Summary The batch fermentation of whey permeate to lactic acid was improved markedly by the addition of enzymehydrolyzed whey protein. Acid concentrations greater than 90 g/l were achieved at a productivity of 4.3 g/l per h and a 98% substrate use. Cell mass concentration reached 6 g/l. The acid productivity achieved is somewhat higher than that typical for fermentation of whole whey. The process economics, based on in-house hydrolyzate preparation, look promising. Presented in this paper are the experimental results showing the effects of hydrolyzate concentration on acid and cell mass production.  相似文献

The growth of Hawaiian taape, Lutjanus kasmira , was studied by examining otoliths and by analysing length-frequency distribution. Annual hyaline and opaque markings were visible in whole mounts of sagittae, which were verified by enumeration of daily increments with a scanning electron microscope (SEM) and through marginal increment analysis. The von Bertalanffy growth curve was fitted to the data, resulting in: where t. l . is total length (cm) and t is age (years). SEM observations revealed that the slowgrowth hyaline zones were composed of daily increments too small (0.4–0.8 μm) to be resolved optically. Thus, age estimates derived by numerically integrating otolith growth rate data obtained with a light microscope showed a negative bias, resulting in overestimation of growth rates. Parameter estimates obtained from three different types of length-frequency analysis were also unstable. This was due, at least in part, to differences in the size composition of fish sampled with different fishing gears and from different depths.
The growth rate registered in Hawaii falls within the reported growth coefficients of lutjanids, whereas it is one of the highest in the Pacific and clearly higher than a deep-water lutjanid species growth in Hawaii. Probably, this high growth rate may have been enhanced by the relative lack of competitors in the depauperate Hawaiian marine fish community.  相似文献

One-year-old Arctic charr, Sulvelinus alpinus (L.), of the Hornavan strain were tested from February 1985 to January 1986 in an attempt to get an increased understanding of the annual rheotactic behaviour as well as the die1 and seasonal locomotor activity pattern. An annular stream tank equipped with photocells was used to measure the direction of swimming movements as well as the number of passings. From February to late May the locomotor activity was low but increased in July and peaked in September. After November the locomotor activity was again at low winter levels. During the activity peak from July to November the majority ofall movements was directed against the current while no preference for direction was noted during the rest of the year. The high level of swimming movements directed against the current in late summer and autumn may be related to an innate habitat change. From February until June, the charr exhibited a bimodal diurnal activity pattern. In July activity was evenly spread over the whole 24- hour period and in August and September activity was again mainly diurnal with a bimodal pattern. In October and November the activity was mainly nocturnal and in December and January activity was concentrated in the short light period. Both annual and die1 activity are discussed in relation to earlier findings in general locomotor activity in Arctic charr and other salmonids.  相似文献

In a previous paper we demonstrated that the short-range compact regions in atrial natriuretic factor (-hANF) predicted by the average distance map (ADM) correspond to its active sites [Kikuchi,J. Protein Chem.11, 579–581 (1992)]. In the present paper we apply the same method to other bioactive peptides and peptidic enzyme inhibitors. We again observe that active sites in each peptide are contained in short-range compact regions predicted by the ADM for the peptide. This demonstrates that the ADM method predicts the possible location of active sites in biologically active peptides in general. The possibility of practical application of the present method to rational drug design is also discussed.  相似文献

Flux density and spectral measurements were carried out on magnetic fields generated by several types of motor-driven personal appliances used near the body. Among the units tested were several for which the average flux densities, as determined at the surfaces of the appliance, exceeded 0.4 mT. Time-rates-of-change (dB/dt) for several units exceeded 1000 T/s, and several units exhibited high-frequency components in the low-MHz range. Use of such appliances, although normally of short duration, can represent exposure to magnetic fields of relatively high flux density, which may also have high-frequency components. Compared to other household and commercial sources of magnetic fields, those generated by certain motor-driven personal appliances may represent a significant contribution to time-weighted average exposure and may represent an important source of local induced currents in the body. Furthermore, high-frequency transients that represent only a minor contribution to time-weighted average exposure may generate significant instantaneous induced currents. © 1994 Wiley-Liss, Inc.

    Summary Birds show a typical daily pattern of heavy morning and secondary afternoon feeding. We investigate the pattern of foraging by a bird that results in the lowest long-term rate of mortality. We assume the following: mortality is the sum of starvation and predation. The bird is characterized by two state variables, its energy reserves and the amount of food in its stomach. Starvation occurs during the day if the bird's reserves fall to zero. The bird starves during the night if the total energy stored in reserves and the stomach is less than a critical amount. The probability that the bird is killed by a predator is higher if the bird is foraging than if it is resting. Furthermore, the predation risk while foraging increases with the bird's mass. From these assumptions, we use dynamic programming techniques to find the daily foraging routine that minimizes mortality. The principal results are (1) Variability in food finding leads to routines with feeding concentrated early in the day, (2) digestive constraints cause feeding to be spread more evenly through the day, (3) even under fairly severe digestive constraints, the stomach is generally not full and (4) optimal fat reserve levels are higher in more variable environments and under digestive constraints. This model suggests that the characteristic daily feeding pattern of small birds is not due to digestive constraints but is greatly influenced by environmental variability.  相似文献

    It has been shown that probable portions which form contacts in a protein can be predicted by means of an average distance map (ADM) as well as regular structures (-helices and -turns) defined as short-range compact regions (Kikuchiet al., 1988a,c). In this paper, we analyze the occurrence of those portions and short-range compact regions on ADMs for various proteins regarding their folding types. We have found out that each folding type of proteins shows characteristic distribution of such parts on ADMS. We also discuss the possibility of the prediction of folding types of proteins by ADMs.  相似文献

    The response properties of R1–6 type photoreceptors of the blowfly (Calliphora vicina) were investigated using 1- to 600-ms light steps with contrasts from -1.00 to +1.12 at different adapting backgrounds. To prevent activation of voltage-dependent K+-channels, some photoreceptors were treated with ionophoretically injected tetraethylammonium. The linearity and time-course of the photoresponses depended on the adapting background and the duration of the contrast stimulus. At low backgrounds photoresponses were approximately linear regardless of the contrast step duration. However, at higher backgrounds photoreceptors produced, with long contrast steps of opposite polarities, larger and slower hyperpolarizing responses than depolarizing ones, but linear responses with equal time-courses to transient (2ms) contrast changes. The early rising phases of photoresponses were independent of the contrast signal duration, deviated to the same extent from the steady-state potential and steepened as the value of contrast was increased. The photoreceptor contrast gain increased with the background, but its value depended on the duration, the magnitude, and the polarity of the contrast step. Tetraethylammonium linearised the photoreceptor membrane, increased and delayed photoresponses probably by blocking the shunting of the outwardly rectifying potassium channels.     用锋电位触发叠加(STA)技术对大鼠前庭内侧核(NVM)、下段脑干网状结构(RF)和前庭小脑进行了探查,观察了这些结构中对摆动旋转起反应的单位(PPU)与内脏大神经(SN)记录的交感反应之间的关系。发现用NVM的PPU进行STA,在SN可得一阳性反应,其潜伏期为33.28±3.1ms;用下段脑干RF的PPU进行STA,SN的峰反应潜伏期为11.3±0.91ms;用前庭小脑的PPU进行STA,SN的峰反应潜伏期为21.86±1.73ms。本结果提示前庭交感反应的最近的脊髓上中继站是下段脑干RF旁正中区核团,其下行冲动可能是由网状脊髓束中慢速传导的纤维传导的。根据三个脑区PPU引起的SN-STA潜伏期的不同,在前庭交感反应中前庭小脑所处的地位可能在NVM和下段脑干RF核团之间。  相似文献

    Commonly used semiparametric estimators of causal effects specify parametric models for the propensity score (PS) and the conditional outcome. An example is an augmented inverse probability weighting (IPW) estimator, frequently referred to as a doubly robust estimator, because it is consistent if at least one of the two models is correctly specified. However, in many observational studies, the role of the parametric models is often not to provide a representation of the data-generating process but rather to facilitate the adjustment for confounding, making the assumption of at least one true model unlikely to hold. In this paper, we propose a crude analytical approach to study the large-sample bias of estimators when the models are assumed to be approximations of the data-generating process, namely, when all models are misspecified. We apply our approach to three prototypical estimators of the average causal effect, two IPW estimators, using a misspecified PS model, and an augmented IPW (AIPW) estimator, using misspecified models for the outcome regression (OR) and the PS. For the two IPW estimators, we show that normalization, in addition to having a smaller variance, also offers some protection against bias due to model misspecification. To analyze the question of when the use of two misspecified models is better than one we derive necessary and sufficient conditions for when the AIPW estimator has a smaller bias than a simple IPW estimator and when it has a smaller bias than an IPW estimator with normalized weights. If the misspecification of the outcome model is moderate, the comparisons of the biases of the IPW and AIPW estimators show that the AIPW estimator has a smaller bias than the IPW estimators. However, all biases include a scaling with the PS-model error and we suggest caution in modeling the PS whenever such a model is involved. For numerical and finite sample illustrations, we include three simulation studies and corresponding approximations of the large-sample biases. In a dataset from the National Health and Nutrition Examination Survey, we estimate the effect of smoking on blood lead levels.  相似文献
