The addition of methyl tert-butyl ether (MTBE) to gasoline has resulted in public uncertainty regarding the continued reliance on biological processes for gasoline remediation. Despite this concern, researchers have shown that MTBE can be effectively degraded in the laboratory under aerobic conditions using pure and mixed cultures with half-lives ranging from 0.04 to 29 days. Ex-situ aerobic fixed-film and aerobic suspended growth bioreactor studies have demonstrated decreases in MTBE concentrations of 83% and 96% with hydraulic residence times of 0.3 hrs and 3 days, respectively. In microcosm and field studies, aerobic biodegradation half-lives range from 2 to 693 days. These half-lives have been shown to decrease with increasing dissolved oxygen concentrations and, in some cases, with the addition of exogenous MTBE-degraders. MTBE concentrations have also been observed to decrease under anaerobic conditions; however, these rates are not as well defined. Several detailed field case studies describing the use of ex-situ reactors, natural attenuation, and bioaugmentation are presented in this paper and demonstrate the potential for successful remediation of MTBE-contaminated aquifers. In conclusion, a substantial amount of literature is available which demonstratesthat the in-situ biodegradation of MTBE is contingent on achieving aerobic conditions in the contaminated aquifer.  相似文献

目的 为了解老年性骨质疏松情况,用超声骨密度仪对150例老年男性教师做跟骨超振幅衰减值的测试研究。方法:用DMS公怀UBIS5000型骨密度超声仪检查跟骨,研究受检者的年龄、体重、身高和巳患病咎与测得超声振幅衰减值(BUA),声速(SOS),骨硬度(STI)和相对骨折危险性(RRF)等参数之间的关系。结果:本仪器输出3种结果供医师参考:1.一幅灰阶图,2.一幅彩色图和曲线图,3.感兴趣区所测得的参数,受检者的平均BUA为63.87dB/MHz,SOS为1485.21m/s,本文数据经Statpal软件处理,得出回归公式:体重=189.1417 0.2062BUA-0.0224SOS,R=0.0466,P值有显著意义。结论:BUA值为主要参数,SOS为补充参数,作为评估老年男性教师的骨密度情况,假如BUA值突然下降则提示近期可能发生骨质疏松。  相似文献

微生物对石油烃的降解在自然衰减去除土壤和地下水石油烃污染的过程中发挥了重要作用。微生物通过其产生的一系列酶来利用和降解这类有机污染物,其中,编码关键降解酶的基因称为功能基因。功能基因可作为生物标志物用于分析环境中石油烃降解基因的多样性。因此,研究石油降解功能基因是分析土著微生物群落多样性、评价自然衰减潜力与构建基因工程菌的重要基础。本文主要介绍了烷烃和芳香烃在有氧和无氧条件下的微生物降解途径,重点总结了烷烃和芳香烃降解的主要功能基因及其作用,包括参与羟化作用的单加氧酶和双加氧酶基因、延胡索酸加成反应的琥珀酸合酶基因以及中心中间产物的降解酶基因等。  相似文献

Bilayer liposomes from a mixture of dipalmitoylphosphatidylcholine (DPPC) and dipalmitoylphosphatidylethanolamine (DPPC:DPPE=8:2, molar ratio) or DPPC labeled with 14C-DPPC (DPPC:14C-DPPC) were bombarded with thermally activated tritium atoms. The tritiated liposomes were hydrolyzed by phospholipase C, and the tritium incorporation into different parts of the bilayer along its thickness was determined. The tritium flux attenuation coefficients were calculated for the headgroup (k1=0.176±0.032 Å–1) and acylglycerol residue (k2=0.046±0.004 Å–1) layers indicating a preferential attenuation of the tritium flux in the headgroup region and relative transparence of the membrane hydrophobic part. The finding is potentially important to apply tritium bombardment for investigation of spatial organization of transmembrane proteins in their native lipid environment.  相似文献

Methyl tert-butyl ether (MTBE) is a major gasoline oxygenate worldwide and a widespread groundwater contaminant. Natural attenuation of MTBE is of practical interest as a cost effective and non-invasive approach to remediation of contaminated sites. The effectiveness of MTBE attenuation can be difficult to demonstrate without verification of the occurrence of in-situ biodegradation. The aim of this paper is to discuss the recent progress in assessing in-situ biodegradation. In particular, compound-specific isotope analysis (CSIA), molecular techniques based on nucleic acids analysis and in-situ application of stable isotope labels will be discussed. Additionally, attenuation of tert-butyl alcohol (TBA) is of particular interest, as this compound tends to occur alongside MTBE introduced from the gasoline or produced by (mainly anaerobic) biodegradation of MTBE.  相似文献

The magnitude and frequency of events leading to changes in turbidity have been studied in a large (61 km2), shallow (mean depth 3.4 m) wind-exposed lake basin at the western end of Lake Mälaren, Sweden. In this paper we couple changes in suspended particulate inorganic material (SPIM) resulting from wind driven sediment resuspension, and variations in the discharge and sediment load, to spectral variations in subsurface light and estimates of photosynthetically active radiation (PAR). To accomplish this we use a semi-analytical model which predicts the spectral variations in downwelling irradiance (E d()) and the attenuation coefficient of downwelling irradiance (K d()), as a function of the concentrations of chlorophyll, dissolved yellow substances, and suspended inorganic and organic particulate material. Unusually high river discharge, led to large inputs of yellow substances and large in lake yellow substance concentrations (a ys(420) 20 m–1), causing variations in yellow substance concentration to have the greatest role in influencing temporal trends in the attenuation of PAR and variations in the depth of the euphotic zone (Z eup). In spite of this, variations in SPIM could account for approximately 60% of the variation in Z eup attributed to changes in yellow substances alone. Our results show that changes in suspended sediment concentration leads to both long-term and short-term changes in the attenuation of PAR, even in the presence of high concentrations of dissolved yellow substances.  相似文献

根据A型流感病毒密码子使用偏嗜性,选取稀有密码子对A/Puerto Rico/8/34(H1N1)病毒NS1基因内部110个氨基酸区域进行密码子同义突变改造,并全基因合成NS基因,利用反向遗传操作技术拯救出含有密码子去优化NS1基因的重组病毒(deoNS)。体外细胞噬斑形成实验和病毒生长曲线证明该病毒在MDCK细胞内的感染和复制能力比野生型病毒低约1000倍;BALB/c小鼠体内致病力实验证明deoNS病毒不能引起小鼠发病和死亡,该病毒在小鼠肺内的复制滴度比野生型病毒低100~1000倍。本研究探索了通过基因组密码子去优化改造途径降低A型流感病毒毒力的可行性,首次证明流感病毒NS1基因密码子去优化同义突变可以降低病毒毒力,为流感减毒活疫苗的研究提供了新的思路。  相似文献

Consideration of monitored natural attenuation (MNA) as a remedy component for metals-contaminated sites can be achieved using a site-specific screening approach, followed by application of one or a series of sequential extraction measurements. Hazardous waste sites contaminated with metals can be screened for the implementation of monitored natural attenuation on the basis of contaminant-specific soil chemical characteristics (i.e., Kd's, solubilities, and nonexchangeable sorbed fraction). Field cases are used to demonstrate the screening approach and to outline the primary considerations involved in accurately applying sequential extraction procedures to support the of MNA for site remediation. The results of these case studies provide strong evidence that site-specific screening and the use of sequential extraction procedures are effective methods for evaluating natural attenuation for metals impacted sites.  相似文献

Near‐infrared (NIR) radiation has been employed using one‐ and two‐photon excitation of fluorescence imaging at wavelengths 650–950 nm (optical window I) for deep brain imaging; however, longer wavelengths in NIR have been overlooked due to a lack of suitable NIR‐low band gap semiconductor imaging detectors and/or femtosecond laser sources. This research introduces three new optical windows in NIR and demonstrates their potential for deep brain tissue imaging. The transmittances are measured in rat brain tissue in the second (II, 1,100–1,350 nm), third (III, 1,600–1,870 nm), and fourth (IV, centered at 2,200 nm) NIR optical tissue windows. The relationship between transmission and tissue thickness is measured and compared with the theory. Due to a reduction in scattering and minimal absorption, window III is shown to be the best for deep brain imaging, and windows II and IV show similar but better potential for deep imaging than window I.

A Periconia sp was isolated from Torreya grandifolia (a relative of yew that does not synthesize taxol) near Huangshan National Park in the People’s Republic of China. This fungus, not previously known as a tree endophyte, was isolated from the inner bark of a small lower limb. When freshly isolated from the tree and placed in a semi-synthetic medium, the fungus produced readily detectable quantities of the anticancer drug taxol. Other taxol-producing endophytes were also isolated from this source. The production of taxol by Periconia sp was demonstrated unequivocally via spectroscopic and immunological methods. However, successive transfers of the fungus in semi-synthetic medium resulted in gradual attenuation until low production occurred even though fungal growth was relatively unaffected. Several compounds, known previously as activators of microbial metabolism, including serinol, p-hydroxybenzoic acid, and a mixture of phenolic acids, were capable of fully or partially restoring taxol production to otherwise taxol-attenuated cultures. The compound with the most impressive ability to activate taxol production was benzoic acid at 0.01 mM. Benzoic acid was not a taxol precursor.
