Three methods of increasing the productivity of somatic embryogenesis in Medicago sativa L. were investigated. In the basic procedure, somatic embryos were initiated from young petioles and carried through several phases: callus formation, suspension culture, selection of the embryogenic fraction by sieving, development, maturation, desiccation and storage. The suspensions were normally separated into three fractions by sieving. Fraction I (<200 m) containing nonembryogenic cells or cell clusters was discarded. Fraction II (200–500 m) consisting of embryogenic cell clusters was collected for embryo development and maturation. Fraction III (over 500 M) containing the mixture of petiole residues with large pieces of calli and globular somatic embryos was usually discarded. Several methods to scale-up the suspension phase were unsuccessful. Direct subculture of the entire suspension by the addition of fresh liquid medium resulted in the loss of embryogenic capacity by the third subculture. Subculture of fraction II decreased embryogenic cell mass, and hence reduced total productivity. The recycling of fraction III back to fresh B5g liquid medium resulted in high productivity in the first culture but further subculture of this fraction resulted in a rapid decline in the embryogenic capacity.As an alternative, somatic embryos from the first tissue culture cycle were also used as explants for the initiation of secondary embryogenic callus. The embryogenic capacity of these somatic embryo explants declined rapidly as they matured. More than 100 secondary somatic embryos could be induced from embryo explants removed from development medium at 10 days after sieving the suspension, but only 40 somatic embryos were produced from each mature somatic embryo explant, and 13 from desiccated embryos. The secondary somatic embryos were comparable to the primary embryos in quality according to germination tests. Seasonal changes in the phytoplankton community of a small tropical reservoir were monitored over a four year period comprising of an initial two seasonal cycles during which the water column stratified strongly for extended periods each year, and two further seasonal cycles after installation of a mechanical aeration system to induce artificial destratification. In the unmanaged reservoir, the concentration of chlorophyll a at 0.5 m reached maximum values (on one occasion > 90 mg m−3) when the water column was stratified and the epilimnion was very shallow (ca 2 m depth). The hypolimnion at this time was anoxic (less than 2% oxygen saturation) and had a high concentration of bacteriochlorophyll (100–200 mg m−3). The phytoplankton community of the unmanaged reservoir was generally dominated by cyanobacteria (Cylindrospermopsis raciborskii, Anabaena tenericaulis) during the warmer months of the year (November–March) (but replaced by chlorophyta, dinophyceae and euglenophyceae after periods of intense rain) and by bacillariophyceae (Synedra ulna var. chaseana, S. tenera) during the cooler, dry months. In the artificially destratified reservoir (8 h aeration day−1), the phytoplankton community was largely dominated by diatoms except after depletion of the silica content of the water column which caused diatoms to be replaced by cyanobacteria (dominated by A. tenericaulis) and a range of chlorophytes. The changing pattern of stratification and circulation of the water column in the unmanaged reservoir caused repeated disruption of the established phytoplankton assemblage with peaks of high biomass associated with transient cyanobacterial blooms. Continuous aeration and the consequent increase in the ratio mixed: euphotic depth provided conditions suitable for dominance of the phytoplankton by diatoms, as long as silica was available, and resulted in average chlorophyll levels higher than in the unmanaged reservoir (120 ± 10 v. 64 ± 9 mg m−2). Hierarchical fusion analysis based on the biomass of species differentiated the phytoplankton samples into cluster groups that could be related primarily to stratification or mixing of the water column.  相似文献

A simplified and non-destructive method using starch gel electrophoresis has been developed on seeds to identify inbred lines of Vicia faba and assess outcrossing rates and gene dispersal in pollination experiments. Six enzyme systems (Alcohol dehydrogenase, Aspartate aminotransferase, Glucose-6-phosphate isomerase, Isocitrate dehydrogenase, Phosphogluconate dehydrogenase and Shikimate dehydrogenase) were analysed from parental lines, crosses performed between lines bearing dissimilar isozyme patterns in pollination cages with Bombus and F2 progenies obtained from manual selfing of F1 hybrids. The allozymes at each of the seven studied loci segregated in the expected Mendelian fashion and behaved in a co-dominant manner except for the Adh-2 locus where the only variant was a null allele. No evidence of genetic linkage was observed between at least 13 of the 15 pairs of the studied loci. Percentage of cross fertilisation by Bombus between seven pairs of inbred lines ranged between 1.7% and 28.3%. Pollen transfer between a donor line and a recipient line by two species of Bombus did not lead to differences in outcrossing rates (both about 8%). The new PGD marker with two loci at three alleles each is particularly discriminating and valuable in pollination studies and breeding of V. faba.  相似文献

Survival of secondary embryoids of winter oilseed rape (Brassica napus ssp. oleifera cv. Primor) has been used as an assay for the development of artificial seeds involving complex coacervation of alginate (polyanion) with chitosan (polycation). Germination frequency of 100% was achieved for encapsulated embryoids when alginate formed the inner matrix and chitosan the outer layer. When the matrix makeup was reversed, there was no germination of embryoids. The artificial seeds produced were hardened in dilute alkaline solutions of NaOH and Ca(OH)(2). An optimum setting time could be selected based on a quantitative measurement of resistance of hardened capsules to compression and the germination frequency of the encapsulated embryoids. Recent progress in plant transformation for insect resistance has increased the interest in the potential toxicity of proteins towards insect pests. While studies have been targeted to a large array of insect species, phloem-feeding Homoptera have not been investigated yet. The paper describes a routine test for screening toxicity and growth inhibition of purified proteins in artificial diets onAcyrthosiphon pisum (Harris). Twenty-five commercially available proteins of different classes were tested and compared to some non-protein chemicals (an insecticide, an antibiotic …).A. pisum proved to be very sensitive to all proteases tested and to some venoms with general cytolytic properties. A plant lectin, concanavalin A, displayed significant toxicity and growth inhibition, while various proteins such as a soybean proteinase inhibitor, a chitinase, and bovine serum albumin showed measurable impairments of growth only at higher dose (≥250 μg.ml−1). Some proteins were without short-term effect onA. pisum physiology. The influence of these results on aphid-plant interactions are discussed.
Résumé L'effet de protéines alimentaires sur les insectes phloémophages, dont les pucerons, n'a jamais été étudié. Nous proposons ici un test biologique standardisé sur milieu artificiel permettant d'analyser les effets de différentes classes de protéines sur la physiologie d'A. pisum. La validité de ce test est éprouvée (protocole, reproductibilité) et les différentes données récoltées (mortalité et inhibition de croissance) permettent de définir des paramètres toxicologiques tels que concentration létale 50 ou concentration inhibitrice 50. Cette caractérisation toxicologique a été réalisée sur 25 protéines appartenant à des classes différentes, ainsi que plusieurs substances non protéiques utilisées comme témoin de toxicité (insecticide, antibiotique, inhibiteur de synthèse protéique et glucoside phénolique). Les regroupements de protéines par proximités de profils toxicologiques ont été corrélés aux activités biochimiques des différentes protéines. Les implications de ces résultats sur les interactions plante-puceron sont discutées, ainsi que le potentiel d'une stratégie de création de variétés transgéniques résistantes aux pucerons.

The seasonal variation in periphyton dynamics has been studied upon artificial substratum (microscopic glass slides) under various light conditions during the periods May–October 1986 and May–September 1987, in Lake Veluwe. Some additional observations on the periphyton development upon leaves of Potamogeton pectinatus L. have been made simultaneously. Four different light conditions were created in an experimental setup by manipulating the photon flux density through artificial shading.Periphyton upon artificial substratum exhibited a relatively high abundance with a distinct seasonal pattern. Periphyton accrual rates were highest at the beginning of June and in August and September upon slides which were incubated for two weeks. Periphyton mass increased during May and June, decreased or remained about the same during July and subsequently increased until an upper plateau was reached upon slides which were incubated from the beginning of May onwards.Generally, periphyton mass was lower upon slides than upon P. pectinatus. The seasonal variation in periphyton mass was more pronounced upon P. pectinatus leaves than upon the slides.Attenuation by periphyton upon slides ranged from 5 to 65% after two weeks of incubation. Periphyton upon slides which had been incubated for more than two weeks demonstrated an attenuation of more than 85%.Water quality parameters other than photon flux density were probably more important in determining the periphyton dynamics, since only minor differences were observed in periphyton mass between the various light conditions. Chlorophyll-a content was higher with increased shading on various sampling dates.Periphyton, especially older periphyton consisted largely of settled silt and clay particles and to a lesser extent of detrital matter on both substrata. Living epiphytes were only a relatively small fraction.It is concluded that a reduction of resuspension of sediment particles, giving less suspended matter in the water column, will result in lower periphytic mass. Consequently, the quantity of photosynthetically active radiation reaching the submerged macrophytes is expected to increase considerably.  相似文献

荒漠草原(生态区)横贯我国西北地区东部,生态地位十分重要。近二十年来,通过封育禁牧、退耕还林(草),植被覆盖显著改善,但是生态系统质量和稳定性依然不高。由于长期将荒漠草原单纯视作草原的一部分,对其生态系统过渡性、脆弱性和复杂性本质特征认识不足,造成了荒漠草原生态学研究与区域生态建设实践之间不同程度的脱节。在分析荒漠草原生态区未来在我国生态安全格局中突出的但是被一定程度上忽视的地位的基础上,进一步归纳了荒漠草原生态系统的一般特征,指出了生态恢复与重建研究中存在的主要问题。进而以人工植被引入荒漠草原生态工程为案例,分析了人工植被驱动荒漠草原生态恢复与重建的过程与机制,归纳了“植被-水文-土壤”互馈作用驱动生态系统层级响应模式,并展望了今后的发展趋势与研究方向。  相似文献

Preservation of the genetic diversity of the captive orangutan, especially the wild-caught founders, is critical in maintaining a long-term population in zoological parks. One solution to the problem of maintaining maximum genetic diversity would be to initiate a program of artificial insemination for genetically underrepresented individuals through the banking and interinstitutional use of cryopreserved semen. However, little is known about basic orangutan semen characteristics, and current methodology is inadequate to support such a program. In this paper, we report the results of semen collection from an adult Sumatran orangutan (Pongo pygmaeus abelli), using an artificial vagina without anesthesia or electrical stimulation. A total of 27 ejaculates were evaluated during a 1-year period. The total and liquid volumes of the ejaculates at 1 h following collection were 6.1 ± 0.6 ml and 2.6 ± 0.4 ml, respectively (mean ± SEM). The liquid portion continued to exude semen for 2 h; however, 90% of the motile sperm was exuded within the first 30 min. The total number of sperm in the ejaculate was 164 ± 106 ± 16.5, and the percentage of motile cells was 60 ± 2.7%. We conclude that the artificial vagina provides a promising technique for semen collection in the orangutan, and view these results as an initial step in developing methods for in vitro sperm capacitation, sperm cryopreservation, and artificial insemination. © 1993 Wiley-Liss, Inc.  相似文献
