To investigate the potential use of n-alkanes (alkanes), long-chain alcohols (alcohols) and long-chain fatty acids (acids) for estimating the diet composition of sheep, in a feeding trial. A total of 18 sheep were assigned randomly to three different diets (diet A, diet B and diet C) containing up to eight herbage species (Leymus chinensis, Leymus dasystachys, Elymus sibiricum, Chenopodium album, Puccinellia chinampoensis, Medicago sativa, Saussurea sinuata and Bromus inermis). Faecal recoveries of alkanes, alcohols and acids were determined, and diet compositions were estimated using different combinations of alkanes, alcohols and acids. The faecal concentrations of individual alkanes, alcohols and acids were corrected using the mean recovery of the dietary treatment that the respective animal belonged to (diet recovery), or the mean recovery across all dietary treatments (general recovery). In general, diets did not affect the faecal recovery values for alkanes, alcohols and acids, and no difference in accuracy was found between diet composition estimates based on dietary recovery and general recovery. The accuracy of diet composition estimates declined as the number of dietary components increased from four to eight herbage species (P < 0.001). Better (P < 0.05) estimates of diet composition were obtained with the combinations of two or three marker types instead of alkanes alone. Moreover, results showed that excluding minor diet components from the calculations decreased (P < 0.05) the accuracy of diet composition estimates, whereas including extra non-grazed herbage species did not reduce (P > 0.05) the quality of diet composition estimates. These results confirmed the usefulness of alkanes, alcohols and acids as markers for determining complex diet composition of sheep. However, a negative impact on the accuracy of diet composition estimates, caused by missing minor diet components from the calculation of diet composition, could happen when plant wax markers are used to estimate the diet composition of free-ranging animals.  相似文献

Saturated hydrocarbons are quantitatively the most abundant fraction among all petroleum hydrocarbons. Significant advances have been made in the understanding of the anaerobic biodegradability of alkanes in terms of the microorganisms involved and the biochemical pathways over the past two decades. They can be used as carbon and energy sources by diverse physiological groups of microorganisms (isolates or consortia) grown under chlorate-reducing, nitrate-reducing, sufidogenic or methanogenic conditions. Two general biochemical mechanisms have been proposed for the initial activation of alkanes including addition of fumarate and carboxylation. However, glycyl radical enzymes dependent fumarate addition which yields alkyl-substituted succinate appear to be the most commonly shared mechanism for the anaerobic attack of alkanes under various redox conditions by phylogenetically diverse microorganisms. The genes encoding the candidate alkylsuccinate synthase have been recently described in alkane-degrading sulfate- and nitrate-reducers as well as in hydrocarbon-rich environments. Alternative mechanisms may also be available depending on the alkane-degrading microbial community and electron acceptors utilized.  相似文献

Spilled crude petroleum from oil wells contains numerous hydrocarbons, some of which are toxic and threaten life. We have studied the mobility and persistence of hydrocarbons in waterlogged soils that contain large proportions of fermented organic matter (Histosols) and large concentrations of dissolved organic carbon (DOC) in the State of Tabasco, Mexico. We sampled soil and phreatic water at sites polluted by oil spills for several decades, as well as at sites that had only recently (few weeks) been polluted, and compared their hydrocarbon contents with those of unaffected sites in the same area. Samples were analyzed for 16 non-alkylated polyaromatic hydrocarbons (PAHs) and n-alkanes from nC9 to nC34. The spilled hydrocarbons had remained predominantly in the organic surface horizons of the soil where spillage occurred; there was little evidence of movement within the soil. The fraction of low molecular weight compounds was larger at sites of recent spills than where spills happened several decades ago. Nevertheless, sites of old spills still contained large concentrations of hydrocarbons, among which those of low molecular weight represented from 30 to 49% of total PAHs and from 50 to 84% of total n-alkanes, indicating that volatilization or microbial degradation is slow in these soils. In the peat horizons the measured organic carbon partition coefficients (K oc ) for the higher molecular weight PAHs were consistently smaller than those estimated by empirical equations by up to two orders of magnitude. The dissolved organic carbon of these peat soils seems to influence this behavior. At sites of old spills, partition coefficients for the PAHs were larger than at sites of recent spills.  相似文献

Sunflower occupies the fourth position among oilseed crops the around the world. Eceriferum (CER) is an important gene family that plays critical role in very-long-chain fatty acids elongation and biosynthesis of epicuticular waxes under both biotic and abiotic stress conditions. The aim of present study was to investigate the effect of sunflower CER genes during drought stress condition. Thus, comparative analysis was undertaken for sunflower CER genes with Arabidopsis genome to determine phylogenetic relationship, chromosomal mapping, gene structures, gene ontology and conserved motifs. Furthermore, we subjected the sunflower cultivars under drought stress and used qRT-PCR analysis to explore the expression pattern of CER genes during drought conditions. We identified thirty-seven unevenly distributed CER genes in the sunflower genome. The phylogenetic analysis revealed that CER genes were grouped into seven clades in Arabidopsis, Helianthus annuus, and Gossypium hirsutum. Expression analysis showed that genes CER10 and CER60 were upregulated in sunflower during drought conditions, indicating that these genes are activated during drought stress. The results obtained will serve to characterize the CER gene family in sunflower and exploit the role of these genes in wax biosynthesis under limited water conditions.Key messageCuticular waxes protect the plants from drought stress, so we observed the expression of wax bio synthesis genes in recently sequences genome of Helianthus annuus. Samples from a long-term bioremediation experiment contaminated with two crude oils, Arabian Heavy and Gullfax, was used to analyze the compositional change of petroleum hydrocarbons. A time course of five different homologous series of petroleum hydrocarbons were analysed by GC/FID and GC/MS. The homologous series were n-alkanes, acyclic isoprenoids, alkylated naphthalenes, alkylated phenanthrenes, and alkylated dibenzothiophenes. Several biomarker compounds were monitored during the experiment to evaluate the possible use as conserved reference compounds for the quantification of other oil compounds, that is, nor-hopanes, hopanes, methyl-hopanes, steranes, mono- og triaromatic steranes. The 17α(H),21β(H)-hopane was found to be stable toward biodegradation and was used as reference compound. The internal standard quantification method was used to quantify changes of the homologous series of oil compounds, and a graphic presentation was used to compare the decrease of the individual compounds. This was found to be an easy way of comparing relative changes in oil. The disappearance of the compounds was extensive and in 6 to 7 months less than 6% remained. The decrease of the n-alkanes (>C15) and acyclic isoprenoids was almost uniform within each homologous series and thus independent of physical-chemical characteristics. Evaporation affected compounds with boiling points lower than n-C15. The alkylated aromatic and sulfur-aromatic compounds decreased according to the degree of alkylation and the decrease showed to be delayed by 10 to 20% by each additional alkyl group. The lack of isomeric-specific degradation of most of the aromatic and sulfur-aromatic compounds, until extensive decrease in concentration had occurred, suggests these compounds have to be dissolved, before any biodegradation occurs.  相似文献

The reaction of AgX (X=ClO4, NO3 or SO3CH3) acceptors with excesses of tris(pyrazol-1-yl)methane ligands L (L=CH(pz)3, CH(4-Mepz)3, CH(3,5-Me2pz)3, CH(3,4,5-Me3pz)3 or CH(3-Mepz)2(5-Mepz)) yields 1:1 [AgX(L)], 2:1 [Ag(L)2]X or 3:2 [(AgX)2(L)3] complexes. The ligand to metal ratio in all complexes is dependent on the number and disposition of the Me substituents on the azole ring of the neutral ligand and on the nature of the Ag(I) acceptor. All complexes have been characterized in the solid state as well as in solution (medium- and far-IR, 1H and 13C NMR and conductivity determinations) and the solid-state structures of [Ag(NO3){(pz)3CH}](∞/∞) and [Ag{(3,5-Me2pz)3CH}2]NO3 determined by single crystal X-ray studies.  相似文献

An investigation of the treatment of a complex waste oil–recycling hydrocarbon wastewater was made by employing white-rot fungal strains immobilized in a pinewood chip-packed reactor. The reactor was operated in sequencing batch mode. The fungal reactor was evaluated in a preliminary bench-scale reactor followed by intermediate-scale operation. Substantial chemical oxygen demand (COD) reduction (> 96% in 48-h batch cycles) and removal of specific influent compound constituents, alkanes (n-C8, n-C10–C12) and aromatic compound (o-xylene), was shown in diluted and undiluted (COD > 37 g L?1) influent. Industrial application of the fungal reactor was evaluated in a 14-m3 pilot plant erected on-site at a waste oil–processing facility. The scale-up implication and optimization of the field plant is discussed in relation to the process-monitoring programme.  相似文献
