate and upper leaves were markedly more tolerant of dehydration after drought-induced stress than when non-stressed. Dehydration tolerance in some cases, was inversely proportional to dehydration rate. It was possible, however, to equalize the rates of dehydration of drought-stressed and non-drought-stressed leaves without affecting the greater tolerance of the drought-stressed leaves. Dehydration avoidance by osmotic adjustment was markedly developed in the slowly dehydrated attached leaves of drought-stressed plants, but not in the rapidly dehydrated excised leaves. This is evidence of drought acclimation. It must, therefore, be concluded that the slow dehydration of the drought-stressed plants also leads to the increase in dehydration tolerance by permitting drought-induced acclimation. The overall drought resistance of cabbage leaves depends on the three components: drought avoidance, dehydration avoidance and dehydration tolerance. The latter two increase during acclimation but the cuticular control of drought avoidance does not.  相似文献

Oviposition by Pieris rapae L. on cabbage was deterred by homogenized cabbage tissue sprayed onto intact plants. Ether extracts of cabbage also were deterrent, but water extracts were not. Hexane extracts of other host plants deterred oviposition and water extracts had little or no effect. Polar as well as non-polar extracts of non-host plants inhibited oviposition. Polar deterrents in the non-host crucifers, Erysimum cheiranthoides and Capsella bursa-pastoris may explain the avoidance of these plants by P. rapae. Chemical deterrents, as well as stimulants, apparently play a major role in the acceptance or rejection of plants as hosts by ovipositing female butterflies.
Zusammenfassung Die Eiablage von Pieris rapae auf Kohl wurde gehemmt von einem Homogenisat aus Kohlblättern, das auf intakte Pflanzen gesprüht wurde. Ether-Extrakte der Kohlblätter wirkten im Gegensatz zu Wasserextrakten abschreckend. Ebenso zeigten Hexanextrakte anderer Wirtspflanzen eine hemmende Wirkung auf die Eiablage, während Wasserextrakte anderer Wirtspflanzen keine Wirkung aufwiesen. Dagegen aber verhinderten sowohl polare als auch unpolare Extrakte von Nicht-Wirtspflanzen die Eiablagen. Polare, abschreckende Substanzen in den Nicht-Wirtspflanzen Erysimum cheiranthoides und Capsella bursa-pastoris (Cruciferae) könnten Ursache dafür sein, dass P. rapae diese Pflanzen meidet. Chemische Substanzen mit sowohl abschreckender als auch stimulierender Wirkung spielen offensichtlich eine bedeutende Rolle für legebereite Schmetterlingsweibchen bei der Auswahl von Pflanzen als geeigneten Eiablageplatz.

Hypocotyl-derived callus cultures of Brassica campestris L. ssp. pekinensis cv. Kim-jung (Chinese cabbage) were grown on Murashige and Skoog medium containing no additional salt, NaCl or Na2SO4. Na2SO4 was more than twice as inhibitory in comparison to the same concentration of NaCl when growth and fresh:dry weight ratios of established callus were measured. Levels of protein, starch, sucrose and α-amino nitrogen were not significantly altered in salt-grown callus. Concentrations of reducing sugars and chlorophyll were 2–3 times greater in callus grown on either salt. Proline concentration increased 15–20 fold on the highest levels of salt. Final concentrations (reached in 20–24 days) were closely correlated to the initial Na+ concentration of the medium, regardless of salt type. The osmotic potential in callus transferred to NaCl or Na2SO4 reached a maximum negative value after 16 days. For both salts, subsequent increases were correlated to increases in fresh:dry weight and growth. On both salts, turgor remained relatively constant (0. 6–0.75 MPa). Changes in Na+, K+, Mg2+ and Ca2+ content were correlated to initial Na+ concentration in the medium, not salt type. Accumulation of Na+ was accompanied by loss of K+ and Mg2+. Six to seven times less sulfate was measured in callus grown on Na2SO4 than chloride in callus grown on similar concentrations of NaCl.  相似文献
