中国石竹离体快繁与试管苗玻璃化研究   总被引:1,自引:0,他引:1  
研究了培养基组成成分对中国石竹品种Diana F1 White组培程序中种子萌发、诱芽增殖、试管苗玻璃化及生根等重要环节的影响。结果表明:(1)种子适宜的萌发培养基为1/2MS,在此培养基中种子萌发率为83.33%,播种后30d时无菌苗高度达6.99cm,叶片数达26.7。(2)在芽诱导阶段玻璃化苗发生率最高可达53.85%。将光照强度提高至2000lx后,采用培养基MS+6-BA3.0mg/L+NAA0.3mg/L+琼脂8.0g/L+蔗糖8.0g/L进行诱芽培养,玻璃化苗发生率降至3.33%,外植体出芽率达到90%,单个外植体出芽数达到7.2个。(3)适宜的生根培养基为1/2MS+1.0mg/LIBA+琼脂8g/L+蔗糖40g/L,培养30d时生根率为100%,平均根条数达到24.7条,平均根长度达到4.7cm。试管苗在消毒后的腐殖土中移栽,成活率达95%。该研究结果为DianaF1试管苗的快速繁殖提供科学依据。  相似文献

Fungal plant pathogens, especially rust fungi (Pucciniales), are well known for their complex life cycles, which include phases of sexual and asexual reproduction. The effect of asexual multiplication on population genetic diversity has been investigated in the poplar rust fungus Melampsora larici‐populina using a nested hierarchical sampling scheme. Four hierarchical levels were considered: leaf, twig, tree and site. Both cultivated and wild poplar stands were sampled at two time points at the start and end of rust epidemics. A total of 641 fungal isolates was analysed using nine microsatellite markers. This study revealed that the genetic signature of asexual multiplication in the wild poplar stand was seen only at lower hierarchical levels (leaf and twig). Moreover, we observed an erosion of clonal structure through time, with an increase in both gene and genotypic diversity. New genotypes contributed to host infection over time, which demonstrates the importance of allo‐infection in the epidemic process in this host‐pathogen system. Compared with the wild stands, the nearly lack of detection of clonal structure in the cultivated stands reflects the higher infection level on cultivated poplars. More generally, this genetic analysis illustrates the utility of population genetics approach for elucidating the proportion of asexual reproduction in the multiplication of isolates during an epidemic, and for proper quantification of asexual dispersal in plant pathogens.  相似文献

通过比较不同的外植体、植物生长调节剂种类和浓度配比、生根培养基类型以及生根苗的移栽基质,建立华泽兰组织培养快速繁殖体系。结果表明,外植体表面消毒以75%酒精预处理10s,再用0.1%HgCl2浸泡10min,以嫩茎节为外植体诱导效果最好。培养基MS+6-BA1.00mg.L-1+IBA0.05mg.L-1利于形成丛生芽,用于继代增殖,30d的增殖系数为9.43;1/2MS+IBA0.05~0.10mg.L-1适宜诱导生根获得再生植株,生根率100%;生根苗适宜移栽于田园土中,成活率93.3%。  相似文献

广西绞股蓝是极具开发潜力的绞股蓝属植物。以其茎尖和叶片为外植体进行的组织培养试验表明,茎尖为最适的外植体,愈伤组织诱导和不定芽增殖的最适培养基为MS+6-BA 1.0mg.L-1+NAA 0.02mg.L-1,在此培养基中,茎尖和叶片的愈伤组织诱导率平均达95%,丛芽增殖系数平均达14。广西绞股蓝最适的生根培养基为MS+NAA 0.20mg.L-1,生根率92.50%。炼苗后组培苗的移栽成活率达95.5%。  相似文献

Production of novel mutants with a high ability to mitigate pollutants is important for phytoremediation. We investigated the use of ion beam irradiation to produce mutants of Ficus pumila L. with an improved ability to mitigate atmospheric nitrogen dioxide (NO2). More than 25,000 shoot explants were irradiated with an ion beam (12C5+, 12C6+, or 4He2+), from which 263 independent plant lines were obtained. The plants were analyzed for NO2 uptake by fumigation with 1 ppm 15N-labeled NO2 for 8 h in light, followed by mass spectrometric analysis. The mean NO2 uptake values of each of the 263 lines differed over a 110-fold range. Propagation was attempted using cuttings from 44 lines showing the greatest NO2 uptake; in total, 15 lines were propagated. Two of the 15 lines showed a mean NO2 uptake 1.7- to 1.8-fold greater than that of the wild-type. This increase in NO2 uptake was heritable in both lines; their progenies showed a significantly greater ability to take up and assimilate NO2 than did the wild-type. RAPD analysis demonstrated DNA variation between the progeny plants and the wild type, suggesting that the progeny were true mutants. These mutants of F. pumila may prove useful in mitigating atmospheric NO2.  相似文献

The cosmopolitan marine bivalve genus Lasaea is predominantly composed of highly polyploid asexual lineages with one exception: the diploid, sexual Australian species L. australis. Two undescribed, direct-developing congeners co-occur with the indirect-developing L. australis on the rocky intertidal of southeastern Australia. One of these, L. colmani sp. nov., is also diploid and sexual. The other direct-developing congener is an asexual polyploid composed of a variety of clonal lineages. All three sympatric Australian Lasaea congeners are morphologically distinguishable, although prodissoconch distinctions are required to separate large polyploid clams from equivalently-sized L. australis. Similarities in mitochondrial gene sequence and in shell morphology suggest that L. australis and the Australian sympatric polyploid clones share an exclusive common ancestor despite differing in developmental mode, ploidy and reproductive mode. However, detailed karyological analyses failed to identify a chromosome set morphologically similar to that of L. australis among the sympatric Australian polypoid complement. We propose that generation of the polyploid Australian clones (presumably by hybridization) was followed by radical karyological rearrangement.  相似文献

Tetramerista glabra has a remarkable combination of life history traits. It is a dense-wooded, large, common canopy tree in primary peat swamp rain forest. Its seedlings, although shade tolerant, can grow rapidly in high light conditions, but frequently lack structural stability. Most juvenile stems (94% in the understory and 38% in canopy gaps) collapse under their own weight or from branchfalls. Fallen stems then ramify into vegetative sprouts, which in turn may collapse, perpetuating a vegetative juvenile cycle. Most recruitment is from sprouts rather than from seed. Structural analysis of stem dimensions shows that stems 2–8 cm DBH (diameter at breast height) are close to the theoretical buckling limit, especially for those dependent on neighboring vegetation to maintain vertical form. Trees > 4 cm DBH persisting as upright stems develop stilt root support and become structurally independent at ca 8 cm DBH. Eventually, as stems thicken, stilt roots anastamose and trees adopt the cylindrical growth form of mature canopy trees (up to 150 cm DBH). Thus, the vegetative life history strategy of the species is to: (i) regenerate a large “ramet bank” from the majority of juveniles that fail structurally while suppressed in the understoty, and (ii) to maximize height growth at the expense of diameter growth in high light conditions. The growth pattern and plastic form of T. glabra shows how a shade tolerant species may adapt to utilize the ephemeral light resource in canopy gaps. The growth strategy of this species allows it to circumvent the normal trade-off between shade tolerance and rapid growth in canopy gaps.  相似文献

Abstract An unusual new species of the gall-inducing scale insect genus Apiomorpha Rübsaamen is described from Queensland. The adult female, its gall, and the first-instar nymph (crawler) are illustrated, and relationships of the new species are estimated using mitochondrial COII data. Adult females induce cigar-shaped galls on leaves of several eucalypts in section Adnataria of subgenus Symphyomyrtus . The bilobed anal lobes of the adult female differ from those of all other Apiomorpha species (single lobe) and the first-instar nymph possesses features, such as broad frontal tubercles and dorsal stripes, that are not present in crawlers of other Apiomorpha species. However, DNA sequence data confirm that the new species falls within Apiomorpha , rather than representing a sister group, and indicate that the new species is not closely related to the A. pharetrata (Schrader) species-group, the only other group within Apiomorpha that induces cigar-shaped galls on leaves. The systematic affiliations of A. gullanae sp. n. are currently not known. Females only are known and there is some indication that reproduction in the new taxon is parthenogenetic. This represents the first putative case of parthenogenesis in Apiomorpha .  相似文献

Dispersal patterns of Lonicera periclymenum determined by genetic analysis   总被引:2,自引:0,他引:2  
Colonization of Lonicera periclymenum L. (honeysuckle) was studied by RAPD analysis of young ramets in two woodlots planted 20 years ago, and in all ramets in older woodlots within a range of 1 km. Mature ramets that climbed in a particular tree always belonged to one individual. Twenty-five percent of the mature individuals had reproduced vegetatively to other trees or patches nearby, which indicates that the larger part of reproduction is sexual. Some young plants that were growing at close distances from each other were genetically highly similar and shared high similarities to the same mature plants. They may be the product of one dispersal event. Detection of parents of young individuals by exclusion was not successful, because of the dominant nature of the bands. Average distances from young plants to genetically most similar mature plants were variable, due to the small number of colonization events. However, four ways of analysis of genetic similarity among all individuals indicated that exchange of genetic material by seed and pollen occurs to a large extent over small distances and within woodlots: (i) using the Mantel test, pairs of individuals with highest similarity were found significantly more often in the same woodlot than in different woodlots; (ii) genetic similarities between individuals decreased significantly with geographical distance, but only for distances up to 300 m; (iii) individuals of woodlots in the Western part of the study area were hardly related to individuals in the Eastern part of the study area, a distance of 2–3 km; (iv) ΦST in the study area was 0.186, indicating a limited gene flow between woodlots. These results are consistent with the dispersal distance as estimated from the average distance between colonized woodlots and the nearest occupied old woodlot in earlier research.  相似文献

It is commonly recognized that large uncertainties exist in modelled biofuel‐induced indirect land‐use change, but until now, spatially explicit quantification of such uncertainties by means of error propagation modelling has never been performed. In this study, we demonstrate a general methodology to stochastically calculate direct and indirect land‐use change (dLUC and iLUC) caused by an increasing demand for biofuels, with an integrated economic – land‐use change model. We use the global Computable General Equilibrium model MAGNET, connected to the spatially explicit land‐use change model PLUC. We quantify important uncertainties in the modelling chain. Next, dLUC and iLUC projections for Brazil up to 2030 at different spatial scales and the uncertainty herein are assessed. Our results show that cell‐based (5 × 5 km2) probabilities of dLUC range from 0 to 0.77, and of iLUC from 0 to 0.43, indicating that it is difficult to project exactly where dLUC and iLUC will occur, with more difficulties for iLUC than for dLUC. At country level, dLUC area can be projected with high certainty, having a coefficient of variation (cv) of only 0.02, while iLUC area is still uncertain, having a cv of 0.72. The latter means that, considering the 95% confidence interval, the iLUC area in Brazil might be 2.4 times as high or as low as the projected mean. Because this confidence interval is so wide that it is likely to straddle any legislation threshold, our opinion is that threshold evaluation for iLUC indicators should not be implemented in legislation. For future studies, we emphasize the need for provision of quantitative uncertainty estimates together with the calculated LUC indicators, to allow users to evaluate the reliability of these indicators and the effects of their uncertainty on the impacts of land‐use change, such as greenhouse gas emissions.  相似文献
