This study evaluated fertility in swamp buffalo after synchronization of ovulation combined with fixed time artificial insemination. At the start of the study, designated day 0, from a group of 98 female Thai swamp buffalo, 55 buffalo (heifers n° = 20 and cows n° = 35) were selected to be synchronized with GnRH (Day 0) followed by PGF2alpha (Day 7) and a second treatment with GnRH (Day 9). All buffalo were inseminated at two fixed times 12 h and 24 h after the second injection of GnRH (Ovsynch+TAI group); a second group of 43 buffalo (heifers n° = 19 and cows n° = 24) were not treated and were artificially inseminated (AI) at natural estrus (AI group). Blood samples were taken 22 days after insemination to evaluate progesterone plasma levels. In the Ovsynch+TAI group, overall conception rate (CR; i.e. the number of cows with progesterone >4.0 ng/ml on day 22 after AI divided by the number of animals inseminated), was 38.1% and overall pregnancy rate (PR; i.e. the number of cows that were pregnant at day 50-60 after insemination divided by the number of animals inseminated), was 32.7%. In the AI group overall CR and PR was 34.9%.Within the Ovsynch+TAI group, CR and PR were reduced (P < 0.05) in heifers compared with cows (CR 15.0% vs. 51.4% for heifers and cows, respectively; PR 15.0% vs. 42.9% for heifers and cows, respectively). Within the AI group the efficacy of treatment was similar between heifers and cows (CR and PR 31.6% for heifers and 37.5% for cows).In conclusion, this study indicates that in swamp buffalo it is possible to synchronize ovulation and use timed artificial insemination with the Ovsynch+TAI protocol.  相似文献

通过同源重组将携带细菌人工染色体(Bacterial Artificial Chromosome,BAC)载体和GFP表达框的pHA2质粒序列插入到PRV病毒的UL23(TK)基因内,获得了重组病毒rPRV-HA2;将该重组病毒的环状基因组电转化到感受态细胞EscherichiacoliDH10B,筛选到病毒的感染性克隆PRV BAC(pPRV)。pPRV转染VeroE6细胞可以重新启动病毒的生产性感染,该拯救病毒的细胞病变和体外增殖特性与rPRV-HA2一致。病毒生长曲线表明TK基因的部分删除和BAC载体的插入不会影响病毒在体外的复制。PRV感染性BAC克隆的成功构建,将方便在大肠杆菌内对病毒基因组进行快速、准确的操作,为进一步开展PRV基因功能和病毒载体研究奠定基础。  相似文献

Under biotic/abiotic stresses, the red alga Kappaphycus alvarezii reportedly releases massive amounts of H2O2 into the surrounding seawater. As an essential redox signal, the role of chloroplast-originated H2O2 in the orchestration of overall antioxidant responses in algal species has thus been questioned. This work purported to study the kinetic decay profiles of the redox-sensitive plastoquinone pool correlated to H2O2 release in seawater, parameters of oxidative lesions and antioxidant enzyme activities in the red alga Kappaphycus alvarezii under the single or combined effects of high light, low temperature, and sub-lethal doses of 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU) and 2,5-dibromo-3-methyl-6-isopropyl-p-benzoquinone (DBMIB), which are inhibitors of the thylakoid electron transport system. Within 24 h, high light and chilling stresses distinctly affected the availability of the PQ pool for photosynthesis, following Gaussian and exponential kinetic profiles, respectively, whereas combined stimuli were mostly reflected in exponential decays. No significant correlation was found in a comparison of the PQ pool levels after 24 h with either catalase (CAT) or ascorbate peroxidase (APX) activities, although the H2O2 concentration in seawater (R = 0.673), total superoxide dismutase activity (R = 0.689), and particularly indexes of protein (R = 0.869) and lipid oxidation (R = 0.864), were moderately correlated. These data suggest that the release of H2O2 from plastids into seawater possibly impaired efficient and immediate responses of pivotal H2O2-scavenging activities of CAT and APX in the red alga K. alvarezii, culminating in short-term exacerbated levels of protein and lipid oxidation. These facts provided a molecular basis for the recognized limited resistance of the red alga K. alvarezii under unfavorable conditions, especially under chilling stress.  相似文献

The catfish IGH locus is large (∼1 Mb) and complex, having undergone multiple internal duplications and transpositions. To define the structure of the locus that contains the single expressed IGHM gene, two overlapping bacterial-artificial-chromosome (BAC) clones spanning the most 3′ end of the channel catfish immunoglobulin heavy (IGH) chain locus have been completely sequenced. The analyses created a contig of 257,153 bp containing 55 VH, 6 D, 12 JH genes and the IGH constant region genes encoding the functional secreted and membrane forms of IgM and the membrane form of IgD. This analysis revealed three major features. First, no C-region genes were found aside from the previously described IGHM1 and IGHD1, with the latter gene being the most 3′ C-region gene of the catfish IGH locus. There was no evidence in the region sequenced for genes that could encode an Ig class similar to the IgZ/IgT described in zebrafish, trout and pufferfish. Second, there are a high number of VH pseudogenes, 28 out of 55 (51%). In contrast, the entire zebrafish IGH locus has 40 functional VH genes and eight pseudogenes (17%). Third, an internal duplication of a 52.4-kb block of VH genes has occurred. These observations suggest that the IGH locus of teleost fish varies significantly from species to species in the diversity of C-region genes as well as the numbers of genes encoding V regions.  相似文献

Glucose-regulated protein 58 (GRP58)-like immunoreactivity in rat liver obtained in the evening or after fasting underwent an electrophoretic band-shift, which disappeared after phosphatase-treatment. Since mass spectrometric analysis raised a possibility that Ser150 of GRP58 is phosphorylated, an antibody against the phosphoserine150 GRP58 was generated. Immunoreactivity to this antibody was increased in the evening and after fasting. Since GRP58 was shown to interact with signal transducer and activator of transduction 3 (STAT3), a leptin-related protein, the effect of leptin was examined. Immunoreactivity to the anti-phosphoGRP58 antibody was markedly elevated after the leptin injection, indicating that Ser150 of GRP58 is phosphorylated after fasting and leptin-treatment.  相似文献

The macrozoobenthos colonization process on artificial substrata in lotic environments was evaluated. Two seasonal periods, autumn–winter and spring–summer, of a relatively undisturbed fluvial system, the Estero Nonguén (VIII Region, Chile), have been studied. Five stations, two in riffles and three on sandy bottoms, extending from a head water stream to 5 km down river, were sampled. For all stations and for each season the results were as follows: (a) 32% of the total number of taxa found in this study colonized the artificial substrata on the first day of exposure, (b) the taxa that colonized sandy bottoms were typical for riffle, (c) the colonization process showed the same abundance, specific and cumulative richness and diversity patterns over time, and (d) equilibrium was reached approximately 2 weeks after initial exposure. This suggests that “Estero Nonguén” stream presents a nearly pristine condition and can be considered as a reference site to carry out biodiversity and ecosystem functionality studies, and giving information on time recovery for sectors previously disturbed. Artificial reefs are man-made structures that are placed on the seabed to mimic natural reefs. The Loch Linnhe Artificial Reef will consist of up to 40 reef modules, when completed, each constructed using concrete blocks. As part of this complex, reef modules of approximately 200 tonnes were deployed in June 2001 and January 2002 and form the basis of this study. Sediment samples were taken adjacent to- and 5 m from the reef perimeters and characterised in terms of sediment oxygenation (redox), particle size, organic carbon (loss on ignition) and coarseness (material >1 mm and <5 mm). Changes in molluscs and ophiuroids (>1 mm) present in the sediment were linked to chemico-physical changes occurring as a consequence of the reef. Both reefs trapped macroalgal phytodetritus at the perimeters, which subsequently decomposed. This accumulated material isolated the seabed from the overlying water column and was associated with a decrease in sediment oxygenation that was linked to changes in community structure. LOI was significantly higher around one reef compared with 5 m from that reef. The reefs did not cause significant changes in the sediment texture (median particle size or coarseness) and neither LOI nor sedimentary texture changes were associated with community change.  相似文献

Studies of rodents have shown that both forced and voluntary chronic exercise cause increased hindlimb bone diameter, mass, and strength. Among species of mammals, "cursoriality" is generally associated with longer limbs as well as relative lengthening of distal limb segments, resulting in an increased metatarsal/femur (MT/F) ratio. Indeed, we show that phylogenetic analyses of previously published data indicate a positive correlation between body mass-corrected home range area and both hindlimb length and MT/F in a sample of 19 species of Carnivora, although only the former is statistically significant in a multiple regression. Therefore, we used an experimental evolution approach to test for possible adaptive changes (in response to selective breeding and/or chronic exercise) in hindlimb bones of four replicate lines of house mice bred for high voluntary wheel running (S lines) for 21 generations and in four nonselected control (C) lines. We examined femur, tibiafibula, and longest metatarsal of males housed either with or without wheel access for 2 months beginning at 25-28 days of age. As expected from previous studies, mice from S lines ran more than C (primarily because the former ran faster) and were smaller in body size (both mass and length). Wheel access reduced body mass (but not length) of both S and C mice. Analysis of covariance (ANCOVA) revealed that body mass was a statistically significant predictor of all bone measures except MT/F ratio; therefore, all results reported are from ANCOVAs. Bone lengths were not significantly affected by either linetype (S vs. C) or wheel access. However, with body mass as a covariate, S mice had significantly thicker femora and tibiafibulae, and wheel access also significantly increased diameters. Mice from S lines also had heavier feet than C, and wheel access increased both foot and tibiafibula mass. Thus, the directions of evolutionary and phenotypic adaptation are generally consistent. Additionally, S-line individuals with the mini-muscle phenotype (homozygous for a Mendelian recessive allele that halves hindlimb muscle mass [Garland et al., 2002, Evolution 56:1,267-1,275]) exhibited significantly longer and thinner femora and tibiafibulae, with no difference in bone masses. Two results were considered surprising. First, no differences were found in the MT/F ratio (the classic indicator of cursoriality). Second, we did not find a significant interaction between linetype and wheel access for any trait, despite the higher running rate of S mice.  相似文献

报道饲养烟青虫HelicoverpaassultaGue澖ee的一种人工饲料,比较了烟青虫取食该饲料和天然饲料的主要生物学指标,测定了6龄幼虫对2种食料的转化和利用率。该人工饲料的主要成分是麦胚、黄豆粉、酵母粉和烟叶粉,天然饲料为烟草叶片。结果表明:取食人工饲料的烟青虫幼虫生长发育快,化的蛹较大,成虫寿命长,产卵量大,所产卵孵化率高;取食人工饲料的烟青虫与取食烟草蒴果的相比,前者相对取食量和近似消化率较高,相对生长率无显著性差异。说明烟青虫对该人工饲料有较好的适应性,可以作为繁殖烟青虫的实用饲料。  相似文献

大黄鱼人工诱导雌核发育后代的微卫星标记分析   总被引:22,自引:0,他引:22  
通过冷休克法诱导2组大黄鱼雌核发育,并采用6对微卫星标记引物 (C7、C13、C27、C44、D19、F27)对雌核发育家系G1、G2中24尾鱼苗及双亲进行PCR扩增,并进行微卫星标记比较分析。结果表明:冷休克诱导雌核发育二倍体获得了35.3%的孵化率和45日龄9.9%的成活率;有4对引物可扩增出清晰的亲本差异条带,其中引物C44、C27扩增结果显示G1子代中均未出现雄鱼基因,全部为雌核发育产物,C27扩增结果显示G2子代中也未出现雄鱼基因,但D19扩增结果显示G2子代中有3尾出现雄鱼基因,属正常受精个体,两家系后代的基因纯合率分别达93.8%和72.9%,平均为83.3%。研究表明雌核发育是促进基因纯合的一个有效途径,微卫星标记技术是是鱼类雌核发育鉴定和遗传分析的有效方法。  相似文献
