One hundred and seventy-six isolates were obtained from corn sheath blight samples in Huanghuai Plain and Haihe Plain (including Shandong, Henan, Hebei Provinces and Northern regions of Jiangsu Province) of China. Anastomosis group identification and 5.8S rDNA-internal transcribed spacer (ITS) sequence analysis showed that the isolates belonged to multinucleate Rhizoctonia AG1-IA, AG1-IB, AG4-HG-I, AG-5 and WAG-Z and binucleate Rhizoctonia AG-A and AG-Ba. Of these, AG-1-IA was the major anastomosis group(AG)(64.20% of total isolates), followed by AG-Ba (12.50%), WAG-Z (10.23%), AG1-IB (5.11%), AG4-HG-I (3.98%), AG-5 (2.27%) and AG-A (1.70%). AG-Ba was isolated for the first time from maize in China. 5.8S rDNA-ITS sequence analysis showed that the isolates could be distinctly separated based on their AG types. The isolates belonging to the same AG (or sub-AG) showed 97%-100% sequence identity.  相似文献

ScleortiumcepivorumBerk.是引起洋葱白腐病的病原真菌,大部分测试的菌株含有dsRNA片段,这些dsRNA片段被称为真菌病毒。经琼脂糖凝胶电泳分析,在不同分离株之间或同一分离株不同提取物之间dsRNA带的数量和位置有很大的变化。ScQ-4菌株菌丝生长缓慢,致病力明显低于其他菌株。采用蔗糖密度梯度离心法证实和确定ScQ-4菌株中dsRNA片段所处的亚细胞位置,结果表明这些dsRNA片段与其寄主线粒体共同纯化,因此它们可能存在于寄主线粒体中,属于线粒体病毒。在营养相容性菌株之间这些dsRNA片段可通过菌丝融合进行传播,获得dsRNA片段的受体菌株的致病力明显降低,这对SclerotiumcepivorumBer.k进行生物防治提供了可能性。  相似文献

从黄淮海地区采集玉米纹枯病标样250余份,分离得到176个丝核菌菌株。融合群测定及5.8SrDNA-ITS区序列分析结果表明,这些菌株分别属于多核丝核菌的AG1-IA、AG1-IB、AG4-HG-I、AG-5、WAG-Z融合群及双核丝核菌的AG-A、AG-Ba融合群。其中AG1-IA是优势融合群,占分离菌株总数的64.20%,其次是AG-Ba,占12.50%,再依次分别是WAG-Z(10.23%)、AGI-IB(5.11%)、AG-4-HG-I(3.98%)、AG-5(2.27%)和AG-A(1.70%)。其中AG-Ba融合群是国内首次在玉米上分离得到。从各融合群中选取代表性的菌株进行5.8S rDNA-ITS区序列分析结果表明,隶属不同融合群或亚群的菌株其5.8S rDNA-ITS区序列存在较大的差异,而相同融合群(或亚群)不同菌株之间其序列的一致性可高达97%-100%。  相似文献

以分月扇舟蛾Clostera anastomosis(L.)为材料,以I-72杨为对照,研究南京林业大学新近引进的18个美洲黑杨(Populus deltoides L.)无性系在实验室条件下的抗虫性,筛选出11个具有较强抗性的无性系,并分析不同无性系的抗性与其叶片中主要营养物质和次生代谢物质含量的关系,初步解释其抗性机理。其研究结果为今后进行林间试验,选育出在生产上具有推广价值的美洲黑杨优良抗虫品系提供科学依据。  相似文献

Isolates of Rhizoctonia spp. associated with stem canker and black scurf disease of potato were examined for their anastomosis group, sequence variations in the ITS‐5.8S rDNA region, pathogenicity and sensitivity to fungicides. A total of 92 isolates were obtained from diseased tuber, stolon and sprouts of the potato plants, collected from five districts of Bolu province, Turkey. Based on the anastomosis group and the similarity of the nucleotide sequence of the ITS‐5.8S rDNA, most of the isolates (81.5%) were identified as AG 3 PT. Other isolates belonged to AG 2‐1 (1.08%), AG 2‐2 IV (1.08%), AG 4 HG II (8.07%), AG 5 (2.17%), binucleate Rhizoctonia AG A (1.08%) and AG K (4.35%). Pathogenicity tests showed that isolates of AG 3 PT, AG 4 HG II and AG 5 caused similar degrees of disease severity on 45‐day‐old potato seedlings, whereas AG 2‐1 was moderately virulent. AG 2‐2 IV and binucleate Rhizoctonia spp. were weakly pathogenic or non‐pathogenic on potato seedlings. In this study, anastomosis groups of Rhizoctonia spp. isolates associated with potato in Turkey were characterized for the first time using molecular techniques and classified at the level of subgroups. Furthermore, the effect of selected fungicides was evaluated on disease development caused by soil‐borne inoculums of different anastomosis groups (AGs). Flutolanil and Bacillus subtilis QST 713 were found to be most effective against the Rhizoctonia isolates tested. These results revealed significant differences among the fungicides on disease development resulted from the different AGs.  相似文献

The significance of anastomosis formation and the hyphal healing mechanism (HHM) for functionality and integrity of the arbuscular mycorrhizal (AM) fungal mycelial network remains poorly documented. Four Glomeraceae and three Gigasporaceae were cultured monoxenically. Anastomosis formation was assessed using the grid line method, while HHM was time-lapse monitored. In intact mycelial networks, the number of anastomosis per hyphal length was higher for Glomeraceae than for Gigasporaceae strains. Glomeraceae strains studied always formed anastomosis between different hyphae, whereas anastomosis in the Gigasporaceae more often concerned hyphal bridges within the same hyphae. In both families the HHM corresponded to a four-step process; first septum formation; second initiation of growing hyphal tips (GHTs); third GHT elongation, orientation and contact; and fourth GHT fusion and cytoplasmic/protoplasmic flux re-establishment. These four steps differentiated Glomeraceae from Gigasporaceae. The type and number of anastomosis per hyphal length, and the HHM differed considerably between Glomeraceae and Gigasporaceae families representing a supplementary character that distinguishes these two families and may be of significance in ecological studies of AM fungi.  相似文献

Binucleate Rhizoctonia endophytes from the roots of nursery-grown Pinus sylvestris (Scots pine) seedlings and the orchid Goodyera repens from Scots pine forests were characterized on the basis of morphological characters, anastomosis group membership and PCR-assisted ribosomal DNA fingerprinting. Common hyphal and colony morphological traits displayed by the Finnish binucleate Rhizoctonia isolates and a range of Canadian orchid root endophytes enabled them to be placed in the anamorphic genus Ceratorhiza . Five main anastomosis groups were identified and included groups that contained different combinations of Scots pine, G. repens and Canadian Ceratorhiza spp. isolates. Two Scots pine root endophytes anastomosed with a phytopathogenic Japanese tester isolate, confirming their membership of anastomosis group I, which is known to include the teleomorphic species Ceratobasidium cornigerum . Hierarchical cluster analysis of RFLPs in the internal transcribed spacer of ribosomal DNA enabled the division of isolates into one of five RFLP groups. The RFLP and anastomosis groupings were closely correlated; isolates within each of four RFLP groups, which shared 100% RFLP identity, anastomosed in the cross-pairing anastomosis group tests. However, all represented different vegetatively compatible populations (clones) because the diagnostic killing reaction, a cellular vegetative incompatibility response, was identified at hyphal fusion junctions. These findings indicate a high degree of intraspecific variation within Ceratobasidium cornigerum , which includes isolates able to enter into either mutualistic or pathogenic root association with susceptible host plants. The common anastomosis group/RFLP groupings identified also strongly support the hypothesis that conifer tree roots can act as large inoculum reservoirs for these orchid endophytes, allowing the development of inter-plant connections, via commonly shared hyphal linkages, in boreal forest ecosystems.  相似文献

Excitatory amino acids have been implicated in the pathogenesis of hepatic encephalopathy. In the present study, kainate, quisqualate and N-methyl-D-aspartate (NMDA) subclasses of L-glutamate receptors were measured in adult rat brain by quantitative receptor autoradiography following surgical construction of an end-to-side portacaval anastomosis (PCA). PCA resulted in sustained hyperammonemia and decreased binding of L-glutamate to the NMDA receptor when compared to sham-operated controls. Decreases in binding ranged from 17 to 39% in several regions of cerebral cortex, hippocampus, striatum, and thalamus. Binding to quisqualate and kainate receptor subtypes was not altered. PCA leads to astrocytic changes in brain but does not result in any measurable loss of neuronal integrity. It is therefore proposed that decreased glutamate binding to the NMDA receptor following PCA results from increased extracellular glutamate caused by decreased reuptake into perineuronal astrocytes and a compensatory down-regulation of these receptors. Such changes could be of pathophysiological significance in hepatic encephalopathy.  相似文献

Abstract: HPLC determination of histamine, serotonin, dopamine, and noradrenaline in the brain tissue of rats with portocaval anastomoses (PCA) has revealed a selective increase in histamine concentration. In the posterior hypothalamus, the steady-state level of the amine metabolites showed an inverse pattern; N-tele -methylhistamine(t-MeHA), as estimated by gas chromatography-mass spectrometry, was not changed significantly by portocaval shunting, whereas 5-hydroxyindoleacetic acid (5-HIAA) and homovanillic acid were more than doubled. Interestingly, the net increase in t-MeHA concentration in response to pargyline (80 mg/kg i.p.) was almost the same for PCA and sham-operated rats. This implies that the great enhancement of the histamine level in this area might be a consequence of the persistent stimulation of its synthesis and the unchanged activity of histaminergic neurons. In the rest of the brain, on the other hand, the steady-state level of t-MeHA was higher after PCA (3.8-fold), as were the levels of 5-HIAA and homovanillic acid. Surprisingly, t-MeHA remained unchanged after monoamine oxidase blockade. Of the pargyline-induced alterations in the concentrations of indoles and catechols, the most pronounced were those in the serotonin level; serotonin was elevated more than twofold in hypothalamus and more than 12-fold in the rest of the brain, with a concomitant 80% decrease in 5-HIAA. The dopamine and, to a much smaller extent, noradrenaline levels were also increased, and the levels of homovanillic acid and 3,4-dihydroxyphenylacetic acid fell below the detection limit. The study suggests that at least two different mechanisms operate in the brains of PCA rats to counteract the excessive synthesis of neuromediators, e.g., increased deposition and increased metabolism.  相似文献

Patients with liver cirrhosis show disturbances in sleep and in its circadian rhythms which are an early sign of minimal hepatic encephalopathy (MHE). The mechanisms of these disturbances are poorly understood. Rats with porta-caval shunt (PCS), a model of MHE, show sleep disturbances reproducing those of cirrhotic patients. The aims of this work were to characterize the alterations in circadian rhythms in PCS rats and analyze the underlying mechanisms. To reach these aims, we analyzed in control and PCS rats: (a) daily rhythms of spontaneous and rewarding activity and of temperature, (b) timing of the onset of activity following turning-off the light, (c) synchronization to light after a phase advance and (d) the molecular mechanisms contributing to these alterations in circadian rhythms. PCS rats show altered circadian rhythms of spontaneous and rewarding activities (wheel running). PCS rats show more rest bouts during the active phase, more errors in the onset of motor activity and need less time to re-synchronize after a phase advance than control rats. Circadian rhythm of body temperature is also slightly altered in PCS rats. The internal period length (tau) of circadian rhythm of motor activity is longer in PCS rats. We analyzed some mechanisms by which hypothalamus modulate circadian rhythms. PCS rats show increased content of cGMP in hypothalamus while the activity of cGMP-dependent protein kinase was reduced by 41% compared to control rats. Altered cGMP-PKG pathway in hypothalamus would contribute to altered circadian rhythms and synchronization to light.  相似文献
